The aim of the present study was to assess the remineralizing potential of 10% casein phosphopeptide-amorphous calcium phosphate complex (CPP-ACP) -Recaldent used alone or with fluoride (CPP-ACFP) as compared to fluoride mouthrinse for the non-invasive treatment of occlusal non-cavitary caries lesions in vivo.A total of eighty subjects were randomly assigned to four groups, i.e. group 1 - 10% CPP-ACP, group 2- 10% CPP-ACP + 0.2% NaF, group 3- 0.05% NaF mouthrinse and group 4 �control. The lesions were assessed using visual examination and DIAGNOdent measurements. After 12 weeks of using the corresponding product, the results showed that the highest decrease in fluorescence was produced by CPP-ACFP (from 16.28�1.40 to 6.53�0.59, p=0.002), followed by CPP-ACP (from 15.70�1.82 to 8.18�0.70, p=0.004) and fluoride mouthrinse (from 14.32�1.72 to 7.88�0.68, p=0.005), significantly higher than in the control group (p[0.05). No significant differences were found between the three treatment groups.

Objective: Carboxymethyl chitosan (CMC) is a non-collagenous protein analog which has a similar role as dentin matrix protein 1. CMC stabilizes amorphous calcium phosphate (ACP); hence forming nanocomplexes of CMC-ACP. The purpose of this study was to evaluate the effect of CMC concentration in CMC-ACP on dentin remineralization.Methods: Cavities were formed on the occlusal surfaces of freshly extracted premolar teeth. All samples were demineralized and immersed in phosphate-buffered saline and stored in a shaking incubator at 37°C. The teeth were randomly divided into five groups. Group 1 was control group (no treatment), whereas Groups 2, 3, 4, and 5 were treated with CMC-ACP containing 1%, 2.5%, 5%, and 10% CMC. The remineralized layer on the dentin surface was evaluated using scanning electron microscopy and energy-dispersive X-ray analysis.Results: The highest dentin remineralization capacity was achieved in Group 5 (10% CMC), whereas diminishing effects were observed in Group 4 (5% CMC), Group 3 (2.5% CMC), and Group 2 (1% CMC). Although no significant differences in calcium levels were observed between 2.5%, 5%, and 10% CMC groups, phosphate levels differed significantly in all treatment groups.Conclusion: Optimal dentin remineralization was achieved by the application of CMC–ACP containing 2.5% CMC.

Abstract Objective: To evaluate the effect of a recently introduced prophylactic agent, casein phosphopeptide–amorphous calcium phosphate (CPP-ACP), on shear bond strength of brackets and compare it with the effect of acidulated phosphate fluoride (APF). Materials and Methods: Forty-eight freshly extracted mandibular bovine incisors were used. Teeth were randomly divided into four groups (n = 12) as follows: group 1 served as control, and no pretreatment was performed on the enamel; group 2, enamel was treated with 1.23% APF and CPP-ACP, respectively; group 3, enamel was treated with CPP-ACP; and group 4, enamel was treated with 1.23% APF for 4 minutes. In all groups, brackets were bonded using a conventional acid-etch and bond system (Transbond XT, 3M Unitek, Monrovia, Calif). Bonded specimens were first stored in deionized water at 37°C for 24 hours, subjected to thermal cycling for 1000 cycles, and further stored in distilled water for 6 weeks before debonding procedures. After debonding, teeth and brackets were examined under a stereomicroscope at 10× magnification for any adhesive remaining, in accordance with the modified adhesive remnant index. Results: The shear bond strengths of all experimental groups were significantly higher than that of the control group (P &lt; .01). There was no significant difference between the shear bond strengths of the experimental groups (P &gt; .05). Conclusion: The use of CPP-ACP either alone or combined with APF could be considered as an alternative prophylactic application in orthodontic practice since it did not compromise bracket bond strength.

Different formulas of topical fluoride have been used to manage root carious lesions. This clinical trial aimed to investigate the efficacy of a dental varnish containing casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and fluoride compared with fluoride alone in reversing/arresting root caries in xerostomic patients over 1 year. A total of 80 patients (age range 45–92 years) with primary root caries (<i>n</i> = 184 root carious lesions) and unstimulated salivary flow rate of &#x3c;0.2 mL/min were randomly allocated to receive either dental varnish containing CPP-ACP and 5% fluoride (group 1: MI varnish; GC, Japan) (<i>n</i> = 41, 83 lesions), or dental varnish with 5% fluoride alone (group 2: NUPRO White; Dentsply, USA) (<i>n</i> = 39, 101 lesions). Clinical assessments with Severity Index (SI) for root caries, DIAGNOdent measurements, and varnish application were carried out at baseline, 3, 6, and 12 months. Standard oral hygiene instructions with 1,450 ppm fluoride toothpastes were provided for both groups. After 3 months, 63.9% (<i>n</i> = 46) of root caries in group 1 became hard (SI: 0) compared with 39.3% (<i>n</i> = 35) in group 2 (<i>p</i> &#x3c; 0.01). After 6 and 12 months, the differences in SI were insignificant (group 1, <i>n</i> = 60, 83.3%) (group 2, <i>n</i> = 66, 74.2%) (<i>p</i> = 0.36), and (group 1, <i>n</i> = 60, 89.6%) (group 2, <i>n</i> = 67, 81.7%, <i>n</i> = 1 soft, 1.2%) (<i>p</i> = 0.29), respectively. In both groups, noncavitated leathery lesions were more likely to become hard when compared to the cavitated root caries. A significant decrease in plaque index, surface roughness, lesion dimension, and DIAGNOdent readings with a significant increase in lesion distance from the gingival margin was reported in both groups (<i>p</i> &#x3c; 0.05). This study has provided evidence that fluoride dental varnish either with or without calcium and phosphate has the potential to arrest/reverse root caries, especially noncavitated lesions for patients with xerostomia.

Objectives: To identify the effect of monoclonal antibody bZP3 at ovarian follicles that undergo atresia and diameter of various ovarian follicles.Materials and Methods: This is a true experimental research with post only control group design. Samples were 36 female mices (Mus musculus) which is divided into 6 groups, there are 3 control groups (group 1, 2, and 3) injected by Phospatase Buffer Saline (PBS) 50µl and 3 treatment groups (group 4, 5, and 6) injected by Mab bZP3 50µl. Group 1 and 4 terminated at 5th day, group 2 and 5 terminated at 10th day, and group 3 and 6 terminated at 20th day. Evaluation of atretic ovarian follicles and diameter of ovarian follicles performed by hematoxylin eosin (HE) and the data processed by parametric statistic.Results: There are no significant in different among groups in the aspect of atretic follicles and diameter of folicles (p>0.05), but descriptively, number of follicles undergo atresia of the follicle primary, secondary, and tertiary treatment group was higher than the control group, except on the 20th day of observation time.Conclusion: administration of Mab bZP3 had no effect to amount of atretic follicles and diameter of folicles during observation time.

SUMMARY Objectives: The objective of this in vitro study was to quantify the amount of mineral change in demineralized dentin at pH 5.5 after the application of dental varnishes containing fluoride with casein phosphopeptide–amorphous calcium phosphate, fluoride and bioglass, or fluoride alone. Methods and Materials: A total of 12 extracted human sound mandibular premolar root samples were coated with an acid-resistant varnish, leaving a 2 × 3 mm window at the outer root surface. These root specimens were then randomly divided into four groups and separately subjected to the demineralizing cycle at a pH of 4.8 for five days to create artificial caries-like lesions in dentin. Subsequently, each sample was imaged using quantitative x-ray microtomography (XMT) at a 15-μm voxel size. Each test group then received one of the following treatments: dental varnish containing casein phosphopeptide–amorphous calcium phosphate and fluoride (CPP-ACP, MI varnish, GC Europe), bioglass and fluoride (BGA, Experimental, Dentsply Sirona), or fluoride alone (NUPRO, Dentsply Sirona), as well as a control group, which received no treatment. These samples were kept in deionized water for 12 hours. The thin layer of varnish was then removed. All samples including the nonvarnish group were subjected to the second demineralizing cycle at pH 5.5 for five days. The final XMT imaging was then carried out following the second demineralizing cycle. XMT scan was also carried out to varnish samples at 25 μm voxel size. The change in mineral concentration in the demineralized teeth was assessed using both qualitative and quantitative image analysis. Results: There was an increase in radiopacity in the subtracted images of all varnish groups; a significant increase in mineral content, 12% for the CPP-ACP and fluoride (p≤0.05 and p≤0.001), 25% BGA (p≤0.001), and 104% fluoride alone varnish (p≤0.001). There was an increase in the size of radiolucency in the lesion area with a significant decrease in mineral content in the nonvarnish group, 10% (p≤0.05 and p≤0.001). Conclusions: There was encouraging evidence of a remineralization effect following the application of dental varnish on dentin and also an observed resistance to demineralization during the acidic challenge in all cases. However, a dental varnish containing fluoride alone appeared to have a much greater effect on dentin remineralization when compared with CPP-ACP with fluoride and bioglass with fluoride.

Objective We investigated the effects of bumetanide alone and in combination with dexamethasone on facial nerve regeneration in rats with facial paralysis. Study Design A prospective controlled animal study. Setting An animal laboratory. Subjects and Methods Facial paralysis was induced in 32 Wistar rats that we then divided into 4 groups: group 1, control; group 2, bumetanide; group 3, dexamethasone; group 4, bumetanide and dexamethasone. Electroneurography was performed 1, 2, and 4 weeks later, and nerve regeneration was evaluated by electron and light microscopy and Western blotting in week 4. Results Regarding the comparison between preoperative values and week 4, the latency difference in group 1 (1.25 milliseconds) was significantly higher than those of groups 2 to 4 (0.56, 0.34, and 0.10 milliseconds, respectively; P = .001). The latency increment in groups 2 and 3 was higher than that of group 4 ( P = .002 and P = .046) in week 4, whereas groups 2 and 3 did not differ significantly ( P = .291). Amplitude difference was not statistically significant from week 4 among all groups (all P > .05). The number of myelinated axons was significantly higher in all treatment groups than in the control group ( P = .001). Axon number and intensity were significantly higher in group 4 as compared with groups 2 and 3 ( P = .009, P = .005). Conclusion After primary neurorrhaphy, dexamethasone and bumetanide alone promoted nerve recovery based on electrophysiologic and histologic measures. Combination therapy was, however, superior.

Background: Endodontic infection may persist despite root canal instrumentation. Thus, the use of intracanal medicaments plays an essential role in eliminating resistant bacteria like Enterococcus faecalis, known to be associated with persistent infections in endodontically treated teeth. Although calcium hydroxide is the gold standard intracanal medicament, it has been reported that Enterococcus faecalis is immune to its effects. Therefore, several studies assessed the efficacy of other intracanal medicaments, but none to date evaluated Casein Phosphopeptide-Amorphous Calcium Phosphate. Objectives: This in-vitro randomized controlled study aimed to assess the antibacterial efficacy of Casein phosphopeptide-amorphous calcium phosphate as an intracanal medicament against Enterococcus faecalis and compared it to calcium hydroxide. Methods: 60 extracted single root canal permanent teeth were prepared and later divided into three equal groups according to the intracanal medicament used. Group 1: No intracanal medicament (negative control), Group 2: Calcium hydroxide paste, and Group 3: Casein phosphopeptide-amorphous calcium phosphate paste. The intracanal medicaments were placed on the canals for 7 days. The outcome of this procedure was measured by counting colony-forming units. Statistical analysis was carried out using One-Way ANOVA and Tukey’s Post Hoc Test to determine significant differences between the groups. Results: The mean bacterial count for Group 2 was significantly lower than Group 1 and Group 3. Calcium hydroxide showed significantly more antibacterial efficacy against Enterococcus faecalis than Casein phosphopeptide-amorphous calcium phosphate and the negative control groups. Conclusion: Casein Phosphopeptide-amorphous calcium phosphate is ineffective in inhibiting Enterococcus faecalis growth compared to Calcium hydroxide.

Abstract Aim This review of the literature examines the role of the natural components of saliva in maintaining tooth mineralization and the role of different casein phosphopeptide amorphous calcium phosphate-based (CPP-ACP) compounds in controlling demineralization/ remineralization and their clinical applications. Background A group of peptides, known as CPP, have been shown to stabilize calcium and phosphate preserving them in an amorphous or soluble form known as amorphous calcium phosphate (ACP). Calcium and phosphate are essential components of enamel and dentine and form highly insoluble complexes, but in the presence of CPP they remain soluble and biologically available. This CPP-ACP complex applied to teeth by means of chewing-gum, toothpaste, lozenges, mouth rinses, or sprays is able to adhere to the dental biofilm and enamel hydroxyapatite providing bioavailable calcium and phosphate ions. Review Results Significantly high levels of calcium and phosphate have been found in both biofilm and subsurface incipient caries lesions and in lower level demineralization of enamel or dentine surfaces previously treated with CPP-ACP based compounds. When placed on the surface of a tooth with early carious lesions, pastes with CPP-ACP complexes can prevent tooth demineralization and improve enamel remineralization and enhance fluoride activity. Conclusion Remineralization of white spot lesions has been achieved clinically by applying pastes based on these compounds, and a similar effect to self-applied fluorides has been observed in reducing the appearance of new caries lesions in patients with xerostomia.
