Effects of different nutrient media, vitamins and peptone on in vitro asymbiotic seed germination of Geodorum densiflorum (Lam.) Schltr. were studied. In vitro developed rhizomes were used to determine the influence of cytokinins on growth and morphogenesis. Seed germination and survival rate of protocorms were highest in MS medium compared to other basal media. The protocorms raised through seed germination directly proliferated into rhizomes in later phase of growth and development. Nodal portions of rhizomes further exhibited growth through formation of direct and callus mediated protocorm like bodies. In general, three types of rhizome movements were noticed; viz. positively geotropic, negatively geotropic and diagravitropic movements. In the PGR free control and in the presence of NAA alone rhizomes exhibited positively geotropic movements. On the contrary, presence of cytokinin either alone or in combination with NAA exhibited diagravitropic movements. Application of TDZ completely suppressed the positively geotropic movement and enhanced the frequency of negatively geotropic movement of rhizomes followed by shoot bud formation. BAP was most effective cytokinin for large scale rhizome mediated multiplication of Geodorum densiflorum and TDZ could be effectively employed for rapid leafy shoot regeneration. Growth regulators, phytohormones, both natural and artificial, are the main means to control plant ontogenesis. They are involved in regulating the processes of cell differentiation and cell divisions, the formation of tissues and organs, the changes in the rate of growth and development, the duration of the certain stages of ontogenesis. The main classes of phytohormones used in plant biotechnology, in particular, in the induction of haploid structures, are auxins and cytokinins. The mechanism of action of phytohormones on a cell is rather complicated and may have a different character. Understanding the characteristics of the action of phytohormones is complicated by the fact that the system of hormonal regulation of plant life is multicomponent. This is manifested in the fact that the same physiological process is most often influenced not by one, but by several phytohormones, covering a wide range of aspects of cell metabolism. In connection with the foregoing, the purpose of our work was to test a set of nutrient media with different basic composition and different proportions of phytohormones to determine the patterns of their influence on the processes of haploid structure induction in rape anther culture using accessions, developed at the Institute of Oilseed Crops NAAS. The material used was two accessions of winter rapeseed (No. 1 and No. 2) and one sample of spring rapeseed, provided by the Rapeseed Breeding laboratory of the Institute of Oilseed Crops. Incised inflorescences were kept against the background of low temperature of 6–8 ° C for several days, and then, under aseptic conditions, anthers with unripe pollen grains were isolated and planted on nutrient media differing in both basic mineral composition and content of phytohormones. MS (Murashige & Skoog 1962) and B5 (Gamborg et al 1968) media were used as basic media. Phytohormones were added to the basic media in various combinations – BA, 2,4-D, NAA at the concentrations of 0.1-0.6 mg/l. In each treatment up to 300 anthers were cultivated. Differences between treatments were evaluated using standard t-test. Studies have shown that in the anther culture of rapeseed on the tested nutrient media, morphogenic structures of different types (embryoids and callus) were originated. Synthetic auxin 2,4-D, regardless of the composition of the basic medium, caused the formation of structures of both types, though with a low frequency. Phytohormone BA of the cytokinin type had a similar effect. In this case, the frequency of structures was slightly higher, and the developed structures were represented mainly by embryoids. The joint action of cytokinin and auxin was the most favorable for the initiation of morphogenic structures. Such combination of phytohormones caused the formation of these structures with a frequency of 24.5-14.7% in the studied genotypes of winter rape. A similar effect of phytohormones on the induction and development of morphogenic structures was also observed in spring rape. In this case, a single basic MS medium was used. The experiment included treatments where phytohormones were absent (control), as well as various combinations of auxin and cytokinin. In the control treatment, the formation of new structures was not noted. In treatments with phytohormones, in addition to the medium with the combination of auxin and cytokinin, the medium in which only cytokinin was present was also rather effective. The treatment in which the action of auxin 2,4-D was combined with the action of another auxin, NAA, turned out to be practically ineffective. Thus, it was found that for the induction of morphogenic structures from microspores in rape anther culture of the tested genotypes, the combination of cytokinin with auxin, or the use of only single cytokinin BA without other phytohormones, had the most positive effect.

Seeds of three cowpea cultivars namely Black eye, ER 7 and Tswana obtained from the Department of Agriculture Research, Gaborone were tested for the presence of seed-borne fungi, and their possible control in vitro using locally available fungicides. Four hundred fifty seeds of each cultivar of cowpea were disinfected with 2% sodium hypochlorite solution for 10 min and washed three times with sterile distilled water before placing them in PDA plates (5 seeds/9 cm Petri plate), incubated at 22±2o C for 12 hour each under continuous light and dark. A total of eight fungi were detected from seeds of cowpea. These were Aspergillus flavus, A. niger, Cylindrocarpon sp., Fusarium equisiti, F. oxyaporum, Penicillium chyrosogenum, Rhizopus oligosporus and R. stolonifer. Rhizopus spp. were dominant fungi recovered from seeds, followed by Penicillium, Aspergillus, Fusarium and Cylindrocarpon. The fungi detected resulted in decay and rotting of seeds, and thereby reducing percentage germination of seeds (22%, 37% and 63 % seed germination in Black eye, ER7 and Tswana varieties of cowpea respectively). Out of four fungicides tested, benlate, captan, dithane M 45 and chlorothalanil. Dithane M45 effectively controlled seed-borne fungi, and enhanced seed germination to an average of 86% (93% germination with no fungi detected in Tswana variety) as compared to chlorothalonile (79%), benlate and captan (77%) and un-treated seeds (45%). The fungal incidence was reduced to 2.3%, 4.3%, 5.3% and 5.3% when seeds were treated with dithane M-45, chlorothalonil, benlate and captan respectively as compared to 62% in non-treated seeds.

Phytase plays an important role in crop seed germination and plant growth. In order to fully understand the plant growth-promoting mechanism by Rahnella aquatilis JZ-GX1,the effect of this strain on germination of maize seeds was determined in vitro, and the colonization of maize root by R. aquatilis JZ-GX1 was observed by scanning electron microscope. Different inoculum concentrations and Phytate-related soil properties were applied to investigate the effect of R. aquatilis JZ-GX1 on the growth of maize seedlings. The results showed that R. aquatilis JZ-GX1 could effectively secrete indole acetic acid and had significantly promoted seed germination and root length of maize. A large number of R. aquatilis JZ-GX1 cells colonized on the root surface, root hair and the root interior of maize. When the inoculation concentration was 107 cfu/mL and the insoluble organophosphorus compound phytate existed in the soil, the net photosynthetic rate, chlorophyll content, phytase activity secreted by roots, total phosphorus concentration and biomass accumulation of maize seedlings were the highest. In contrast, no significant effect of inoculation was found when the total P content was low or when inorganic P was sufficient in the soil. R. aquatilis JZ-GX1 promotes the growth of maize directly by secreting IAA and indirectly by secreting phytase. This work provides beneficial information for the development and application of R. aquatilis JZ-GX1 as a microbial fertilizer in the future.

Lychnis cognate Maxim and Lychnis fulgens Fish. Ex Spreng are two valued ornamental plants in Korea. Soaking of seeds in GA3 solution remarkably promoted germination up to 60%, but the control (0 mg/l) was not effective (> 5%). To select an adequate temperature for seed germination, seeds, previously soaked in a 1000 mg/l GA3 for 24 hrs, were incubated at 15, 20, 25, and 30°C. Seed germination of over 20% was obtained at 15, 20, and 25°C, but only 10% at 30°C. These results indicate that the seeds of L. cognate and L. fulgens are in a such dormant state that they hardly germinate even by dormancy breaker (GA3) and low (15 ? 25°C) temperature treatment. The highest callus induction was observed in the leaf explants of the seedlings on MS containing specific concentrations of 3.0 mg/l BA and 1.0 mg/l NAA. The adventitious shoot was formed < 90% of calli on 1/2 WPM medium. The height of in vitro propagated plantlet was no different media used for regeneration. This in vitro propagation protocol should be useful for conservation of endangered and ornamental plant.Plant Tissue Cult. & Biotech. 25(1): 1-12, 2015 (June)
