Extraction and Antioxidant Activity of Ultrasonic-Assisted Flavonoids from Suaeda salsa

The box-behnken design combined with response surface methodology was used to optimize ultrasonic-assisted extraction of flavonoids from Suaeda salsa (Linn.) Pall. The results indicate that the highest extraction rate of flavonoids by ultrasonic-assisted extraction could arrive 5.17% using 500W ultrasonic power, 69% ethanol as solvent, 77°C extraction temperature, 1:40 solid to liquid ratio for 58 min extraction. The flavonoids-enriched extracts exhibited quite strong reducing property and excellent inhibitions of DPPH radical, hydroxyl radical. Meanwhile, it was found that the bioactivities appeared to be dose-dependent of flavonoids.

An efficient ultrasonic-assisted extraction of inulin from Jerusalem artichoke was investigated by Response Surface Methodology (RSM). The results showed that the yield of ultrasonic-assisted extraction was 62.07 ± 0.39% over the microwave (40.85 ± 0.28%) and hot water extraction (27.42 ± 0.42%). The Box-Behnken Design (BBD) was used to optimize the effects of three parameters (temperature-X1, ultrasonic power-X2 and time-X3) on inulin yield. Analysis of variance showed that the contributions of X1, X3, X12, X13, X22 were significant. The optimal yield of inulin was 82.93 ± 1.03% at 82 °, 120 W and 18 min.

Calophyllum inophyllum (C. inophyllum) is a plant known as “Penaga Laut” in Malaysia. Its seeds contain valuable oil that serve as a potential alternative sources for vegetable oil. In this study, C. inophyllum seeds oil was extracted using an ultrasonic-assisted extraction (UAE) technique. The optimization of extraction parameters namely different types of solvent, extraction time, ultrasonic power, extraction temperature and liquid to solid (L/S) ratio were performed using one-factor-at-a-time (OFAT) method. The optimum extraction conditions obtained were n-hexane as a solvent, extraction time 20 min, ultrasonic power 210 W, extraction temperature 40°C and L/S ratio 20 ml/g, with an oil yield 55.44 ± 0.53 %.

Ultrasonic-assisted extraction (UAE) method was used to polysaccharides from Radix Astragali (RAP), and the orthogonal test was applied to optimize extraction conditions. Furthermore, the antioxidant activities of RAP were evaluated by DPPH and superoxide anion radical scavenging activities assay. The results indicated that the optimal conditions for RAP extraction as followings: solid/liquid ratios of 1:20, extraction time of 10 min, ultrasonic power of 65 W and extraction temperature of 60°C. Meanwhile, RAP had relatively higher scavenging activities on DPPH and superoxide anion radicals.

Litsea cubeba fruit as raw material in this experiment, the use of ultrasonic-assisted method of litsea cubeba oil extraction process are discussed. The effects of several ultrasonic processing parameters, such as ratio of liquid to solid, extraction temperature, ultrasonic power and extraction time on the extraction of litsea cubeba oil were investigated assisted by ultrasonic wave. And the L9 (34) orthogonal test was used to research this four factors, the result from the visual analysis indicated that the optimal extraction process was gotten when ratio of liquid to solid was 14:1 , ultrasonic time was 50min, ultrasonic power is 320W, ultrasonic temperature is 60 °C.Those experimental results showed that ultrasonic-assisted extraction of the Litsea cubeba oil is practicable, and this method is simple and efficient. With the preliminary investigation of the ultrasonic-assisted extraction of Litsea Cubeba oil, the oil yield reached up to 41.35%, this value is the highest of all the oil yields from the current oil extraction methods, and it is worthy of further exploration .

In this study, response surface methodology (RSM) was used to optimize the ultrasonic-assisted extraction parameters of Sojae Semen Praeparatum polysaccharides (SSPP-80), the optimum conditions were determined as follows: ultrasonic frequency of 100 W, ultrasonic power of 80 Hz, ultrasonic temperature of 52℃, ultrasonic time of 23 minutes, and liquid to raw material ratio of 40 mL/g. Based on these conditions, polysaccharides extraction rate was 7.72% ± 0.26%. Then, 2 novel polysaccharides (SSPP-80‐1, SSPP-80‐2) were isolated from SSPP by DEAE-cellulose 52 chromatography. The chemical compositions, physicochemical properties, and structure of SSPPs were investigated by simultaneous thermal analyzer (TGA), scanning electron microscopy (SEM), fourier transform infrared spectroscopy (FI-IR), and high-performance liquid chromatography (HPLC). The results showed that SSPP-80 and 2 fractions were mainly composed of mannose (Man), glucose (Glc), galactose (Gal), xylose (Xyl), and arabinose (Ara). In addition, the antioxidant activities were evaluated against the DPPH and hydroxyl radical in vitro, the IC50 of SSPP-80, SSPP-80‐1 and SSPP-80‐2 against DPPH free radical were 4.407, 8.267, and 5.204 mg/mL, respectively, whereas the IC50values for removing hydroxyl groups were 5.318, 3.516, and 4.016 mg/mL, respectively. It demonstrated that SSPP-80 and 2 fractions had certain antioxidant activity. Theoretical basis for use of Sojae Semen Praeparatum polysaccharides was provided by this study.

Ultrasonic technology was applied to polysaccharides extraction from Ottelia acuminata (Gagnep.) Dandy and Response Surface Methodology (RSM) was used to optimize the effects of processing parameters on polysaccharides yields. Three independent variables such as liquid-solid ratio (ml/g, X1), extraction temperature (°C, X2) and extraction time (hour, X3) were investigated respectively. The statistical analysis indicated that the three variables and the quadratic of X1 and X3 had significant effects on the yields and followed by the significant interaction effects between the variables of X1 and X3, X2 and X3 (p<0.05). A mathematical model with high determination coefficient was gained. The optimal extraction conditions of polysaccharides were determined as follows: liquid-solid ratio 43 ml/g, extraction temperature 90 °C and extraction time 3.45 hours. Under these conditions, the experimental yield of polysaccharides was 107.44 mg/g, which was agreed closely with the predicted value 108.71 mg/g.

This paper studies the optimization of ultrasonic-assisted extraction of polysaccharides from Oenothera biennis L. leaves using Box-Behnken design, and their in vitro antioxidant activity and structure. It is demonstrated that the optimum extraction conditions are: the ultrasonic power at 400 W, solvent-tosample ratio of 23:1 (mL/g), ultrasonication temperature at 53 °C, and time for 36 min, under which the yield of polysaccharides from Oenothera biennis L. leaves, namely 3.54%, remains close to the predicted value in the model. The assays of the scavenging effect on DPPH and hydroxyl free radicals, and of the reducing power indicate that polysaccharides from Oenothera biennis L. leaves possess strong antioxidant activity and certain relationship exists between their dose and effect in the concentration range of the experiment; Fourier IR spectroscopy reveals that characteristic absorption peak of polysaccharides can be seen from the sample; the DE-52 cellulose column chromatography, HPLC profiles, and GC-MS chromatogram identify the obtained OCP-2 as heteropolysaccharides, whose major components, including arabinose, mannose, galactose, and talose, have an average molecular weight of 5435 Da.

Ultrasonic-Assisted extraction (UAE) was applied for saponins extraction from Ophiopholis mirabilis and response surface methodology (RSM) was used to optimize the extraction technology. A first set of experiments were designed to optimize water bath temperature, ethanol concentration, the ratio of liquid to solid, ultrasonic time (min) and ultrasonic power (W) as the main variables affecting the extraction effect. A Box-Behnken design consisting of 15 experimental runs was then applied and a second-order polynomial model was used to explain the interactions between the main factors and the yield of saponins. The developed mathematical model was found to fit well with the experimental data and the optimized conditions were ultrasonic pretreatment time 30.9 min, ethanol concentration 64%, liquid /solid ratio 35:1, ultrasonic power 300W and water bath 70°C. Corresponding saponins content was 0.574%.

Leonurus japonicus is rich in bioactive compounds especially polysaccharide. Single factors test and response surface methodology were used to study the optimum conditions for ultrasonic - assisted extraction of polysaccharide from L. japonicus, and its physiochemical and DPPH scavenging activities also were evaluated. The optimal conditions as underpinned by this paper is as follow: extraction temperature of 70 �C, extraction time of 50 min and extraction power of 210W. The polysaccharide of L. japonicus was shown to be a non-reducing sugar and to exhibit DPPH scavenging activity to an extent. The polysaccharide structures were studied using SEM, AFM, and FT-IR. It showed L. japonicus polysaccharide had been having more than one chains which were branched and entangled with each other. and existed a broad band wavelength of 2925.64, 1635.06, 1539.7, 1046.73, and 477.86 cm-1. The study indicated that L. japonicus may have a beneficial use to the medical and food industries.

Deep eutectic solvents (DESs), a new group of ecofriendly solvent combined with the ultrasonic-assisted extraction (UAE) technique, were first successfully used for extraction of resveratrol from peanut roots. Resveratrol in the extract was analyzed and quantified using a HPLC-UV method. A series of DESs consisting of choline chloride (ChCl) and 1,4-butanediol, citric acid, and ethylene glycol were formulated, finding ChCl/1,4-butanediol was a most proper extraction system. The optimal extraction parameters were obtained using a Box–Behnken design (BBD) test combined with response surface methodology as follows: 40% of water in ChCl/1,4-butanediol (1/3, g/g) at 55°C for 40 min and solid/liquid ratio of 1:30 g/mL. The total extraction content and extraction yield of resveratrol from peanut roots could reach 38.91 mg/kg and 88.19%, respectively, under such optimal conditions. The present study will provide a typical example for using DESs to extract natural bioactive compounds from plants.
