This article addresses issues surrounding the social construction of internet addiction, focusing on conceptualisations of reality, escape, hope, and time. Drawing on a critical realist account of semiosis, the framing of internet addiction in China is analysed using the documentary film Web Junkie as an empirical pivot and point of departure. A contextual overview of relations, interests, and tensions surrounding youth and the internet in China is provided, and the film Web Junkie is briefly presented. The main body of the article consists of a critical analysis of conceptualisations of “reality” and “escape.” The core tension focused on in the analysis is the struggle over time, necessitating engagement with critical thought on hope and utopia. The analysis concludes that struggles over temporal autonomy underlie conflicting claims about “reality” and “escape” that are central to “internet addiction” and its treatment in China today.

There is increasing global competition for better product and process functionality, higher quality, lower costs, and other considerations including energy and environmental challenges. This trend requires that Canadian industry be more innovative and responsive in order to stay competitive internationally. The Canadian capability in Engineering Design is at the core of our ability to achieve this goal. At both the undergraduate and graduate levels, we must improve the capability and capacity of engineering graduates so that they are capable of leading innovation, and converting research results into value-added products and services. This paper addresses the engineering design competency, identifies needs in engineering design training, and describes directions for the design content in engineering education programs.

It is proposed to adapt the new version of the internationally recognized standards for engineering education the Core CDIO Standards 3.0 to the programs of basic higher education in the field of technology, natural and applied sciences, as well as mathematics and computer science in the context of the evolution of STEM. The adaptation of the CDIO standards to STEM higher education creates incentives and contributes to the systematic training of specialists of different professions for coordinated teamwork in the development of high-tech products, as well as in the provision of comprehensive STEM services. Optional CDIO Standards are analyzed, which can be used selectively in STEM higher education. Adaptation of the CDIO-FCDI-FFCD triad to undergraduate, graduate and postgraduate studies in the field of science, technology, engineering and mathematics is considered as a mean for improving the system of three-cycle STEM higher education.

ABSTRACTThe RNA genome of human immunodeficiency virus type 1 (HIV-1) is enclosed in a cone-shaped capsid shell that disassembles following cell entry via a process known as uncoating. During HIV-1 infection, the capsid is important for reverse transcription and entry of the virus into the target cell nucleus. The small molecule PF74 inhibits HIV-1 infection at early stages by binding to the capsid and perturbing uncoating. However, the mechanism by which PF74 alters capsid stability and reduces viral infection is presently unknown. Here, we show, using atomic force microscopy (AFM), that binding of PF74 to recombinant capsid-like assemblies and to HIV-1 isolated cores stabilizes the capsid in a concentration-dependent manner. At a PF74 concentration of 10 μM, the mechanical stability of the core is increased to a level similar to that of the intrinsically hyperstable capsid mutant E45A. PF74 also prevented the complete disassembly of HIV-1 cores normally observed during 24 h of reverse transcription. Specifically, cores treated with PF74 only partially disassembled: the main body of the capsid remained intact and stiff, and a cap-like structure dissociated from the narrow end of the core. Moreover, the internal coiled structure that was observed to form during reverse transcriptionin vitropersisted throughout the duration of the measurement (∼24 h). Our results provide direct evidence that PF74 directly stabilizes the HIV-1 capsid lattice, thereby permitting reverse transcription while interfering with a late step in uncoating.IMPORTANCEThe capsid-binding small molecule PF74 inhibits HIV-1 infection at early stages and perturbs uncoating. However, the mechanism by which PF74 alters capsid stability and reduces viral infection is presently unknown. We recently introduced time-lapse atomic force microscopy to study the morphology and physical properties of HIV-1 cores during the course of reverse transcription. Here, we apply this AFM methodology to show that PF74 prevented the complete disassembly of HIV-1 cores normally observed during 24 h of reverse transcription. Specifically, cores with PF74 only partially disassembled: the main body of the capsid remained intact and stiff, but a cap-like structure dissociated from the narrow end of the core HIV-1. Our result provides direct evidence that PF74 directly stabilizes the HIV-1 capsid lattice.
