With the increasing number of cars, the demand for vehicle maintenance lifts is also increasing. The hydraulic valve is one of its core components, but there are problems with it such as inaccurate positioning and failure. In order to improve the service performance of vehicle maintenance elevators, a novel annular multi-channel magnetorheological (MR) valve structure was creatively proposed based on intelligent material MR fluid (MRF), and its magnetic circuit was designed. The influence of current, damping gap and coil turns on the pressure drop performance of the annular multi-channel MR valve was numerically studied and compared with ordinary type magnetorheological valve pressure drop performance through contrast and analysis. The influence of different loads and currents on the pressure drop performance of annular multi-channel magnetorheological valve was verified by experiments, and the reliability of numerical analysis results was verified. The results show that the single winding excitation coil is 321 to meet the demand. The pressure drop performance of the annular multi-channel magnetorheological valve is 5.6 times that of the ordinary magnetorheological valve. The load has little influence on the regulating range and performance of pressure drop of the MR valve. Compared with the common type, the pressure drop performance of the annular multi-channel MR Valve is improved by 3.7 times, which is basically consistent with the simulation results.

AbstractHippo pathway is initially identified as a master regulator for cell proliferation and organ size control, and the subsequent researches show this pathway is also involved in development, tissue regeneration and homeostasis, inflammation, immunity and cancer. YAP/TAZ, the downstream effectors of Hippo pathway, usually act as coactivators and are dependent on other transcription factors to mediate their transcriptional outputs. In this review, we will first provide an overview on the core components and regulations of Hippo pathway in mammals, and then systematically summarize the identified transcriptional factors or partners that are responsible for the transcriptional output of YAP/TAZ in stem cell and differentiation. More than that, we will discuss the potential applications and future directions based on these findings.

Defining the central components of an intervention is critical for balancing fidelity with flexible implementation in both research settings and community practice. Implementation scientists distinguish an intervention’s essential components (thought to cause clinical change) and adaptable periphery (recommended, but not necessary). While implementing core components with fidelity may be essential for effectiveness, requiring fidelity to the adaptable periphery may stifle innovation critical for personalizing care and achieving successful community implementation. No systematic method exists for defining essential components a priori. We present the CORE (COmponents & Rationales for Effectiveness) Fidelity Method—a novel method for defining key components of evidence-based interventions—and apply it to a case example of reciprocal imitation teaching, a parent-implemented social communication intervention. The CORE Fidelity Method involves three steps: (1) gathering information from published and unpublished materials; (2) synthesizing information, including empirical and hypothesized causal explanations of component effectiveness; and (3) drafting a CORE model and ensuring its ongoing use in implementation efforts. Benefits of this method include: (1) ensuring alignment between intervention and fidelity materials; (2) clarifying the scope of the adaptable periphery to optimize implementation; and (3) hypothesizing—and later, empirically validating—the intervention’s active ingredients and their associated mechanisms of change. Lay abstract Interventions that support social communication include several “components,” or parts (e.g. strategies for working with children and families, targeting specific skills). Some of these components may be essential for the intervention to work, while others may be recommended or viewed as helpful but not necessary for the intervention to work. “Recommended” components are often described as “adaptable” because they can be changed to improve fit in different settings where interventions are offered or with different individuals. We need to understand which parts of an intervention are essential (and which are adaptable) when translating interventions from research to community settings, but it is challenging to do this before studying an intervention in the community. This article presents the CORE (COmponents & Rationales for Effectiveness) Fidelity Method—a new method for defining the essential components of evidence-based interventions—and applies it to a case example of Reciprocal Imitation Teaching, an intervention that parents are taught to deliver with their young children with social communication delays. The CORE Fidelity Method involves three steps: (1) gathering information from multiple sources; (2) integrating information from previous research and theory; and (3) drafting a CORE model for ongoing use. The benefits of using the CORE Fidelity Method may include: (1) improving consistency in intervention and research materials to help all providers emphasize the most important skills or strategies; (2) clarifying which parts of the intervention can be adapted; and (3) supporting future research that evaluates which intervention components work and how they work.

In this paper, we propose a clinical approach to the counseling of distressing subjective paranormal experiences, usually referred to as anomalous or exceptional experiences in the academic field. These experiences are reported by a large part of the population, yet most mental health practitioners have not received a specific training in listening constructively to these experiences. This seems all the more problematic since nearly one person in two find it difficult to integrate such experiences, which can be associated with different forms of psychological suffering. After having described briefly several clinical approaches already developed in this area, we outline the main aspects of clinical practice with people reporting exceptional experiences, in particular the characteristics of the clinician’s attitude toward the narrative of unusual events. We then present the core components of a Psychodynamic Psychotherapy focused on Anomalous Experiences (PPAE) based on three main steps: phenomenological exploration, subjective inscription and subjective integration of the anomalous experience. Such an approach, based on a non-judgmental and open listening, favors the transformation of the ontological shock that often follows the anomalous experiences into a potential source of integration and psychological transformation.

Pancreatic ductal adenocarcinoma (PDAC) is one of the most refractory human malignancies. F-box only proteins (FBXO) are the core components of SKP1-cullin 1-F-box E3 ubiquitin ligase, which have been reported to play crucial roles in tumor initiation and progression via ubiquitination-mediated proteasomal degradation. However, the clinical implications and biological functions of FBXOs in PDAC have not been fully clarified. Herein we perform a comprehensive analysis for the clinical values and functional roles of FBXOs in PDAC using different public databases. We found that FBXO1 (CCNF), FBXO20 (LMO7), FBXO22, FBXO28, FBXO32, and FBXO45 (designated six-FBXOs) were robustly upregulated in PDAC tissues, which predicted an adverse prognosis of PDAC patients. There was a significant correlation between the expression levels of six-FBXOs and the clinicopathological features in PDAC. The transcriptional levels of six-FBXOs were subjected to the influence of promoter methylation levels. There were more than 40% genetic alterations and mutations of six-FBXOs, which affected the clinical outcome of PDAC patients. Furthermore, the expression of six-FBXOs was associated with immune infiltrations and activated status, including B cells, CD8+ T cells, CD4+ T cells, NK cells, macrophages, and dendritic cells. The functional prediction revealed that the six-FBXOs were involved in ubiquitination-related pathways and other vital signaling pathways, such as p53, PI3K/Akt, and Hippo pathway. Therefore, six-FBXOs are the promising prognostic biomarkers or potential targets for PDAC diagnosis and treatment.

The endosomal sorting complex required for transport (ESCRT)-III mediates budding and abscission of intraluminal vesicles (ILVs) into multivesicular endosomes. To further define the role of the ESCRT-III associated protein Mos10/Vps60 in ILV formation, we screened for new interaction partners by SILAC/MS. Here, we focused on the newly identified interaction partner Vps68. Our data suggest that Vps68 cooperates with ESCRT-III in ILV formation. The deletion of VPS68 caused a sorting defect similar to the SNF7 deletion, when the cargo load was high. The composition of ESCRT-III was altered, the level of core components was higher and the level of associated proteins was lower in the deletion strain. This suggests that a shift occurs from an active complex to a disassembly competent complex and that this shift is blocked in the Δvps68 strain. We present evidence that during this shift Snf7 is replaced by Mos10. Vps68 has an unusual membrane topology. Two of its potential membrane helices are amphipathic helices localized to the luminal side of the endosomal membrane. Based on this membrane topology we propose that Vps68 and ESCRT-III cooperate in the abscission step by weakening the luminal and cytosolic leaflets of the bilayer at the abscission site.

The core components of the digital economy, including digital technologies that serve as the new engine of growth for Society 5.0, were identified. The chapter concluded by highlighting the implications of digital technologies, and how standardisation, upgrading curriculum, legislative frameworks, and policies remedy the impediment of growing the digital economy for Society 5.0.

The internet of things (IoT) is aimed at modifying the life of people by adopting the possible computing techniques to the physical world, and thus transforming the computing environment from centralized form to decentralized form. Most of the smart devices receive the data from other smart devices over the network and perform actions based on their implemented programs. Thus, testing becomes an intensive process in the IoT that will require some normalization too. The composite architecture of IoT systems and their distinctive characteristics require different variants of testing to be done on the components of IoT systems. This chapter will discuss the necessity for IoT testing in terms of various criteria of identifying and fixing the problems in the IoT systems. In addition, this chapter examines the core components to be focused on IoT testing and testing scope based on IoT device classification. It also elaborates the various types of testing applied on healthcare IoT applications, and finally, this chapter summarizes the various challenges faced during IoT testing.

Moving from Fichte’s assumption that “the essence of the I is its activity”, this paper tries to analyze the meaning and implications of the idea of “activity” [Tathandlung] in order to explicate the peculiarities of Fichte’s critical, transcendental, and moral idealism. Fichte’s idea of activity will be examined with reference to such basic concepts as collision [Anstoss], interaction [Wechselwirkung], inter-determination [Wechselbestimmung], and striving [Streben]. However, it is freedom which frames and connects the core components of Fichte’s thinking and sets up the goal of his philosophy of action. What freedom accounts for, can be identified both at the transcendental level, in the internal dynamic of infinity and finitude constituting the subjectivity of the I, and at the moral and social levels of Fichte’s thought, as the goal of the human action in history and in the society. In assuming the unitary character of Fichte’s philosophical system, concluding remarks are developed concerning the moral meaning of the act of striving for freedom and, conversely, the immorality of attitudes and feelings such as fear, resignation, and fatigue.
