Staphylococcus Epidermidis and Pseudomonas Aeruginosa Adhesion Intensity on a TiO2 Ceramic in an in Vitro Study

Extrusion is a perspective forming technology for obtaining objects with certain profile, important for the TiO2 application as biomaterial. Extruded samples were calcinated at 1100 °C, thermally treated in different atmospheres: at 1450 °C in air and at 1300 °C in vacuum. An approach was made to examine the adhesion and colonization intensity of Staphylococcus epidermidis and Pseudomonas aeruginosa on TiO2 ceramic in vitro. It was found that Ps.aeruginosa demonstrated higher adhesion and colonization intensity as S.epidermidis and TiO2 samples treated in vacuum demonstrated higher attachment of microorganisms as TiO2 samples treated in air. It was supposed that surface charge promoted the bacterial adhesion on the vacuum treated samples.

Se evaluó la actividad antibacteriana in vitro de aceites esenciales de diferentes especiesdel género Citrus frente a cepas ATCC de Staphylococcus aureus, Staphylococcus epidermidis,Klebsiella pneumoniae, Pseudomonas aeruginosa y Escherichia coli, determinandola concentración mínima inhibitoria (CMI) y la concentración mínima bactericida(CMB). Las bacterias se replicaron en medios de agar y caldos específicos. Se determinóel momento de máxima densidad óptica (DO620) para emplearlo como tiempode incubación; luego se hicieron pruebas de evaluación de sensibilidad con la exposiciónde las cepas a concentraciones a 1000 g/mL del extracto en caldo. Para solubilizarse empleó dimetilsulfóxido (DMSO) al 1%. Posteriormente, se le determinó laconcentración mínima inhibitoria mediante metodologías de microdilución en caldoy la concentración mínima bactericida. Encontrándose una actividad de los aceitesesenciales del género Citrus, con valores de CMI ≥ 600 mg/mL frente a S. aureus,S. epidermidis, K. pneumoniae, P. aeruginosa y E. coli. En función a los resultados obtenidos,se concluye que las diferentes especies del género Citrus son consideradas comopromisorias para el control del componente bacteriano.

AbstractThe aerial parts of Gontscharovia popovii (B. Fedtsch. and Gontsch.) Boriss. were collected at full flowering stage. The essential oil was isolated by hydrodistillation and analyzed by a combination of capillary GC and GC-MS. Thirty-one components were identified with the main constituent being carvacrol (71.9%), followed by linalool (5.5%), p-cymene (4.5%) and γ-terpinene (4.4%). The in vitro antimicrobial activity of the essential oil of G. popovii was studied against seven Gram-positive and Gram-negative bacteria (Bacillus subtilis, Enterococcus faecalis, Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Pseudomonas aeruginosa and Klebsiella pneumoniae) and three fungi (Candida albicans, Saccharomyces cerevisiae and Aspergillus niger). The results of the bioassays showed that the oil exhibited strong antimicrobial activity against all the tested fungi and bacteria except for the resistant bacterium Pseudomonas aeruginosa.

El estudio tuvo como objetivo analizar la composición química del aceite esencial de las hojas de Petroselinum crispum (Mill) Nyman ex A.W. Hill “perejil” y determinar su actividad antibacteriana. El aceite esencial se obtuvo tratando 20 kg de hojas frescas con un equipo de destilación por arrastre con vapor de agua; obteniéndose cuatro mililitros y un rendimiento de 0,02 por ciento v/p. En el análisis cualitativo se analizó su solubilidad en solventes orgánicos; los componentes químicos se determinaron por cromatografía de Gases/ Espectrometría de Masas (CG/EM), lográndose la elucidación de los siguientes compuestos: 1R-α-pineno, β-pineno, β-felandreno, p, α-dimetil estireno, (E)-2-caren-4-ol, 1,3-benzodioxol, 4-metoxi-6-(2-propenil)-, y 1,3-benzodioxol,4,7-dimetoxy-5-(2-propenil)-. La determinación de la actividad antibacteriana in vitro, se realizó por el método de difusión en agar, demostrándose actividad significativa frente a Staphylococcus aureus ATCC 25933 y Staphylococcus epidermidis cepa clínica, a las concentraciones de 100 y 50 por ciento; y para Pseudomonas aeruginosa ATCC 2783 a la concentración de 100 por ciento, teniendo de poca a ninguna actividad frente a Bacillus subtilis y Escherichia coli. Los resultados obtenidos muestran que el aceite esencial de Petroselinum crispum (Mill) Nyman ex A.W. Hill “perejil” presenta actividad antibacteriana.

We compared association of Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa and Enterococcus faecalis with nine vascular implants after co-culture. Vascular implants were composed of various materials such as warp knitted polyester (with or without gelatin and silver ions), expanded polytetrafluoroethylene and biological materials – surface treated porcine pericardial patch and Omniflow II. The lowest overall number of associated bacteria was detected for polytetrafluoroethylene implants and porcine pericardial patch. The highest overall number of associated bacteria was detected for Omniflow II implant. The major source of variation, i.e. primary factor influencing colonization, is the implant type (56.22%), bacterial species is responsible for only 1.81%, and interaction of those two factors – 13.09% of variation.

AbstractObjectives:To evaluate the long-term efficacy of urinary Foley catheters (latex and silicone) impregnated with (1) chlorhexidine and silver sulfadiazine (CXS) and (2) chlorhexidine, silver sulfadiazine, and triclosan (CXST) in inhibiting extra-luminal bacterial adherence and to compare their efficacy with that of silver hydrogel latex (SH) and nitrofurazone-treated silicone (NF) catheters.Design:The antimicrobial spectrum of these catheters was evaluated using a zone of inhibition assay. A novel in vitro urinary tract model was developed to study the potential in vivo efficacy of antimicrobial catheters in preventing extraluminal bacterial colonization. The “meatus” was inoculated daily with Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Enterococcus faecalis, Pseudomonas aeruginosa, and Candida albicans. The “bladder” portion of the model was cultured daily to determine bacterial growth.Results:Both CXS and CXST catheters had a broader antimicrobial spectrum than SH and NF catheters. In the in vitro model, CXST latex and silicone catheters exhibited significantly better efficacy (3 to 25 days) against uropathogens, compared with CXS (1 to 14 days) and control (0 to 5 days) catheters (P = .01). CXST latex catheters exhibited significantly longer protection against Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, and Pseudomonas aeruginosa, compared with SH catheters (P = .01). CXST silicone catheters resisted colonization with Staphylococcus aureus and Staphylococcus epidermidis for a significantly longer period (23 to 24 days) than did NF catheters (9 to 11 days) (P = .01).Conclusion:Catheters impregnated with synergistic combinations of chlorhexidine, silver sulfadiazine, and triclosan exhibited broad-spectrum, long-term resistance against microbial colonization on their outer surfaces (Infect Control Hosp Epidemiol 2003;24:506-513)

Introdução: Sabonetes antissépticos vêm sendo cada vez mais utilizados pela população. No entanto, muito se questiona sobre sua eficiência e a necessidade do uso dos mesmos, tendo em vista que muitos ingredientes ativos presentes nesses sabonetes podem ocasionar riscos à saúde humana e ao meio ambiente. Assim, objetivou-se neste estudo avaliar a ação antibacteriana dos sabonetes comerciais com e sem ingredientes antissépticos. Metodologia: Foram avaliados cinco sabonetes, sendo três antissépticos (Prote & Clean, Lifebuoy e Protex) e dois sabonetes neutros (Nivea e Johnson’s), por meio de ensaios in vitro realizados através das técnicas de difusão em ágar (poço) e microdiluição em caldo frente à Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli e Pseudomonas aeruginosa. Resultados: Todos os sabonetes antissépticos avaliados apresentaram atividade antibacteriana, com destaque para o Prote & Clean que inibiu todas as bactérias testadas. Os sabonetes neutros, apesar de não apresentarem ingredientes ativos, também apresentaram atividade antibacteriana. Conclusão: Foi possível identificar que todos os sabonetes testados possuíram atividade antibacteriana para uma ou mais bactérias patogênicas. Porém, tendo em vista os riscos potenciais de alguns princípios ativos presentes nos sabonetes antissépticos para a saúde e meio ambiente, sugere-se a utilização dos mesmos somente em locais mais propícios às infecções ou em casos de aconselhamento médico, sendo recomendado o uso de sabonetes neutros nos domicílios.
