Two experiments were conducted to assess the effects of storage conditions on the quality of large round bales (LRB). Samples were taken at 75-d intervals from LRB stored inside, outside single bales covered with a 15-mm black polyethylene plastic bag, outside uncovered as single bales, outside uncovered in a single row and outside uncovered in a double stacked row. Bales were sampled at depths of 0–15, 15–30 and 30–45 cm. Time, storage and depth factors had an influence on the chemical components of the LRB except for protein. Weathering had the most effect after 150 d of storage and with LRB stored uncovered and in the top 15-cm layer. Significant (P < 0.05) interactions in in vitro dry matter digestibilities were found for storage time × depth of sample, storage time × storage methods and for depth of sample × storage methods. Under the conditions of our experiments, weathering damages were limited to the superficial layer of LRB left uncovered. Key words: Large round bales, weathering, storage, moisture

White and yellow popcorn were stored in different containers at high temperature (35°C) and high relative humidity (85%) conditions for 3 months. Gradual decreases in popping volumes with the lengthening of storage time were observed in both white and yellow popcorn. Internal mold infection was quite low in both white (5.7%) and yellow (3.0%) popcorn at the beginning of storage tests. Few differences were found in total infection levels up to 60 days of storage, except that the Aspergillus glaucus group became established in place of field fungi. A gradual increase in mold infection levels was then observed during the remaining 30 days of storage. Visible mold growth was also observed on the tips of some kernels by the end of storage studies. Internal mold infection in white popcorn stored in an open container was lower (18.3%) than white popcorn stored in a closed plastic bag (75.0%) and closed plastic jar (85.3%), whereas the internal mold infection in yellow popcorn stored in an open container was higher (23.3%) than yellow popcorn stored in a closed plastic bag (6.3%) and closed plastic jar (2.6%). The A. glaucus group were the predominant molds found at the end of storage tests. The ability of toxigenic molds to invade the popcorn was determined using a dry spore inoculum. None of the inoculated molds, which included Aspergillus flavus, Penicillium martensii, and Penicillium viridicatum, were able to invade the popcorn during storage. However, the A. glaucus group predominated at the end of storage tests in the inoculated samples.

ABSTRACT: This study aimed to evaluate the effect of using lycopene and organic minerals in diets for laying hens on the egg quality and stability of eggs stored for 30 days under different storage environments. An entirely randomized design was adopted in 2x3x3 factorial scheme (mineral sources x lycopene levels x storage periods) with six replicates of eight hens per experimental unit. The experimental diets were: feed containing inorganic minerals (IM) without added lycopene; IM with added lycopene (400mg kg-1); IM with added lycopene (800mg kg-1); organic minerals (OM) without added lycopene; OM with added lycopene (400mg kg-1); OM with added lycopene (800mg kg-1). After 112 days of feeding experimental diets, it was selected 60 eggs treatment-1, which were later labeled, stored in room and refrigerated temperature, and subjected to different storage periods (0, 15 and 30 days). Variables analyzed were: Haugh unit, yolk index, yolk color, albumen and yolk pH, and lipid oxidation (TBARS). Stability of eggs is not altered as a function of mineral sources and levels of lycopene studied. However, increasing storage time affects the quality of the eggs of laying hens at both storage conditions.

The objective of this study was to evaluate the influence of the milking method, the storage conditions and the SCC (Somatic Cell Count) increase on the quality of raw milk. Monthly evaluations were performed out over a year in 21 tanks by monitoring the refrigeration temperature and the storage time of the milk in the tank. The tanks were grouped into three temperature levels. Milk storage time intervals were established in each tank: up to 24 h of storage; between 24 and 48 h; and above 48 h. The effect of SCC on the composition was evaluated in three categories: Low SCC; Medium SCC; High SCC. In the analyzed period, 10.8 % presented low SCC, followed by 46.5 % with medium SCC, while 42.7 % had high SCC. There was a positive correlation between SCC and protein, and a negative correlation between SCC and lactose. It is concluded that the milking method does not influence the microbial contamination of the milk; however, longer storage time and increased temperature influenced an increase in microorganism counts in milk. In evaluating the hygienic/sanitary quality of the milk, 42.7 % had high SCC and the total bacterial counts presented values above the values recommended by legislation.

Abstract: Jatropha seeds are classified as orthodox. However, since it is an oil seed species, adequate storage conditions are required to ensure their longevity. The objective of this work was to evaluate the physiological quality of jatropha seeds stored in different environments and packaging, for periods of 3, 9 and 15 months. Three types of seed packaging bags (high density plastic bag, aluminized envelope and multiwall paper bag) were used, and the storage environments were cold and dry chamber (20 °C and 15% RH, constant), refrigerator (7 ± 3 °C, 48 ± 8% RH) and laboratory conditions (25 ± 3 °C, 51 ± 7% RH). The initial moisture content and seed germination were 7.1% and 89%, respectively. During storage, the physiological quality (germination and vigor) and moisture content of the seeds were evaluated. Seed water content ranged from 3.3 to 7.7%, depending on the permeability of the packaging and the storage environment. The highest longevity (15 months) without loss of viability was observed for jatropha seeds with initial moisture of 7.1%, packed in semipermeable plastic. Seed vigor was maintained, regardless of the environment and the type of packaging used, for up to nine months of storage.

Abstract The detection of SARS-CoV-2 infection is the premise of quarantine. In many countries or areas, samples need to be shipped or inactivated before SARS-CoV-2 testing. In this study, we checked the influence of sample storage conditions on SARS-CoV-2 nucleic acid testing results, including sample inactivation time, storage temperature, and storage time. All of these conditions caused an increase in the cycle threshold values of the nucleic acid tests and led to the misclassification of at least 10.2% of positive cases as negative or suspected. The results highlight the importance of immediate testing of samples for SARS-CoV-2 nucleic acid detection.

Weathering and mold growth in alfalfa (Medicago sativa L.) hay stored at 20% moisture in large round bales was extensive. The losses of dry matter were 9, 40 and 30%, respectively, for hay stored inside, outside and partially protected. Prolonged storage reduced the quality of the edible portion of hay. AbstractThe study evaluated the effect of storage time and conditions of nutritional pellets (NP) containingDuddingtonia flagranschlamydospores on itsin vitrotrapping ability againstHaemonchus contortusL3 larvae. The treated batch (200 NP) contained 4 ×  106chlamydospores of the FTH0-8 strain, whereas the control batch (200 NP) was produced without spores. Both NP batches were exposed to four experimental storage conditions: (T1) shelves (indoors); (T2) refrigeration (4°C); (T3) outdoors under a roof; and (T4) 100% outdoors. Each group comprised 48 NP with spores and 48 NP without spores (control). The ability ofD. flagransspores to trapH. contortusL3 larvae was evaluated for 8 weeks for each storage condition. For that purpose, six randomly selected NP with spores were compared to their respective control NP. Each NP was individually crushed. The crushed material (1 g) was placed on the surface of a 2% water agar plate with 200H. contortusL3 larvae. Plates were sealed and were incubated at room temperature for 8 days. The whole content of every plate was transferred to a Baermann apparatus to recover the remaining larvae. There was a clear larval reduction in the NP with spores, compared to the respective control NP in the four storage conditions (P< 0.05). The mean reductions ( ± SEM) of the storage conditions were 67 ± 4.9 (T2), 77 ± 6.1 (T1), 81.5 ± 3.8 (T4) and 82.1 ± 2.5 (T3). Larval reductions were similar at all times and were not affected by storage conditions or storage time (R2< 0.2;P>0.05). The long-term shelf-life of the chlamydospores in the NP suggests that this spore dosage technology is a viable option.
