ABSTRACT Sponges are one source of producing bioactive components from the sea. Bacteria Symbiosis with sponges are thought to have the potential to produce bioactive compounds that have been isolated from sponges. One of the potential of bioactive compounds which have been found and developed from sponges was antibacterial. The aim of this study was to determine the antibacterial activity of bacteria associated with Phyllospongia lamellose sponges against pathogenic bacteria namely Staphylococcus aureus and Esherichia coli and then proceed with identification of biochemistry in isolates which showed the greatest inhibitory activity. Three isolates of sponges symbiont bacteria were obtained through the isolation and purification stage which were then followed by testing of antibacterial activity with paper disk diffusion method. The antibacterial test results showed the diameter of the inhibitory zone against Staphylococcus aureus, were: SL1(8.67 mm), SL2 (9.33 mm) and SL3(9.00 mm) categorized as medium. Whereas the inhibiotion zone shown on Esherichia coli bacteria is also classified as medium, were: SL1(9.67 mm), SL2 (9.00 mm) and SL3 (9.33 mm). The three isolates continued to the identification stage biochemically. Each isolation was assumed as follows: Desulfotomaculum (SL1), Brochothrix (SL2) and Sulfidobacillus (SL3). Keyword         :Phyllospongia lamellose, isolation, biochemically identification ABSTRAK Spons merupakan salah satu sumber penghasil komponen bioaktif yang berasal dari laut. Bakteri yang bersimbiosis dengan spons diduga memiliki potensi dalam menghasilkan senyawa-senyawa bioaktif yang telah diisolasi dari spons. Potensi senyawa bioaktif yang telah ditemukan dan dikembangkan dari spons salah satunya ialah sebagai antibakteri. Penelitian ini bertujuan untuk mengetahui aktivitas antibakteri dari bakteri yang berasosiasi dengan spons Phyllospongia lamellose terhadap bakteri patogen yakni Staphylococcus aureus dan Escherichia coli dan kemudian dilanjutkan dengan identifikasi secara biokimia pada isolat yang menunjukkan daya hambat terbesar. Diperoleh 3 isolat bakteri simbion spons melalui tahap isolasi dan purifikasi yang kemudian dilanjutkan dengan pengujian aktivitas antibakteri melalui metode difusi kertas cakram. Hasil uji antibakteri menunjukkan diameter zona hambat terhadap bakteri Staphylococcus aureus yaitu: SL1(8.67 mm), SL2(9.33 mm) dan SL3 (9.00 mm) termasuk dalam kategori sedang. Sedangkan zona hambat yang ditunjukkan terhadap bakteri Escherichia coli juga tergolong kategori sedang yaitu: SL1(9.67 mm), SL2(9.00 mm) dan SL3 (9.33 mm). Ketiga isolat dilanjutkan ke tahap identifikasi secara biokimia. Masing-masing isolat diduga sebagai berikut: Desulfotomaculum (SL1), Brochothrix (SL2) dan Sulfidobacillus (SL3). Kata kunci          :Phyllospongia lamellose, isolasi, identifikasi biokimia.

Black tiger shrimp Penaeus monodon post larvae were challenged with Vibrio harveyi and butanol extract of selected marine sponge Aaptos aaptos to determine its antibacterial bioactive potential in vibriosis treatment. Based on the preliminary toxicity study, the A. aaptos butanol extract with concentrations of 31.25, 62.5, and 125 mg/L were selected in the study. Black tiger shrimp post larvae were challenged with V. harveyi at 107 cfu/mL and immersed A. aaptos butanol extract with the concentration of 125 mg/L showed significantly in (P<0.05) decrease mortality of the post larvae treated. Besides at this concentration, V. harveyi population in the rearing water and the post larvae treated decreased compared to control (untreated post larvae). Histological observation indicated that there was no changing on hepatopancreas of the black tiger shrimp post larvae. Based on this result, it is suggested that the butanol extract of A. aaptos is a potential bioactive compounds source in the treatment of vibriosis which may replaced the current antibiotics application.

AbstractThe development of alternatives to antibiotics is crucial to limiting the incidence of antimicrobial resistance, especially in prophylactic and metaphylactic use to control post-weaning diarrhea (PWD). Feed additives, including bioactive compounds, could be a promising alternative. This study aimed to test two bioactive compounds, sodium salicylate (SA) and a chestnut extract (CE) containing hydrolysable tannins, on the occurrence of PWD. At weaning, 72 piglets were assigned to four treatments that combined two factors: CE supplementation (with 2% of CE (CE+) or without (CE-)) and SA supplementation (with 35 mg/kg BW of SA (SA+) or without (SA-)). Then, 4 days after weaning, all piglets were infected with a suspension at 108 CFU/ml of enterotoxigenic Escherichia coli (ETEC F4ac). Each piglet had free access to an electrolyte solution containing, or not, SA. This SA supplementation was administered for 5 days (i.e., from the day of infection (day 0) to 4 days post-infection (day 4). During the 2 weeks post-infection, supplementation with SA had no effect (P > 0.05) on growth performances nor on fecal scores. A significant SA × time interaction (P < 0.01) for fecal scores and the percentage of diarrhea indicated that piglets with SA did not recover faster and did have a second episode of diarrhea. In contrast to SA treatment, inclusion of CE increased (P < 0.05) growth performances and feed intake. In the first week post-infection, CE decreased (P < 0.001) the overall fecal scores, the percentage of piglets with diarrhea, the days in diarrhea, and ETEC shedding in the feces. There was a SA×CE interaction (P < 0.05) for ETEC shedding, suggesting a negative effect of combining SA with CE. This study highlighted that, in contrast to SA, CE could represent a promising alternative to antibiotics immediately after weaning for improving growth performance and reducing PWD.

Medicinal plants have served as sources of medicine to treat and suppress the diseases, because many pathogens are gaining resistance to the current synthetic drugs. In addition, high cost and adverse side effects are commonly associated with popular Synthetic drugs. Therefore, there is need for continuous search for new drugs in order to overcome this emerging resistance. Plants synthesize bioactive compounds which are of great potential in agriculture, antimicrobial and anti-insect activity. The concentration of bioactive compounds in each plant species depends on the environmental conditions, age of the plant, relative humidity of harvested materials and method of extraction. Little is known on the phytochemical and antimicrobial potential of Alba and Rosea cultivars of Catharanthus roseus ethanol extracts. The leaves of Alba and Rosea cultivars were investigated for their phytochemical and antimicrobial properties. The study was conducted at Maseno University, Kenya. Plant Leaves were collected around Maseno University. Leaves of Alba and Rosea cultivars of Catharanthus roseus were air-dried in the shade, thereafter crushed into powder and ethanol extraction done using the Rotary evaporator. Antimicrobial activity of the pathogenic microorganisms was Candida albicans and Escherichia coli.  Disc diffusion method was used for antimicrobial tests. concentrations of ethanol leaf extracts consisted of 2.5, 5and 7.5 mg/mL with three replications.  Data on growth inhibition were subjected to analysis of variance (ANOVA) . Phytochemical analysis revealed the presence of tannins, flavonoids, terpenoids, saponins, alkaloids and phenols except steroids and glycosides. The ethanol leaf extracts were active against Candida albicans and and Escherichia coli. Alba leaves extracts showed higher inhibitory zones compared to Rosea leaves. The observed differences in antimicrobial activity could be due to differences in cell wall synthesis, structure and composition. The results of present study further confirm the use of these plants traditionally for the treatment of different ailments.

Bacteria of the genus Thermoactinomyces have the ability to produce antibacterial bioactive compounds. This bioactive compound can be used for combating diarrheal agents such as Escherichia coli and Staphylococcus aureus. This study aims to determine the antibacterial activity of the metabolite extract from Thermoactinomyces sp. (H24) against Escherichia coli and Staphylococcus aureus. Methanol was used as a solvent for the extraction of bacterial bioactive compounds. Antibacterial activity was analyzed by the diffusion method with several extract concentrations (0.75 mL, 1.5 mL, 2.25 mL, and 3 mL), 10% DMSO as the negative control, and ciprofloxacin as the positive control. Our result shows that Termoactinomyces sp. (H24) extract has an inhibitory effect on the growth of  E. coli and S. aureus with an effective concentration of 2.25 mL (inhibition strength: very strong).

ABSTRACTLissoclinum badium is a type of ascidian that contains bioactive compounds. This study aims to determine of presence of antibacterial activity from extracts and fractions of Lissoclinum badium collected from Mantehage Island Manado against Escherichia coli and Staphylococcus aureus bacteria. Samples were extracted by maceration method using 95% ethanol solvent and fractionated using solvents of chloroform, n-hexane and methanol. Antibacterial activity was carried out by the disk diffusion agar method. The results showed that the ethanol extracts an methanol fraction had activity to inhibit the growth of Escherichia coli bacteria with strong category. Meanwhile, against the Staphylococcus aureus the ethanol extracts, chloroform and n-hexane fractions had ability to inhibit the growth of bacteria with weak category.. Keywords: Antibacterial activity, Lissoclinum badium, Escherichia coli, Staphylococcus aureus.  ABSTRAKLissoclinum badium merupakan salah satu jenis tunikata yang memiliki kandungan senyawa bioaktif yang dapat digunakan untuk pengobatan berbagai penyakit. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antibakteri dari ekstrak dan fraksi  Lissoclinum badium yang diperoleh dari Pulau Mantehage Manado terhadap Escherichia coli dan Staphylococcus aureus. Sampel diekstraksi dengan metode maserasi menggunakan pelarut etanol 95% dan fraksinasi menggunakan pelarut metanol, kloroform, dan n-heksan. Aktivitas antibakteri dilakukan dengan  metode difusi agar cakram kertas. Hasil penelitian menunjukkan bahwa ekstrak etanol dan fraksi metanol memiliki aktivitas untuk menghambat bakteri Escherichia coli dan Staphylococcus aureus dengan daya hambat kuat. Sedangkan untuk fraksi kloroform dan fraksi n-heksan memiliki aktivitas untuk menghambat bakteri Staphylococcus aureus saja dengan daya hambat sedang. Kata kunci: Aktivitas antibakteri, Lissoclinum badium, Escherichia coli, Staphylococcus aureus.
