Abstract Digestibility of AA in feed ingredients for pigs has been generally determined by feeding semipurified diets containing test ingredients as a sole source of nitrogen. However, malnutrition caused by feeding semi-purified diets may affect the digestibility of AA. Therefore, the hypothesis of this study was that the standardized ileal digestibility (SID) of AA in distillers’ dried grains with solubles (DDGS) are not affected by the addition of casein in experimental diets. In Exp. 1, 20 growing pigs were allotted to dietary treatments including 2 diets containing either 480 g/kg DDGS or 308 g/kg DDGS and 60 g/kg casein. The SID of Lys and Phe for DDGS in the diet containing casein were greater (P < 0.05) than those without casein. Exp. 2 was conducted with 20 growing pigs assigned to dietary treatments containing 0, 55, 110, or 165 g/kg casein while the concentration of DDGS decreased at 466.8, 311.2, 155.6, or 0 g/kg. The SID of most indispensable AA in DDGS linearly (P < 0.05) decreased with increasing concentration of casein. Experiment 3 was conducted to verify that the results of Exp. 1 were affected by the addition of casein or by the concentration of DDGS or both. Twenty growing pigs were assigned to dietary treatments prepared as a 2 × 2 factorial arrangement with the concentration of DDGS at 466.8 or 155.6 g/kg and casein at 0 or 110 g/kg. The SID of most indispensable AA in DDGS at 466.8 g/kg were greater (P < 0.01) than in DDGS at 155.6 g/kg regardless of dietary casein. Overall, the addition of 60 g/kg casein to experimental diets may increase the SID of AA in low-protein quality ingredients but the addition of higher concentration of casein and low concentration of dietary protein from test ingredients may decrease the SID of AA.

Vegetarian diets provide an abundance of nutrients when carefully planned. However, vegetarian diets may have lower protein quality compared to omnivorous diets, a reflection of less favorable amino acid profiles and bioavailability. Hence, the current recommended dietary allowance for protein may not be adequate for some vegetarian populations. The purpose of this study was to determine dietary protein quality using the DIAAS (Digestible Indispensable Amino Acid Score) method in vegetarian and omnivore endurance athletes. DIAAS scores reflect the true ileal digestibility of the indispensable amino acids that are present in food items, and these scores can be used to compute the available protein in diet plans. Thirty-eight omnivores and 22 vegetarians submitted seven-day food records that were analyzed for nutrient content, and DIAAS scores were computed by diet group. Average available protein (g) was compared along with participants’ lean body mass and strength (quantified using the peak torque of leg extension). DIAAS scores and available protein were higher for omnivorous versus vegetarian athletes (+11% and +43%, respectively, p < 0.05). Omnivorous participants had significantly higher lean body mass than vegetarian participants (+14%), and significant correlations existed between available protein and strength (r = 0.314) and available protein and lean body mass (r = 0.541). Based upon available protein, as determined through the DIAAS, vegetarian athletes in this study would need to consume, on average, an additional 10 g protein daily to reach the recommended intake for protein (1.2 g/kg/d). An additional 22 g protein daily would be needed to achieve an intake of 1.4 g/kg/d, the upper end of the recommended intake range.

AbstractDiet composition, especially the relative abundance of key macronutrients, is well known to affect animal wellbeing by changing reproductive output, metabolism and length of life. However, less attention has been paid to the ways the quality of these nutrients modify these macronutrient interactions. Nutritional Geometry can be used to model the effects of multiple dietary components on life-history traits and to compare these responses when diet quality is varied. Previous studies have shown that dietary protein quality can be increased for egg production in Drosophila melanogaster by matching the dietary amino acid proportions to the balance of amino acids used by the sum of proteins in the fly’s in silico translated exome. Here, we show that dietary protein quality dramatically alters the effect of protein quantity on female reproduction across a broad range of diets varying in both protein and carbohydrate concentrations. These data show that when sources of ingredients vary, their relative value to the consumer can vastly differ and yield very different physiological outcomes. Such variations could be particularly important for meta analyses that look to draw generalisable conclusions from diverse studies.

ABSTRACT Background In general, pulse protein is limiting in the indispensable amino acid methionine, and antinutritional factors in pulses can affect methionine bioavailability. Complementation with grains such as rice can improve pulse protein quality, but knowledge of methionine bioavailability in pulses and grains is necessary to correct for available methionine when planning and assessing dietary protein intake. Objectives The study objectives were to determine the bioavailability of methionine in rice and chickpeas separately and to assess the effect of complementation of chickpeas and rice. Methods Eleven healthy young men (&lt;30 y, BMI &lt;25 kg/m2) were studied in a repeated-measures design using the indicator amino acid oxidation (IAAO) method, with l-[1-13C]phenylalanine as the indicator. Each received 7 or 10 methionine intakes in random order: 4 intakes of l-methionine—0.5, 1, 2, and 3 mg⋅kg−1⋅d−1 (reference diet); 3 intakes of methionine from rice and from chickpeas; and 3 intakes from the mixed meal of chickpeas plus rice (test diets). The bioavailability of methionine and the effect of complementation were assessed by comparing the IAAO response to varying intakes of methionine in rice, in cooked Canadian chickpeas, and in rice plus chickpeas combined compared with the IAAO response to l-methionine intakes in the reference protein (crystalline amino acid mixture patterned after egg protein) using the slope ratio method. Results The bioavailability of methionine from rice and from chickpeas was 100% and 63%, respectively. Complementation of cooked chickpeas with rice decreased the oxidation of l-[1-13C]phenylalanine by up to 14% (P &lt; 0.05), suggesting an improved protein quality of the combined chickpeas plus rice protein. Conclusions When chickpeas are the main protein source in the diet of young adult men, the combination of rice and chickpeas in a 3:1 ratio is recommended to improve dietary protein quality. The WHO (2007) Technical Report on protein and amino acid requirements in human nutrition states that the best estimate for a population average requirement is 105 mg nitrogen/kg body weight per day, or 0·66 g protein/kg body weight per day. In many developing countries protein intake falls significantly short of these values. Apart from protein quantity, protein quality including bioavailability and digestibility, from different food sources, are currently on the global agenda. The 1st International Symposium on Dietary Protein for Human Health held in Auckland, in March 2011, and the consecutive Food and Agricultural Organization of the United Nations (FAO) Expert Consultation on Dietary Protein Quality, both highlighted the importance of assessing the quality of protein from different food sources through determination of amino acid content. Throughout the developed world, animal products and cereals are the two most important sources of protein; in developing countries this order is reversed. In low income countries only 3 % of total dietary energy, as an indicator of diet composition, is derived from meat and offal, 11 % from roots and tubers and 6 % from pulses, nuts and oilseeds. The remainder of the dietary energy is mainly derived from cereal-based staple food. Although the production of livestock has increased in developing countries, the consumption of protein in these countries with people consuming the most limited amounts of protein are continually decreasing. Undernutrition, including insufficient consumption of protein, remains a persistent problem in the developing world, and although many diets within these developing countries are deficient in thequantityof protein compared to recommendations, thequalityof the protein also strongly comes into focus.

The current constant pressure on fishmeal production for aquaculture feeds stresses the need to search for alternative sources of protein. A possible protein ingredient is insect meal, being more cost effective than fishmeal and soybean meal. Black soldier fly (Hermetia illucens) larvae meal is used in many studies with positive results regarding the bioproductive performances of salmonids. The balanced amino acid profile is comparable with the one of soybean meal. The crude protein and amino acid digestibility is high and similar to that of fishmeal. For rainbow trout (Oncorhynchus mykiss), the protein biological value of the larvae is qualitatively lower than that of fishmeal. Atlantic salmon fed with experimental diets with larvae meal showed high bioproductive performances and a better protein biological value compared with salmon fed with a standard fishmeal diet. However, the nutritional quality and chemical composition of the larvae can be drastically improved through the feeding media, which make it a very interesting candidate as a substitute for fishmeal. The aim of this review is to present the current stage of H. illucens use in salmonid feeds, detailing the chemical composition and protein quality. Furthermore, the effect of larvae meal based diets on salmonids is presented.

The common bean (Phaseolus vulgaris L.) represents a sustainable and affordable source of protein, namely, to populations with vegetarian dietary habits. Despite the national germplasm genetic diversity, little is known about the Portuguese accessions’ nutritional and protein quality, leading to their underuse in breeding programs. To fill this gap, a representative collection (106 accessions) was cropped under two contrasting environments (traditional versus heat stress) and evaluated in terms of nutritional quality by near-infrared spectroscopy. Protein quality was assessed, under the stressful environment, considering the individual amino acid contents and the activity of trypsin inhibitors through mass spectrometry (LC-MS/MS) and spectrophotometry, respectively. On top of strong genotypic control, the nutritional composition (protein, fat, fiber, moisture and ash) was also highly influenced by the environment and by genotype × environment interaction, with a clear nutritional quality ranking change for the accessions in heat stress conditions. Classified into three clusters, the accessions from the cluster with the highest individual amino acid and protein contents also showed higher trypsin inhibitor activity (TIA). Since different levels of TIA had no translation into contrasting protein digestibility, breeders focusing on common beans’ protein quality improvement, especially under challenging warming climate conditions, may take advantage of this group of accessions.
