Background: Ziziphus genus (jujube) is a fruit, which is commonly found and used in Eastern traditional medicine for various conditions. Immunomodulation and anti-oxidant activities of jujube fruit extract are reported. This study aimed to observe immunomodulation effects of green (unripe) and ripe jujube fruit extract on peripheral blood mononuclear cell (PBMC) proliferation by carboxyfluorescein diacetate (CFSE) fluorescence, cytokine profiling by enzyme-linked immunosorbent assay (ELISA), and intracellular H2O2 level by dichlorodihydrofluorescein diacetate (DCFH-DA) fluorescence. Results: Treatments of PBMC with both green and ripe pulp extracts resulted in non-significant increases, about 1.01 - 1.15 times, in PBMC proliferation compared to the untreated group (p > 0.05). The pro-inflammatory cytokine (IL-1β) and regulatory Th2 cytokine (IL-10) levels were significantly higher in PBMC treated with ripe jujube fruit extract than in the cells stimulated with green jujube extract (p < 0.05). Th1 cytokine (IFN-γ) level was not significantly different between the ripe and green jujube extract. A higher percentage of intracellular H2O2 reduction was observed in PBMC treated with the green pulp extract compared to the ripe pulp extract (p < 0.05). Conclusions: This study provides practical information about an effect of jujube staging on some cytokine inductions. The reduction of intracellular oxidative stress seems to involve these cytokine induction phenomena.

ABSTRACT Leptospira interrogans glycolipoprotein (GLP) has been implicated in pathological and functional derangement seen in leptospirosis. The goal of this study was to evaluate GLP's ability to induce cellular activation, as assessed by cytokine production and expression of surface activation markers. GLP extracted from either pathogenic L. interrogans serovar Copenhageni or nonpathogenic Leptospira biflexa serovar Patoc (GLPp) was used to stimulate peripheral blood mononuclear cell cultures from healthy donors. Supernatant cytokine levels were measured by enzyme-linked immunosorbent assay. Expression of CD69 and HLA-DR on lymphocytes and monocytes, as well as lipopolysaccharide (LPS) binding, were measured by flow cytometry. At 6 h of incubation, GLP induced a significant rise in tumor necrosis factor alpha levels, which dropped progressively until 72 h of incubation. Interleukin-10 peak levels were obtained at between 24 and 48 h, with sustained levels until 72 h of incubation. The response magnitude was proportional to the GLP dose. CD69 expression on T lymphocytes and monocytes increased significantly, as did HLA-DR expression on monocytes. GLPp induced no CD69 or HLA-DR expression. GLP did not block biotinylated LPS binding to monocytes, suggesting that different pathways are used to induce cell activation. In conclusion, GLP induces cellular activation and may play a major role in the pathogenesis of leptospirosis.

Abstrak Latar Belakang : Tuberkulosis paru disebabkan oleh bakteri Mycobacterium tuberculosis, yakni baksil tahan asam yang hidup secara intraseluler dan merupakan penyebab utama kematian penyakit menular. Oleh karenanya, perlu diagnosis dini untuk pencegahan. Sel Th1 sangat berperan pada sistem pertahanan tubuh dalam menghadapi infeksi bakteri intraseluler. Pada tuberkulosis paru, terjadi ketidakseimbangan sistem imun host. Tujuan penelitian ini adalah menganalisis kadar IFN-γ dan IL-10 pada PBMC (peripheral blood mononuclear cell) penderita tuberkulosis aktif, laten dan orang sehat, setelah stimulasi dengan antigen ESAT-6. Metode Penelitian : Subjek penelitian adalah 10 TB aktif, 10 LTBI dan 10 orang sehat, yang diambil dari RS Paru Karang Tembok Surabaya. Dilakukan pengambilan darah vena kemudian diisolasi PBMCnya dan dikultur dengan ESAT-6. Kemudian supernatan diambil untuk dilakukan pemeriksaan kadar IFN-γ dan IL-10 dengan metode pemeriksaan Enzyme Linked Immunosorbent Assay (ELISA).Hasil : Kadar IFN-γ setelah di stimulasi antigen ESAT-6, pada orang sehat berkisar antara 95,84 – 135,47 pg/mL, pada TB laten 74,6 – 142,2 pg/mL dan TB aktif 72,67 – 154,21 pg/mL. Kadar  IL-10 setelah di stimulasi antigen ESAT-6, pada orang sehat berkisar antara 161,89 – 282,57 pg/mL, pada TB laten 145,28 – 218,64 pg/mL dan TB akti 232,05 – 294,07 pg/mL. Hasil analisis menunjukkan terdapat perbedaan bermakna (p=0,000) kadar IL-10 pada ketiga kelompok.Simpulan : Terdapat perbedaan pola sekresi/ kadar IFN-γ dan IL-10 pada PBMC penderita TB aktif, TB laten dan orang sehat. Kata kunci : IFN-γ, IL-10 , TB aktif.

Abstract Background It is well known that Autoimmune thyroid disease is multifactorial with multiple genetic and environmental factors, immune malfunction also incriminated in the development of this disease, The exact pathogenesis of this disease remains unclear despite the fact that the production of autoantibodies destroys self-tolerance and agitate the adaptive immune system. Our study will answer the question is there a difference in Toll like receptor 9 (TLR 9) expression in peripheral blood mononuclear cell (PBMCs) from Grave’s disease patients. Objective to measure TLR9 percentage expression on peripheral blood mononuclear cells in patients with Graves’ disease. Methods 60 subjects were included in this study; 30 with Graves’ disease and 30 healthy individuals as control group. All the patients were subjected to the following: Full history, clinical examination, thyroid functions, Thyroid ultrasound, Radioisotope thyroid scan: to assess uptake of thyroid gland and Toll like receptor 9 (TLR 9) percentage expression on peripheral blood mononuclear cells will be analyzed using flow cytometry technique. Results The present study proved that patients with Graves’ disease had higher levels of percentage expression of TLR 9 on peripheral blood lymphocytes. Conclusion percentage expression of TLR9 on peripheral blood lymphocytes is higher in Graves’ patients.
