Foodborne disease is a major public health problem causing considerable morbidity and mortality annually. In the present study, the antibiotic susceptibility patterns of Escherichia coli isolated from food, cooking utensils and palms of food handlers in some restaurants in Zaria, Nigeria were evaluated. A total of 250 samples (220 food samples, 7 hand samples of food handlers, 10 plate samples within restaurants and 13 spoon samples) were collected from five locations in Zaria, Nigeria and analysed for microbial contaminations using standard microbiological techniques. The antibiotic susceptibility pattern of the isolates was determined using Kirby-Bauer modified disc agar diffusion technique. Results revealed that out of 158 acclaimed Enterobacteriaceae isolates evaluated, 19 % (30) were confirmed to be E. coli, while 81 % were Klebsiella. spp, Citrobacter fruendii, Enterobacter spp, Shigella spp, Salmonella spp, Serratia spp, and Cronobacter sakazaki. The majority of the isolates were resistant to amoxiclav (26.08%), ampicillin (26.08%), tetracycline (26.08%) and metronidazole (13.04%). A 33.3% of the isolates were multidrug-resistant. The E. coli isolates were mostly multiple antibiotic resistance with 43.3% having multiple antibiotic resistance index (MARI) ≥ 0.2. In conclusion, E. coli evolved resistance to ampicillin, Amoxicillin Clavulanic acid, and Tetracycline and other tested antimicrobial drugs which would make the treatment of Escherichia coli infections difficult

The aim of this longitudinal study was to verify the prevalence and antibiotic susceptibility pattern of Escherichia coli (E. coli) in cattle reared on Bathan and intensive farming system in Bangladesh. Fecal materials originated from recto anal junction (RAJ) of 100 cattle used for primary screening on MacConkey agar. The diversities among the pink color colony producing isolates on MacConkey agar were verified by conventional cultural methods and biochemical tests. Phenotypically positive E. coli isolates were further investigated for the variations in the antimicrobial resistance profiles to 10 selected antibiotics, by the disk-diffusion method. This study revealed that the overall prevalence of E. coli was 70% of in the rectal swab sample of cattle. However, the prevalence of E. coli was found significantly higher (p= 0.002) in cattle under intensive farming (84%) than cattle on Bathan (56%). Antibiotic susceptibility pattern shows that among the tested isolates 83%, 73%, 68% and 64% were sensitive to chloramphenicol, gentamicin, ciprofloxacin and ampicillin, respectively. On the other hand, all the 70 (100%) E. coli isolates were found resistant to tetracycline and sulfamethoxazole. A high antibiotic resistance profile was also found against amoxicillin (90%), ampicillin (87%), nalidixic acid (86%) and erythromycin (83%). In total, 24 (34%) isolates were resistant against ?2 antimicrobials. The result clearly shows that antibiotic resistant E. coli isolates are commonly present in cattle of different management systems (intensive and Bathan). Therefore, careful selection of appropriate antibiotics with optimal doses might be ensured to prevent the emergence of antibiotic resistance bacteria.Microbes and Health, June 2017, 6(1): 1-4

Objective: This work aimed to detect the antibiotic susceptibility pattern of Escherichia coli isolated from children, as it is the most predominant pathogen of urinary tract infection (UTI). Methods: About 530 urine samples were collected and tested using the modified Kirby–Bauer disk diffusion method to find the susceptibility pattern of isolated bacteria. Results: Out of a total of 530 samples, 114 (21.50%) showed significant growth. A total of 8 different types of bacteria were isolated from the growth of positive samples. Among the isolates, E. coli 66 (57.8%) was found to be the most predominant organism followed by Klebsiella pneumoniae 18(15.8%), Proteus spp. 10 (8.8%), Staphylococcus aureus 8 (7.0%), Acinetobacter spp. 4 (3.5%), CoNS 4 (3.5%), Enterobacter spp. 2 (1.8%), and Pseudomonas aeruginosa 2 (1.8%). In the present study, out of 66 E. coli, 37 (56.1%) were multidrug-resistant strain. E. coli showed 94.0% resistance to ceftriaxone followed by ceftazidime 86.5% and cefotaxime 70.3%. Imipenem (91.9%) followed by amikacin (89.2%) seems to be the effective drug against UTI causing E. coli in children. Conclusion: Multidrug resistance may possess difficulties with the choice of therapeutic options for the treatment of severe infections.

The study reported isolation, identification and antibiotic susceptibility of Escherichia coli isolated from raw meat from Modakeke and Ile-ife, Osun State, Nigeria, with the view to determining the antibiogram profiling of the bacterial isolates.  In this study, five samples of fresh meat were collected from different abattoirs in Ile-Ife and Modakeke, Osun State. Isolates of Escherichia coli were isolated, identified morphologically based on their growth on nutrient agar and subjected to antibiotic susceptibility test on Mueller Hinton agar. The mean microbial load from the meat samples ranged from 8.85 x 102cfu/ml to 5.77 x 104cfu/ml. A total of 69 E. coli isolates were recovered from the meat sampled. All the isolates appeared cream, translucent, entire, convex, circular, smooth and glistering. The isolates were identified as Gram negative rods, non-motile, lactose fermenters, positive for indole test and negative for citrate utilization test. All the E. coli isolates were resistant to augmentin, ceftriazone, nitrofurantoin and gentamycin. 98.55% of E. coli isolated was resistant to amoxillin and the least resistant was recorded in ofloxacin (8.70%). However, 91.30% of the E. coli isolates was sensitive to ofloxacin, 81.16% to ciprofloxacin and 36.23% to pefloxacin while none was sensitive to augmentin, ceftriazone, nitrofurantoin and gentamycin. A total of 19 different multiple antibiotic resistance patterns were observed among the isolates. Thirty isolates (43.48%) showed multiple antibiotic resistance to 5 and 10 different antibiotic types each. The study concluded that occurrence of E. coli infection is high in the study area with high level of multiple antibiotic resistance.

Objectives: To investigate the prevalence and antibiotic resistance pattern of biofilm-forming Uropathogenic Escherichia coli (UPEC) from urine samples isolated from UTI infected patients of Koshi zonal hospital, Biratnagar.Methods: A total of 51 urine samples from urinary tract infected patients were collected from Koshi zonal hospital, Biratnagar in the period of July to August 2017. Following the isolation and identification of biofilm-forming uropathogenic Escherichia coli, antibiotic susceptibility test was performed by a modified Kirby-Bauer disc diffusion technique. The biofilm detection was done by Congo red agar method.Results: In the present study, 45% of the urine samples showed a predominant growth of E. coli, among which 70% of isolates exhibited positive biofilm formation. Biofilm forming isolates revealed 100%, 87.5%, 75%, 63% and 12.5% resistant to erythromycin, amoxicillin, cefotaxime, levofloxacin, and nitrofurantoin respectively. Approximately 87.5% of biofilm-forming isolates were found multi-drug resistant.Conclusion: The study revealed the major issue of UTI by E. coli which may be due to poor sanitation, not the proper cleanliness of genitals and unsafe sexual intercourse. The multiple antibiotic resistance operon (marORAB) in Escherichia coli controls intrinsic susceptibility and resistance to multiple, structurally different antibiotics and other noxious agents. A plasmid construct with marA cloned in the antisense direction reduced LacZ expression from a constitutively expressed marA::lacZ translational fusion and inhibited the induced expression of LacZ in cells bearing the wild-type repressed fusion. The marA antisense construction also decreased the multiple antibiotic resistance of a Mar mutant. Two antisense phosphorothioate oligonucleotides, one targeted to marO and the other targeted to marA of the mar operon, introduced by heat shock or electroporation reduced LacZ expression in the strain having the marA::lacZ fusion. One antisense oligonucleotide, tested against a Mar mutant of E. coli ML308-225, increased the bactericidal activity of norfloxacin. These studies demonstrate the efficacy of exogenously delivered antisense oligonucleotides targeted to the marRAB operon in inhibiting expression of this chromosomal regulatory locus.

Milk is such a food which can meet almost all nutritional needs of human lives. Raw or unprocessed milk supports the growth of wide variety of microorganisms. The major interests of this study were examining the microbial quality of raw milk collected from different locations of Jessore city in Bangladesh and determining antibiotic susceptibility pattern of some isolated bacteria. To do so, 12 raw milk samples were collected from different areas of Jessore city. Microbial analysis comprised of enumeration of TVC (total viable count), TCC (total coliform count) and TSC (total staphylococcal count). The highest TVC, TCC and TSC were 1.95x109 CFU/ml, 2.5x107 CFU/ml and 1.02x107 CFU/ml respectively. Prevalent bacterial populations were Klebsiella spp., Enterobacter spp., Shigella spp. Staphylococcus spp., Escherichia coli and Citrobacter spp. In order to observe the antibiotic susceptibility pattern, the antibiotic sensitivity test was performed for some randomly selected isolates of E. coli and Klebsiella spp. More than 90% isolates of Klebsiella spp. were found to be resistant against Erythromycin whereas more than 90% isolates were sensitive against Imipenem. On the other hand, 100% E. coli isolates were observed as resistant against Erythromycin and in case of Trimethopreme 100% isolates were sensitive. Multidrug resistance pattern was also found. These results suggest the necessity of hygienic practices during handling, processing and post-processing of raw milk to improve the microbiological quality and safety of raw milk.DOI: http://dx.doi.org/10.3329/bjvm.v13i1.23723Bangl.
