In vitro Antioxidant Properties of Certain Indigenous Medicinal Plants From Western Ghats of India

Hypoglycemic and antioxidant properties of extracts of medicinal plants Galega officinalis L. (aboveground part) and yacon (Smallanthus sonchifolius Poepp. & Endl.) (leaves) as potential sources of biologically active substances with antidiabetic action have been studied. The pronounced hypoglycemic effect of Galega officinalis extract, devoid of alkaloids, at a dose of 600 mg/kg in experimental diabetes mellitus (DM) has been proven. The established effect is evidenced by a decrease in the concentration of glucose and glycosylated hemoglobin in the blood, increase glucose tolerance of cells, increase C-peptide and insulin content in the plasma of rats’ blood. The effective hypoglycemic effect of the extract in the studied pathology was confirmed by histological examination of the pancreas. The cytoprotective effect of the studied extract on pancreatic cells at a dose of 1200 mg/kg was experimentally confirmed. In the standard cut area, an increase was found in the number of Langerhans islets, their average area, diameter, volume, and a number of β-cells relative to these indicators in animals with diabetes. Comparative screening of the antioxidant properties of 30, 50, 70, and 96% water–ethanol extracts of yacon indicates the highest potential of 50% water-ethanol extract to block free radicals in in vitro model experiments. The non-alkaloid fraction of Galega officinalis extract showed moderate antioxidant activity and was inferior to yacon extract in its ability to neutralize reactive oxygen species (ROS) and bind metal ions of variable valence. The level of antioxidant potential of the studied extracts is due to differences in the quantitative content of compounds of phenolic nature in their compositions. The obtained data on the biological effects of Galega officinalis extract on the structural and functional state of β-cells of the pancreas and antioxidant properties of Galega officinalis and yacon extracts substantiate the prospects of using these plants to create antidiabetic medicines and functional foods based on them.

The antibacterial activities of the ethanol extracts of 35 medicinal plants were screened against Escherichia coli, Bacillus cereus, and Staphylococcus aureus by following standard antimicrobial susceptibility testing procedures. The ethanol extracts of Coptis chinensis and Glycyrrhiza uralensis showed antibacterial activity against all three test bacterial species. The screened extracts were also tested for their antioxidant activities using the DPPH free radical scavenging activity assay, and for their cytotoxic and antioxidant activity in vitro using an assessment of cell viability on mouse embryonic fibroblast cells. The ethanol extracts of both C. chinensis and G. uralensis showed good radical scavenging antioxidant properties and attenuated the rate of cell death caused by oxidative damage. Our results showed that C. chinensis and G. uralensis are promising sources of natural products with good antibacterial and antioxidant activities.

Abstract Background Porcine reproductive and respiratory syndrome (PRRS) caused by the PRRS virus (PRRSV) results in economic losses in the swine industry globally. Several studies have investigated the use of plant extracts for the prevention and control of PRRS outbreaks. Thai medicinal plants may be useful for treating PRRSV infection in pigs. Therefore, we investigated the in vitro anti-PRRSV and antioxidant properties of seven Thai medicinal plants: Caesalpinia sappan Linn., Garcinia mangostana Linn., Houttuynia cordata , Perilla frutescens , Clinacanthus nutans , Phyllanthus emblica , and Tiliacora triandra .Results In the antiviral screening test, we observed that the T. triandra extract strongly inhibited the infectivity of PRRSV into MARC-145 cells [virus titer 3.5 median tissue culture infectious doses (TCID 50 )/ml (log 10 )] at 24 h post-infection, whereas the C. sappan Linn. extract strongly inhibited PRRSV replication [virus titer 2.5 TCID 50 /ml (log 10 )] at 72 h postinfection. C. sappan Linn. extract had the highest total phenol content [220.52 mM gallic acid equivalents (GAE)/g] and the lowest half-maximal inhibitory concentration [IC 50 ; 1.17 mg/ml in 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2.58 mg/ml in 2,2-azino-bis (3-ethylbenzothiazo-line-6-sulfonic acid) diammonium salt (ABTS)].Conclusion Our study showed that T. triandra could inhibit the infectivity of PRRSV, whereas C. sappan Linn was the most effective for inhibiting PRRSV replication in MARC-145 cells. This study elucidates the antiviral activities of Thai medicinal plant extracts in vivo . The result promising that the extracts of Thai medicinal plants, especially that of T. triandra and C. sappan Linn, can be developed into pharmaceutical drugs for the prevention of PRRS in pigs.
