Comprehensive Analysis of Differentially Expressed Profiles of mRNA N6-Methyladenosine in Colorectal Cancer

Aim: To comprehensively profile the landscape of the mRNA N6-methyladenosine (m6A) modification in human colorectal cancer (CRC).Methods: Methylated RNA immunoprecipitation sequencing (MeRIP-seq) was explored to compare the difference in mRNA N6-methyladenosine (m6A) methylation between CRC tissues and adjacent normal control (NC) tissue. RNA-sequencing (RNA-seq) was performed to transcribe differentially expressed mRNAs. Conjoint analysis of MeRIP-seq and RNA-seq data was conducted to predict RNA-binding proteins (RBPs).Results: MeRIP-seq identified 1110 differentially m6A methylated sites (DMMSs) and 980 differentially m6A methylated genes (DMMGs) in CRC, with 50.13% of all modified genes showing unique m6A-modified peaks in CRC. RNA-seq showed 915 upregulated genes and 1463 downregulated genes in CRC. QRT-PCR verified the RNA-seq results by detecting the expression of some mRNAs. Conjoint analysis of MeRIP-seq and RNA-seq identified 400 differentially m6A methylated and expressed genes (DEGs), and pathway analysis detected that DMMGs and DEGs were closely related to cancer. After analyzing these DMMGs and DEGs through the GEPIA database, we found that the expression of B3GNT6, DKC1, SRPK1, and RIMKLB were associated with prognosis, and the expression of B3GNT6 and RIMKLB were associated with clinical stage. 17 RBPs were identified based on the DMMGs and DEGs, among which FXR1, FXR2, FMR1, IGF2BP2, IGF2BP3, and SRSF1 were obviously highly expressed in CRC, and FMR1, IGF2BP2, and IGF2BP3 were closely related to methylation, and might be involved in the development of CRC.Conclusion: This study comprehensively profiled m6A modification of mRNAs in CRC, which revealed possible mechanisms of m6A-mediated gene expression regulation.

Consumers are willing to pay a price for explainable, but not for green AI. Evidence from a choice-based conjoint analysis

A major challenge with the increasing use of Artificial Intelligence (AI) applications is to manage the long-term societal impacts of this technology. Two central concerns that have emerged in this respect are that the optimized goals behind the data processing of AI applications usually remain opaque and the energy footprint of their data processing is growing quickly. This study thus explores how much people value the transparency and environmental sustainability of AI using the example of personal AI assistants. The results from a choice-based conjoint analysis with a sample of more than 1.000 respondents from Germany indicate that people hardly care about the energy efficiency of AI; and while they do value transparency through explainable AI, this added value of an application is offset by minor costs. The findings shed light on what kinds of AI people are likely to demand and have important implications for policy and regulation.

Conjoint analysis: the assumptions, applications, concerns, remedies and future research direction

PurposeSince the inception of the conjoint analysis technique in the year 1971, papers addressing the epistemological aspects of conjoint analysis are scant. Hence, this paper attempts to address the vacuum of qualitative discourse addressing the epistemological and methodological aspects of conjoint analysis including different issues, challenges, probable solutions, limitations and future direction of conjoint analysis in the recent decade.Design/methodology/approachFor exploring the methodological and epistemological aspects of conjoint analysis, the seminal papers on conjoint analysis were reviewed. Moreover, the authors' experience for the state-of-art review was also taken into consideration.FindingsThe findings suggest that conjoint analysis that roots back since 1971 has not seen much exploration in Asian regions and is mainly used for new product development in the field of marketing or allied areas. Moreover, the reliability and validity of conjoint analysis is always a matter of concern for the researchers that hinders this technique's wider adaptability. Thus, the paper presents some probable solutions to address the focal issues useful for improved reliability and validity of the conjoint analysis technique.Research limitations/implicationsThis paper attempts to familiarize the researchers with epistemological and methodological aspects of conjoint analysis with certain solutions to evolve beyond existing conjoint analysis dimensions in terms of improved validity, reliability, epistemological and methodological aspects of conjoint analysis (CA). Moreover, it acts as a call for research in different research domains, especially in the Asian continent.Originality/valueThere exist certain seminal research papers on epistemological aspects of conjoint analysis. However, there is a dearth of such attempt in the recent decade addressing the application issues of conjoint analysis incorporating the recent issues as well. Therefore, this paper is an attempt to usher the future researcher to understand the methodological aspects of conjoint analysis. It may prevent them from violating the basic assumptions and methodological threshold. This research technique is preferred equally by academicians and practitioners, thus making it imperative to have clarity beforehand for improved research rigor.

Decision Tool of Medical Endoscope Maintenance Service in Chinese Hospitals: A Conjoint Analysis

Background: Quality and cost of medical device maintenance are dominant factors influencing hospitals’ decision in choosing medical endoscope products. Effective and efficient medical device maintenance are also paramount for providing cost-effective and high quality of medical care. This research aims to facilitate decision-making at hospitals in choosing the suitable endoscope device and the associated maintenance service; it also aims to facilitate decision-making at suppliers to develop the right products and services to fulfill customer demands.Methods: A cross-sectional survey was undertaken in 50 Chinese hospitals, including primary and tertiary hospitals. Moreover, 65 medical staff and 56 medical engineers were recruited from 50 Chinese hospitals. A comprehensive set of attributes were defined and investigated. Conjoint analysis and orthogonal design were used for survey design and statistical analysis. Results: Attribute importance and utility values of decision-making factors were analyzed at the aggregate, occupation, and medical institution levels. (1) At the aggregate level, the most critical factor is "maintenance response" and the least important one is "maintenance efficiency". (2) At the occupation level, medical staff paid more attention to "maintenance response" and medical engineers paid more attention to "maintenance quality". (3) At the medical institution level, Primary hospitals paid more attention to "maintenance price", while tertiary hospitals paid more attention to "maintenance quality". Conclusions: In general, this study provides a more scientific decision-making tool to both hospitals in choosing maintenance service of medical equipment such as endoscopy, and it also helps manufacturers and suppliers improve the after-sales service.

Beef from Calves Finished with a Diet Based on Concentrate Rich in Agro-Industrial By-Products: Acceptability and Quality Label Preferences in Spanish Meat Consumers

Conjoint analysis was used to estimate the relative importance of some of the main extrinsic attributes and quality labels of beef in three Spanish cities (Córdoba, Marbella, and Santa Pola) in a study performed with 300 individuals. Consumers were segmented according to their frequency of consumption. Willingness to pay for different meats was also calculated from the conjoint analysis results. Consumer liking of beef that had been finished with an alternative concentrate rich in agro-industrial by-products and aged for three different durations as compared to conventionally finished beef was also evaluated using the same consumers. The most important attribute for Spanish consumers was the price (28%), followed by origin (25%), animal welfare certification (19%), protected geographical indication (14%), and organic agriculture certification (14%). Most consumers preferred beef from Spain at the lowest possible price and with the highest number of quality labels. Consumers were willing to pay a premium of 1.49, 3.61, and 5.53 EUR over 14 EUR/kg for organic certification, protected geographical indication, and animal welfare certification, respectively. Sensory analysis revealed that, for regular consumers, beef finished with an alternative concentrate rich in agro-industrial by-products offered several hedonic advantages (color, flavor, and tenderness) when compared to beef finished using a conventional diet, while occasional consumers did not find any difference between the two kinds of meat.
