Introduction: The quantitative myasthenia gravis score is a commonly used scale for evaluating muscle weakness associated with myasthenia gravis (MG). It has been reported that some items used in the scale have low discriminative properties. However, there has been no research investigating the applicability of the quantitative MG score (QMGS) in Chinese patients with MG. In addition, the scoring method and ranges of grip strength items in QMGS need to be further evaluated.Methods: This study included 106 Chinese patients with MG, enrolled between September 2020 and February 2021, who were evaluated using the QMGS. Each item in the QMGS was analyzed for distribution. Three methods of evaluating grip strength, grip strength decrement, maximum grip strength, and relative grip strength, were compared. The correlation between the QMG total score minus grip strength score, and three evaluating methods, was analyzed.Results: The grip strength, swallowing, speech, diplopia, ptosis, and facial muscles items showed a clustered distribution. Most patients (94%) presented their maximum grip strength in the first four grip strength measurements. The QMG total score minus the grip strength score had a weak correlation with grip strength decrement (R grip r = 0.276; L grip r = 0.353, both p < 0.05) and moderate correlations with maximum grip strength (R grip r = −0.508; L grip r = −0.507; both p < 0.001) and relative grip strength (R grip r = −0.494; L grip r = −0.497, both p < 0.001).Conclusions: This study suggested that partial items in the QMGS have low discriminative properties for Chinese populations and the maximum grip strength value is the better method to evaluate grip strength compared to the other two scoring methods. Based on the quartiles of maximum grip strength, we propose new scoring ranges for the grip strength items.

In this study, we applied different text-mining methods to the originality scoring of the Unusual Uses Test (UUT) and Just Suppose Test (JST) from the Torrance Tests of Creative Thinking (TTCT)–Verbal. Responses from 102 and 123 participants who completed Form A and Form B, respectively, were scored using three different text-mining methods. The validity of these scoring methods was tested against TTCT’s manual-based scoring and a subjective snapshot scoring method. Results indicated that text-mining systems are applicable to both UUT and JST items across both forms and students’ performance on those items can predict total originality and creativity scores across all six tasks in the TTCT-Verbal. Comparatively, the text-mining methods worked better for UUT than JST. Of the three text-mining models we tested, the Global Vectors for Word Representation (GLoVe) model produced the most reliable and valid scores. These findings indicate that creativity assessment can be done quickly and at a lower cost using text-mining approaches.

Abstract. Bimodality and other types of non-Gaussianity arise in ensemble forecasts of the atmosphere as a result of nonlinear spread across ensemble members. In this paper, bimodality in 50-member ECMWF ENS-extended ensemble forecasts is identified and characterized. Forecasts of 2 m temperature are found to exhibit widespread bimodality well over a derived false-positive rate. In some regions bimodality occurs in excess of 30 % of forecasts, with the largest rates occurring during lead times of 2 to 3 weeks. Bimodality occurs more frequently in the winter hemisphere with indications of baroclinicity being a factor to its development. Additionally, bimodality is more common over the ocean, especially the polar oceans, which may indicate development caused by boundary conditions (such as sea ice). Near the equatorial region, bimodality remains common during either season and follows similar patterns to the Intertropical Convergence Zone (ITCZ), suggesting convection as a possible source for its development. Over some continental regions the modes of the forecasts are separated by up to 15 °C. The probability density for the modes can be up to 4 times greater than at the minimum between the modes, which lies near the ensemble mean. The widespread presence of such bimodality has potentially important implications for decision makers acting on these forecasts. Bimodality also has implications for assessing forecast skill and for statistical postprocessing: several commonly used skill-scoring methods and ensemble dressing methods are found to perform poorly in the presence of bimodality, suggesting the need for improvements in how non-Gaussian ensemble forecasts are evaluated.

Weighted committees allow shareholders, party leaders, etc. to wield different numbers of votes or voting weights as they decide between multiple candidates by a given social choice method. We consider committees that apply scoring methods such as plurality, Borda, or antiplurality rule. Many different weights induce the same mapping from committee members’ preferences to winning candidates. The numbers of respective weight equivalence classes and hence of structurally distinct plurality committees, Borda commitees, etc. differ widely. There are 6, 51, and 5 plurality, Borda, and antiplurality committees, respectively, if three players choose between three candidates and up to 163 (229) committees for scoring rules in between plurality and Borda (Borda and antiplurality). A key implication is that plurality, Borda, and antiplurality rule are much less sensitive to weight changes than other scoring rules. We illustrate the geometry of weight equivalence classes, with a map of all Borda classes, and identify minimal integer representations.

Purpose: Research on the typical and impaired grammatical acquisition of Arabic is limited. This study systematically examined the morphosyntactic abilities of Arabic-speaking children with and without developmental language disorder (DLD) using a novel sentence repetition task. The usefulness of the task as an indicator of DLD in Arabic was determined. Method: A LITMUS (Language Impairment Testing in Multilingual Settings) sentence repetition task was developed in Palestinian Arabic (LITMUS-SR-PA-72) and administered to 30 children with DLD ( M = 61.50 months, SD = 11.27) and 60 age-matched typically developing (TD) children ( M = 63.85 months, SD = 10.16). The task targeted grammatical structures known to be problematic for Arabic-speaking children with DLD (language specific) and children with DLD across languages (language independent). Responses were scored using binary, error, and structural scoring methods. Results: Children with DLD scored below TD children on the LITMUS-SR-PA-72, in general, and in the repetition of language-specific and language-independent structures. The frequency of morphosyntactic errors was higher in the DLD group relative to the TD group. Despite the large similarity of the type of morphosyntactic errors between the two groups, some atypical errors were exclusively produced by the DLD group. The three scoring methods showed good diagnostic power in the discrimination between children with DLD and children without DLD. Conclusions: Sentence repetition was an area of difficulty for Palestinian Arabic–speaking children with DLD. The DLD group demonstrated difficulties with language-specific and language-independent structures, particularly complex sentences with noncanonical word order. Most grammatical errors made by the DLD group resembled those of the TD group and were mostly omissions or substitutions of grammatical affixes or omissions of function words. SR appears to hold promise as a good indicator for the presence or absence of DLD in Arabic. Further validation of these findings using population-based studies is warranted. Immunotherapy targeting the PD-1–PD-L1 axis yielded good results in treating different immunologically ‘‘hot’’ tumors. A phase II study revealed good therapeutic activity of pembrolizumab in selected prostatic carcinoma (PC)-patients. We performed a systematic literature review (PRISMA guidelines), which analyzes the immunohistochemical expression of PD-L1 in human PC samples and highlights the pre-analytical and interpretation variables. Interestingly, 29% acinar PCs, 7% ductal PCs, and 46% neuroendocrine carcinomas/tumors were PD-L1+ on immunohistochemistry. Different scoring methods or cut-off criteria were applied on variable specimen-types, evaluating tumors showing different clinic-pathologic features. The positivity rate of different PD-L1 antibody clones in tumor cells ranged from 3% (SP142) to 50% (ABM4E54), excluding the single case tested for RM-320. The most tested clone was E1L3N, followed by 22C3 (most used for pembrolizumab eligibility), SP263, SP142, and 28-8, which gave the positivity rates of 35%, 11–41% (depending on different scoring systems), 6%, 3%, and 15%, respectively. Other clones were tested in <200 cases. The PD-L1 positivity rate was usually higher in tumors than benign tissues. It was higher in non-tissue microarray specimens (41–50% vs. 15%), as PC cells frequently showed heterogenous or focal PD-L1-staining. PD-L1 was expressed by immune or stromal cells in 12% and 69% cases, respectively. Tumor heterogeneity, inter-institutional preanalytics, and inter-observer interpretation variability may account for result biases.

Long-read direct RNA sequencing developed by Oxford Nanopore Technologies (ONT) is quickly gaining popularity for transcriptome studies, while fast turnaround time and low cost make it an attractive instrument for clinical applications. There is a growing interest to utilize transcriptome data to unravel activated biological processes responsible for disease progression and response to therapies. This trend is of particular interest for precision medicine which aims at single-patient analysis. Here we evaluated whether gene abundances measured by MinION direct RNA sequencing are suited to produce robust estimates of pathway activation for single sample scoring methods. We performed multiple RNA-seq analyses for a single sample that originated from the HepG2 cell line, namely five ONT replicates, and three replicates using Illumina NovaSeq. Two pathway scoring methods were employed—ssGSEA and singscore. We estimated the ONT performance in terms of detected protein-coding genes and average pairwise correlation between pathway activation scores using an exhaustive computational scheme for all combinations of replicates. In brief, we found that at least two ONT replicates are required to obtain reproducible pathway scores for both algorithms. We hope that our findings may be of interest to researchers planning their ONT direct RNA-seq experiments.
