Abstract In this study, the prevalence of Avian orthoavulavirus-1 (AOAV-1) (also commonly known as Newcastle disease virus) was investigated in caged birds kept in bird markets in the Lahore district of Pakistan. A total of 354 swab samples were obtained from 14 different species of clinically healthy birds. The overall virus prevalence was 12.7% in 9 out of the 14 species. Phylogenetic analysis of the complete fusion protein (F) gene showed that 23 isolates from different avian species belonged to sub-genotype VII.2 while three isolates of pigeon origin clustered with sub-genotype XXI.1.2. The VII.2 viruses isolated had a high nucleotide identity to viruses repeatedly isolated from poultry in Pakistan from 2011 to 2018. To date, sub-genotype XXI.1.2 viruses have only been identified in Pakistan. These findings suggest that the Newcastle disease (ND) outbreaks occurring in Pakistan involve multiple hosts and environments. The study emphasises the importance of continuing to monitor multiple avian species for the presence of AOAV-1s and implementing effective ND control strategies.

Global efforts to assess honey bee health show viruses are major stressors that undermine colony performance. Identifying factors that affect virus incidence, such as management practices and landscape context, could aid in slowing virus transmission. Here we surveyed viruses in honey bees from 86 sites in the Pacific Northwest, USA, and tested effects of regional bee density, movement associated with commercial pollination, julian date, and hive management on virus prevalence. We also explored patterns of virus co-occurrence and spatial autocorrelation to identify whether local transmission was a primary driver of pathogen distribution. Our surveys found widespread prevalence of Deformed wing virus (DWV), Sacbrood virus (SBV), and Black queen cell virus (BQCV). BQCV and SBV were most prolific in commercial apiaries, while Chronic bee paralysis virus (CPBV) was more common in hobbyist apiaries than commercial apiaries. DWV was most common in urban landscapes and was best predicted by mite prevalence and julian date, while the incidence of both SBV and BQCV were best predicted by regional apiary density. We did not find evidence of additional spatial autocorrelation for any viruses, although high co-occurrence suggests parallel transmission patterns. Our results support the importance of mite management in slowing virus spread and suggest that greater bee density increases transmission. Our study provides support that viruses are widespread in honey bees and connects known mechanisms of virus transmission to the distribution of pathogens observed across the Pacific Northwest.

A study was conducted to investigate epidemiological aspects of papaya virus E (PpVE), a cytorhabdovirus commonly found in papaya (Carica papaya L.) plantings of Ecuador. Besides papaya, PpVE was found in three Fabaceae weeds, including Rhynchosia minima, Centrosema plumieri and Macroptilium lathyroides; the latter being the species with the highest virus prevalence. Greenhouse experiments showed that in M. lathyroides, single infections of PpVE induce only mild leaf mosaic, whereas in mixed infections with cowpea severe mosaic virus, PpVE contributes to severe mosaic. In papaya, PpVE did not induce noticeable symptoms in single or mixed infections with papaya ringspot virus. Transmission experiments confirmed that whiteflies (Bemisia tabaci) transmit PpVE in a semi-persistent, non-propagative manner.

A month-long wastewater sampling project was conducted along the northeast periphery of Mexico City, specifically in the state of Hidalgo, to assess the presence of SARS-CoV-2. To determine the prevalence of infection and obtain a range of COVID-19 cases in the main metropolitan zones. Viral RNA residues (0–197,655 copies/L) were measured in wastewater from the five central municipalities in the state. By recording the number of RNA viral copies per liter, micro-basins delimitation, demographic and physiological data, an interval of infected people and virus prevalence was estimated using a Monte Carlo model (with 90% confidence) in the micro-basin of five municipalities with metropolitan influence or industrial activity. Our procedure determined that the percentage of the infected population ranges from 1.4% to 41.7%, while the official data reports 0.1–0.3%. This model is proposed as a helpful method of regional epidemiological monitoring through the analysis of viral prevalence.

Abstract Background Hepatitis E virus (HEV) is a nonenveloped RNA virus causing hepatitis E worldwide. The increase in transfusion-transmitted cases of HEV infections from asymptomatic blood donors causing serious illnesses among immunosuppressed recipients has been reported in the past few years. China is one of the most prevalent regions of HEV; as a result, it is important to evaluate the risk of transfusion-transmitted HEV. Methods A total of 1864 serum samples (including demographic characteristics) from blood donors were randomly collected from February to March 2018 in Dali city. Anti-HEV IgG, IgM and IgA antibodies and HEV antigen were examined by enzyme-linked immunosorbent assay (ELISA). HEV RNA was detected by real-time PCR. Multivariable logistic regression modelling was used to examine the risk factors associated with HEV prevalence. Results Overall, the positive rates of anti-HEV IgG, IgM, and IgA antibodies were 13.36% (249/1864), 1.13% (21/1864), and 1.82% (34/1864), respectively. However, none of the 1864 serum samples were HEV antigen positive or HEV RNA positive. Females (16.69%) had a significantly higher HEV seroprevalence than males (13.04%) (odds ratio [OR] 1.34 [95% CI, 1.02–1.75]). Bai (18.85%) donors had a significantly higher HEV seroprevalence than Han (12.21%) blood donors (odds ratio [OR], 1.65 [95% CI, 1.24–2.19] for Bai). Conclusions HEV showed a seroprevalence among blood donors in Yunnan Province, some of which were even recent infections, indicating a threat to the safety of blood transfusions. Whether to formulate a strategy for HEV screening in blood centres needs further research.

Although in humans West Nile virus is mainly the cause of mild or sub-clinical infections, in some cases a neuroinvasive disease may occur predominantly in the elderly. In Italy, several cases of West Nile virus infection are reported every year. Tuscany was the first Italian region where the virus was identified; however, to date only two cases of infection have been reported in humans. This study aimed at evaluating the prevalence of antibodies against West Nile virus in the area of Siena Province to estimate the recent circulation of the virus. Human serum samples collected in Siena between 2016 and 2019 were tested for the presence of antibodies against West Nile virus by ELISA. ELISA positive samples were further evaluated using immunofluorescence, micro neutralization, and plaque reduction neutralization assays. In total, 1.9% (95% CI 1.2–3.1) and 1.4% (95% CI 0.8–2.4) of samples collected in 2016–2017 were positive by ELISA and immunofluorescence assay, respectively. Neutralizing antibodies were found in 0.7% (95% CI 0.3–1.5) of samples. Additionally, 0.9% (95% CI 0.4–1.7) and 0.65% (95% CI 0.3–1.45) of samples collected in 2018–2019 were positive by ELISA and immunofluorescence assay, respectively. The prevalence of neutralizing antibodies was 0.5% (95% CI 0.2–1.3). Although no human cases of West Nile infection were reported in the area between 2016 and 2019 and virus prevalence in the area of Siena Province was as low as less than 1%, the active asymptomatic circulation confirms the potential concern of this emergent virus for human health.
