Abstract The filarial nematode Onchocerca volvulus causes onchocerciasis (river blindness), a neglected tropical disease affecting 21 million people, mostly in Sub-Saharan Africa. Targeting the endosymbiont Wolbachia with antibiotics leads to permanent sterilization and killing of adult worms. The gold standard to assess Wolbachia depletion is the histological examination of adult worms in nodules beginning at 6 months post-treatment. However, nodules can only be used once, limiting the time points to monitor Wolbachia depletion. A diagnostic to longitudinally monitor Wolbachia depletion from microfilariae (MF) at more frequent intervals < 6 months post-treatment would accelerate clinical trials of antiwolbachials. We developed a TaqMan qPCR amplifying the single-copy gene wOvftsZ to quantify Wolbachia from as few as one MF that had migrated from skin biopsies and compared quantification using circular and linearized plasmids or synthetic dsDNA (gBlock®). qPCR for MF from the rodent nematode Litomosoides sigmodontis was used to support the reproducibility and validate the principle. The qPCR using as few as 2 MF from O. volvulus and L. sigmodontis reproducibly quantified Wolbachia. Use of a linearized plasmid standard or synthesized dsDNA resulted in numbers of Wolbachia/MF congruent with biologically plausible estimates in O. volvulus and L. sigmodontis MF. The qPCR assay yielded a median of 48.8 (range 1.5–280.5) Wolbachia/O. volvulus MF. The qPCR is a sensitive tool for quantifying Wolbachia in a few MF from skin biopsies and allows for establishing the qPCR as a surrogate parameter for monitoring Wolbachia depletion in adult worms of new antiwolbachial candidates.

Summary The prevalence of smartwatches and other wearable medical technology has been increasing yearly. These watches offer a sensitive tool for capturing cardiac dysrhythmias and can lead to patients seeking earlier medical advice. This case report highlights the importance of clinicians seeking and using the information provided by wearable medical technology which in this case resulted in both the timely treatment of non-sustained ventricular tachycardia and lung adenocarcinoma.

Background: Accurate surveillance of triatomine household infestation is crucial for Chagas disease vector control. However, no gold standard detection method with high levels of sensitivity or specificity is currently available. Several intrinsic features of triatomine bug behaviour and the lifecycle of Trypanosoma (T.) cruzi lead to deposition of environmental DNA (eDNA) in infested houses. This study evaluated the use of FTA cards and cotton-tipped swabs as low-technology, cost-effective tools for simultaneous detection of T. cruzi and vector eDNA in the laboratory and field. Methods/Principal Findings: This study had two components: (1) laboratory evaluation and optimisation of QIAcard® FTA® classic cards to detect Rhodnius (R.) prolixus eDNA by altering five different environmental variables (darkness, triatomine number, temperature, feeding status and degradation at ambient temperature); (2) detection of R. prolixus and T. cruzi eDNA from cotton-tipped house wall swabs from an endemic region in Casanare Department, Colombia. eDNA was extracted from all specimens and amplified using a multiplex TaqMan qPCR assay targeting the R. prolixus 12S rRNA gene and T. cruzi satellite DNA region. R. prolixus eDNA from five 3rd/4th instar nymphs was successfully amplified from FTA cards after as little as 15 minutes of contact time under standard insectary conditions. Factors significantly increasing eDNA detection from FTA cards were increasing temperature from 21oC to 27-32oC, triatomine bug density from 1-25 bugs and recent blood-feeding. eDNA was detectable from FTA cards stored at room temperature for at least two weeks. In cotton-tipped swabs from the field, the sensitivity and specificity of R. prolixus eDNA detection was 60.6% (n=20/33) and 100% (n=33/33), respectively. T. cruzi eDNA was amplified from 93.9% (n=31/33) of infested houses. Conclusions/Significance: FTA cards are a highly sensitive tool for entomological surveillance of R. prolixus and exhibit little variability under different environmental conditions. Additionally, cotton-tipped swabs are a relatively sensitive tool for entomological and parasitological surveillance of R. prolixus and T. cruzi in situ, but more feasible due to low cost. Both methods could be utilised by citizen science initiatives to contribute to the control of Chagas disease in endemic communities.

Reservoirs are dynamic ecosystems subject to different pressures that influence and compromise their ecological structure. The main objective of this study was to evaluate the potential of using the macroinvertebrate to assess the water quality of four reservoirs (one site in Miranda—M and Pocinho—P; four sites in Aguieira—Ag1 to Ag4; and five sites in Alqueva—Al1 to Al5). The sites were sampled in autumn 2018 (A18), spring and autumn 2019 (S19 and A19) and spring 2020 (S20). In situ physical and chemical parameters were measured and a sample of water and macroinvertebrate were collected for further analyses. Total phosphorus exceeded the allowed concentrations (maximum values recorded: M—0.13 mg/L, P—0.09 mg/L, Ag3—0.22 mg/L and Al5—0.18 mg/L). Total abundance varied between 4 and 3088. Taxonomic richness was always low, between 1 and 12 taxa. The highest Shannon–Wiener value (1.91) was recorded in Ag1_A18 and Al2_A18. Pielou’s evenness varied widely across all reservoirs, from 0.06 to 0.92. Almost all the organisms found were associated with polluted water, according to the index ratings. Organisms tolerant of disturbances (e.g., Chironomidae and Oligochaeta) were associated with sites with the worst water quality, according to the Water Framework Directive (WFD), (M, Ag3, Ag4 and Al5) while organisms with moderate tolerance to disturbances (e.g., Cordullidae and Polycentropodidae) were associated with sites with better water quality (P, Ag1, Ag2 and Al1 to Al4). The macrozoobenthos index (MZB) used proved to be a sensitive tool to Portuguese reservoirs, corroborating most of the results obtained in the remaining analyses, as well as providing a clear ecological potential complementing the analysis carried out by the WFD. Based on this, the macroinvertebrate community appeared to be sensitive and able to characterize the reservoirs’ water quality.

This study presents a novel perspective for the study of functional lateralization in a virtual reality environment. In the model study of handedness, the recognition of the dominant and non-dominant hand in real and virtual conditions was assessed using selected tests, such as a real light exposure test of Piórkowski’s apparatus and classical clinical tests, as well as virtual test tasks, in healthy adults. Statistically significant differences between the dominant and non-dominant hand were observed for tests carried out both in classical conditions and the virtual environment. The results and findings of other studies suggest that the virtual reality approach is a very promising and sensitive tool in the research on functional asymmetries in healthy and disease for motor skills and cognition processes.
