The locomotor activity of zebrafish (Danio rerio) has a pronounced, well-studied circadian rhythm. Under constant illumination, the period of free-running locomotor activity in this species usually becomes less than 24 hours. To evaluate the entraining capabilities of slow magnetic variations, zebrafish locomotor activity was evaluated at constant illumination and fluctuating magnetic field with a period of 26.8 hours. Lomb-Scargle periodogram revealed significant free-running rhythms of locomotor activity and related behavioral endpoints with a period close to 27 hours. Obtained results reveal the potential of slow magnetic fluctuations for entrainment of the circadian rhythms in zebrafish. The putative mechanisms responsible for the entrainment are discussed, including the possible role of cryptochromes.

AbstractThe decline in snowpack across the western United States is one of the most pressing threats posed by climate change to regional economies and livelihoods. Earth system models are important tools for exploring past and future snowpack variability, yet their coarse spatial resolutions distort local topography and bias spatial patterns of accumulation and ablation. Here, we explore pattern-based statistical downscaling for spatially-continuous interannual snowpack estimates. We find that a few leading patterns capture the majority of snowpack variability across the western US in observations, reanalyses, and free-running simulations. Pattern-based downscaling methods yield accurate, high resolution maps that correct mean and variance biases in domain-wide simulated snowpack. Methods that use large-scale patterns as both predictors and predictands perform better than those that do not and all are superior to an interpolation-based “delta change” approach. These findings suggest that pattern-based methods are appropriate for downscaling interannual snowpack variability and that using physically meaningful large-scale patterns is more important than the details of any particular downscaling method.

Circadian rhythms are generated by interlocked transcriptional-translational feedback loops of circadian clock genes and their protein products. Mice homozygous for a functional deletion in the Period-2 gene ( Per2m/m mice) exhibit short free-running circadian periods and eventually lose behavioral circadian rhythmicity in constant darkness (DD). We investigated Per2m/m mice in DD for several months and identified a categorical sex difference in the dependence on Per2 for maintenance of circadian rhythms. Nearly all female Per2m/m mice became circadian arrhythmic in DD, whereas free-running rhythms persisted in 37% of males. Remarkably, with extended testing, Per2m/m mice did not remain arrhythmic in DD, but after varying intervals spontaneously recovered robust, free-running circadian rhythms, with periods shorter than those expressed prior to arrhythmia. Spontaneous recovery was strikingly sex-biased, occurring in 95% of females and 33% of males. Castration in adulthood resulted in male Per2m/m mice exhibiting female-like levels of arrhythmia in DD, but did not affect spontaneous recovery. The circadian pacemaker of many gonad-intact males, but not females, can persist in DD for long intervals without a functional PER2 protein; their circadian clocks may be in an unstable equilibrium, incapable of sustaining persistent coherent circadian organization, resulting in transient cycles of circadian organization and arrhythmia.

How daily clocks in the brain are set by light to local environmental time and encode the seasons is not fully understood. The suprachiasmatic nucleus (SCN) is a central circadian clock in mammals that orchestrates physiology and behavior in tune with daily and seasonal light cycles. Here, we have found that optogenetically simulated light input to explanted mouse SCN changes the waveform of the molecular clockworks from sinusoids in free-running conditions to highly asymmetrical shapes with accelerated synthetic (rising) phases and extended degradative (falling) phases marking clock advances and delays at simulated dawn and dusk. Daily waveform changes arise under ex vivo entrainment to simulated winter and summer photoperiods, and to non-24 hr periods. Ex vivo SCN imaging further suggests that acute waveform shifts are greatest in the ventrolateral SCN, while period effects are greatest in the dorsomedial SCN. Thus, circadian entrainment is encoded by SCN clock gene waveform changes that arise from spatiotemporally distinct intrinsic responses within the SCN neural network.

The physicalchemistry properties and crystal structure of were nano-ZrO2+3mol.%Y2O3 determined. The kinetics of the formation of H2 as a result of the decomposition of H2O on the surface of nano-ZrO2+3mol.%Y2O3 was studied. Effects of adsorption and desorption process on ZrO2+3 mol.%Y2O3 nanoparticles were studied at different (T=400÷10000C) temperature. The study of H2 in thermal processes at nano-ZrO2+3 mol.%Y2O3 system increased. Such an increase in H2 generation in comparison with a pure H2O as thermal processes had formedactive centers for H2O decomposition on the surface of the catalyst at the expense of δ-electrons emitted on the surface of nano-ZrO2+3 mol.%Y2O3. This showed that the dimensions of the studied nanoscale particles systems are comparable to the free running distance of energy carriers generated by of nano-ZrO2+3 mol.%Y2O3 as a result of thermal processes. These results are promising for hydrogen generation by waer spliting in near future.

Phosphorylation is one of the most important posttranslational modifications and regulates the physiological process. While recent studies highlight a major role of phosphorylation in the regulation of sleep–wake cycles to a lesser extent, the phosphoproteome in the suprachiasmatic nucleus (SCN) is not well-understood. Herein, we reported that the EA treatment elicits partial reparation of circadian rhythmicity when mice were exposure to constant darkness for long time. We investigated the effects of EA on circadian rhythms in constant darkness between EA stimulation and free-running control. Next, mass spectrometry–based phosphoproteome was utilized to explore the molecular characteristics of EA-induced phosphorylation modification in the SCN. A total of 6,192 distinct phosphosites on 2,488 proteins were quantified. Functional annotation analysis and protein–protein interaction networks demonstrated the most significant enriched phosphor-proteins and phosphosites involved in postsynapse and glutamatergic synapse. The current data indicated that most of the altered molecules are structural proteins. The target proteins, NMDAR and CAMK2, were selected for verification, consistent with the results of LC–MS/MS. These findings revealed a complete profile of phosphorylation modification in response to EA.

Free-running model tests have been carried out based on a scaled chemical tanker ship model, having a guidance, control and navigation system developed and implemented in LabVIEW. In order to make the modelling more flexible and physically more realistic, a modified version of Abkowitz model was introduced. During the identification process, the model’s structure is fixed and its parameters have been obtained using system identification. A global optimization algorithm has been used to search the optimum values and minimize the loss functions. In order to reduce the effect of noise in the variables, different loss functions considering the empirical errors and generalization performance have been defined and implemented in the system identification program. The hydrodynamic coefficients have been identified based on the manoeuvring test data of free-running ship model. Validations of the system identification algorithm were also carried out and the comparisons with experiments demonstrated the effectiveness of the proposed system identification method.
