Intestinal parasites are a global problem, mainly in developing countries. Obtaining information about plants and compounds that can combat gastrointestinal disorders and gastrointestinal symptoms is a fundamental first step in designing new treatment strategies. In this study, we analyzed the antiamoebic activity of the aerial part of Croton sonorae. The dichloromethane fraction of C. sonorae (CsDCMfx) contained flavonoids, terpenes, alkaloids, and glycosides. The ultrastructural morphology of the amoebae treated for 72 h with CsDCMfx was completely abnormal. CsDCMfx reduced erythrophagocytosis of trophozoites and the expression of genes involved in erythrocyte adhesion (gal/galnac lectin) and actin cytoskeleton rearrangement in the phagocytosis pathway (rho1 gtpase and formin1). Interestingly, CsDCMfx decreased the expression of genes involved in Entamoeba histolytica trophozoite pathogenesis, such as cysteine proteases (cp1, cp4, and cp5), sod, pfor, and enolase. These results showed that C. sonorae is a potential source of antiamoebic compounds.

Peganum harmala, a well-known medicinal plant, has been used for several therapeutic purposes as it contains numerous pharmacological active compounds. Our study reported an anti-parasitic activity of P. harmala seed extract against Acanthamoeba triangularis. The stress induced by the extract on the surviving trophozoites for Acanthamoeba encystation and vacuolization was examined by microscopy, and transcriptional expression of Acanthamoeba autophagy-related genes was investigated by quantitative PCR. Our results showed that the surviving trophozoites were not transformed into cysts, and the number of trophozoites with enlarged vacuoles were not significantly different from that of untreated control. Molecular analysis data demonstrated that the mRNA expression of tested AcATG genes, i.e., ATG3, ATG8b, and ATG16, was at a basal level along the treatment. However, upregulation of AcATG16 at 24 h post treatment was observed, which may indicate an autophagic activity of this protein in response to the stress. Altogether, these data revealed the anti-Acanthamoeba activity of P. harmala extract and indicated the association of autophagy mRNA expression and cyst formation under the extract stress, representing a promising plant for future drug development. However, further identification of an active compound and a study of autophagy at the protein level are needed.

Genetically uniform plant material, derived from Lychnis flos-cuculi propagated in vitro, was used for the isolation of 20-hydroxyecdysone and polypodine B and subjected to an evaluation of the antifungal and antiamoebic activity. The activity of 80% aqueous methanolic extracts, their fractions, and isolated ecdysteroids were studied against pathogenic Acanthamoeba castellani. Additionally, a Microtox® acute toxicity assay was performed. It was found that an 80% methanolic fraction of root extract exerts the most potent amoebicidal activity at IC50 of 0.06 mg/mL at the 3rd day of treatment. Both ecdysteroids show comparable activity at IC50 of 0.07 mg/mL. The acute toxicity of 80% fractions at similar concentrations is significantly higher than that of 40% fractions. Crude extracts exhibited moderate antifungal activity, with a minimum inhibitory concentration (MIC) within the range of 1.25–2.5 mg/mL. To the best of our knowledge, the present report is the first to show the biological activity of L. flos-cuculi in terms of the antifungal and antiamoebic activities and acute toxicity. It is also the first isolation of the main ecdysteroids from L. flos-cuculi micropropagated, ecdysteroid-rich plant material.

Background: Entamoeba histolytica is a causative agent of amoebiasis, estimated to cause more than 100,000 deaths per year. Metronidazole is used to treatment of E. histolytica infection. However, this drug has several untoward side effects, the current study was designed to analysis of the bioactive chemical products in extracts of Reseda sphenocleoides leaves and evaluation of anti-amoebic activity in vitro. Methods: Bioactive chemical compounds were tests by GC-MS and FT-IR. Entamoeba histolytica was cultured under xenic conditions in Locke's egg (LE) medium and different concentrations of extracts of R. sphenocleoides were added to cultivated parasites. Results: 11 and 18 bioactive phytochemical compounds were showed in the ethanolic and aqueous extracts of R. sphenocleoides leaves respectively by GC-MS analysis. FT-IR analysis of extracts of R. sphenocleoides leaves proved the presence many functional groups for various phytocompounds. The ethanolic extract of R. sphenocleoides leaves was showed inhibits the growth of E. histolytica in vitro at concentrations 5 and 10 mg/ml after 96 hrs of incubation. While the highest concentration 20 mg/ml used in this study inhibited the E. histolytica in all times of incubation. The aqueous extract showed there was no growth of E. histolytica at the concentrations of 5 and 10 mg/ml after 72 and 48 hrs respectively. However, the higher concentfrations (15 and 20 mg/ml) of the aqueous extract were on the growth of the E. histolytica parasite during the period of incubation. It was found that the minimum inhibitory concentration (MIC) for ethanolic and aqueous extracts of R. sphenocleoides leaves was <15 mg/ml, <10 mg/ml respectively. Conclusion: The results specify that the extracts of R. sphenocleoides has a higher capacity in reducing of E. histolytica number in vitro. Moreover, these results showed that the extracts of R. sphenocleoides contains various bioactive compounds and therefore has various medicinal properties that can be used for the treatment of various diseases.

Cationic carbosilane dendrimers are branched molecules with antimicrobial properties. Their activity has been tested against Acanthamoeba polyphaga, a causative agent of Acanthamoeba keratitis, a severe ocular disease in humans. A. polyphaga trophozoites and cysts were exposed to different noncytotoxic cationic carbosilane dendrimers with proven antiamoebic activity. The effects of treatment on cell surface and cell ultrastructure were examined by scanning and transmission electron microscopy, respectively. Two of the dendrimers tested induced dramatic alterations of cellular ultrastructure in both trophozoites and cysts, including vacuolization, depletion of cytoplasmic contents, and reduced cell size. Additionally, we observed severe alterations of the plasma membrane with membrane blebbing in trophozoites and disruption in cysts. These alterations were also observed with chlorhexidine, a drug used for treatment of Acanthamoeba keratitis. Our results support that these compounds may target membranes, and their action is critical for parasite integrity.
