Pseudohypertriglyceridemia is an overestimation of serum triglyceride levels that may incorrectly lead to a diagnosis of hypertriglyceridemia. Glycerol kinase deficiency is a condition in which glycerol cannot be phosphorylated to glycerol-3-phosphate, resulting in elevated levels of serum glycerol. Laboratory assays that measure triglycerides indirectly may be affected by elevated glyerol levels and incorrectly report serum tryglyceride levels. We present a case of a novel missense mutation in the GK gene leading to isolated glycerol kinase deficiency and pseudohypertriglyceridemia in a male infant of a mother with gestational diabetes. This paper reviews glycerol kinase deficiency, describes the challenges in diagnosing pseudohypertriglyceridemia, and provides suggestions on improving diagnostic accuracy. Additionally, a potential maternal-fetal interaction between gestational diabetes and glycerol kinase deficiency is discussed.

Abstract Objective We investigated direct effects of a therapeutic growth hormone dose on lipolysis, glucose and amino acid metabolism. Methods This crossover microdialysis trial involved six healthy male volunteers receiving single subcutaneous injections of both growth hormone (0.035 mg/kg) and placebo (0.9% sodium chloride). The investigation comprised three test days with standard diet. The first day served for adaptation, the second and third one for determining study data during 9 night hours with or without growth hormone. Abdominal subcutaneous microdialysate and blood were continuously collected and forwarded to a separate room next door where hourly taken samples were centrifuged and frozen until analysed. Results Growth hormone achieved the peak serum level after 3 h followed by a plateau-like course for the next 6 h. Glycerol in microdialysate started to rise 2 h following growth hormone injection achieving significance compared to placebo after 9 h (P<0.05). Serum glycerol increased 4 h after growth hormone administration achieving significance after 6 h (P<0.05). Glucose and amino acid concentrations showed neither in microdialysate nor in serum significant differences between growth hormone and placebo. Serum values of insulin and C-peptide revealed no significant difference between growth hormone and placebo. Summary and Conclusion As the result of a high single subcutaneous dose of GH, persistent lipolysis can be shown in continuously collected microdialysate and blood, but no indication for gluconeogenesis or protein anabolism.

The aim of the present study was to evaluate the effects of the inclusion of mixed (animal and vegetable sources) semi-purified glycerin (MSPG) in broiler diets on performance, litter moisture, serum glycerol concentration, carcass and parts yields, meat quality, and the fatty acid profile. In total, 576 1-day-old male Cobb 500 broiler chicks were distributed in a completely randomised design into four treatments (0%, 2.5%, 5.0% and 7.5% dietary inclusion levels of MSPG, as-fed basis), with six replicate pens of 24 birds each. The apparent metabolisable energy corrected for nitrogen balance and energy metabolisability coefficient values of the MSPG were 12.33 MJ/kg DM and 81.61% respectively. There was no significant (P &gt; 0.05) effect of the MSPG concentration on the performance, and carcass and parts yields or broiler meat-quality variables, except for thigh meat colour, in which the yellowness value (b*) decreased linearly (P &lt; 0.05) with an increasing inclusion level of MSPG. Litter moisture and serum glycerol increased linearly (P &lt; 0.05) and the lipid profile of the thigh and drumstick meat of broilers at 42 days of age was slightly altered (P &lt; 0.05) with an increasing MSPG. Mixed semi-purified glycerin can be used at up to 7.5% in broiler diets without impairing performance, meat quality, carcass and parts yields from 1 to 42 days of age. However, special attention should be paid to litter moisture.

This study examined the effects of pre-exercise carbohydrate ingestion on exercise metabolism and endurance running capacity. Eleven active subjects (VO2 max 49.0 ± 1.7 mL·kg–1·min–1, mean ± SE) performed two exercise trials 15 min after ingesting glucose (G; 1 g·kg body mass–1) and placebo (CON). Each subject ran on a level treadmill for 5 min at 60%, 45 min at 70%, and then at 80% of VO2 max until exhaustion. Serum glucose and plasma insulin reached their peak concentrations (p < 0.01) 15 min after glucose ingestion and declined at the onset of exercise. Serum glycerol concentrations were lower (p < 0.01) in the G trial than in the CON trial after 30 min of exercise to exhaustion. In addition, after 45 min of exercise to exhaustion, the levels of free fatty acids were lower in G than in CON (p < 0.05). No differences were observed in carbohydrate oxidation rates during exercise between treatments (G, 2.53 ± 0.08 g·min–1; CON, 2.40 ± 0.09 g·min–1). Time to exhaustion was 12.8% longer in G (p < 0.01) than in CON. These results suggest that glucose ingestion 15 min before prolonged exercise provides an additional carbohydrate source to the exercising muscle, thus improving endurance running capacity.

The influence of caloric restriction on hepatic glyceraldehyde- and glycerol-metabolizing enzyme activities of young and old mice were studied. Glycerol kinase and cytoplasmic glycerol-3-phosphate dehydrogenase activities were increased in both young and old CR (calorie-restricted) mice when compared with controls, whereas triokinase increased only in old CR mice. Aldehyde dehydrogenase and aldehyde reductase activities in both young and old CR mice were unchanged by caloric restriction. Mitochondrial glycerol-3-phosphate dehydrogenase showed a trend towards an increased activity in old CR mice, whereas a trend towards a decreased activity in alcohol dehydrogenase was observed in both young and old CR mice. Serum glycerol levels decreased in young and old CR mice. Therefore increases in glycerol kinase and glycerol-3-phosphate dehydrogenase were associated with a decrease in fasting blood glycerol levels in CR animals. A prominent role for triokinase in glyceraldehyde metabolism with CR was also observed. The results indicate that long-term caloric restriction induces sustained increases in the capacity for gluconeogenesis from glycerol.

In a randomized, balanced, crossover study each of six fit, adult horses ran on a treadmill at 50% of maximal rate of oxygen consumption for 60 min after being denied access to food for 18 h and then 1) fed corn (51.4 kJ/kg digestible energy), or 2) fed an isocaloric amount of alfalfa 2–3 h before exercise, or 3) not fed before exercise. Feeding corn, compared with fasting, resulted in higher plasma glucose and serum insulin and lower serum nonesterified fatty acid concentrations before exercise ( P < 0.05) and in lower plasma glucose, serum glycerol, and serum nonesterified fatty acid concentrations and higher skeletal muscle utilization of blood-borne glucose during exercise ( P < 0.05). Feeding corn, compared with feeding alfalfa, resulted in higher carbohydrate oxidation and lower lipid oxidation during exercise ( P < 0.05). Feeding a soluble carbohydrate-rich meal (corn) to horses before exercise results in increased muscle utilization of blood-borne glucose and carbohydrate oxidation and in decreased lipid oxidation compared with a meal of insoluble carbohydrate (alfalfa) or not feeding. Carbohydrate feedings did not produce a sparing of muscle glycogen compared with fasting.

Claims that ENDUROX™ enhances performance by altering metabolic responses to exercise were tested. In a double-blind crossover design, 10 male subjects were randomly assigned to consume 400 mg of placebo or 800 mg ENDUROX™ for 7 days. Cycle ergometry was performed for 30 minutes at 25%, followed by 10 min at 65% of peak oxygen consumption. After a 1-week washout period, subjects performed the identical exercise protocol following 7 days of reciprocal supplemental conditions. Expired gases were collected and analyzed continuously for oxygen consumption, minute ventilation, and respiratory exchange ratio. Heart rate, blood pressure, rating of perceived exertion, blood lactate, and serum glycerol data were also collected at regular intervals. A two-way ANOVA with repeated measures revealed no significant main or interaction effects involving group differences (p > 0.05) between trials for any variable during rest, 25% or 65% (VO2 peak), or recovery. Our findings do not support the ergogenic claims for ENDUROX™.
