AbstractConventional dendritic cells (cDCs) scan and integrate environmental cues in almost every tissue, including exogenous metabolic signals. While cDCs are critical in maintaining immune balance, their role in preserving energy homeostasis is unclear. Here, we showed that Batf3-deficient mice lacking conventional type 1 DCs (cDC1s) had increased body weight and adiposity during aging. This led to impaired energy expenditure and glucose tolerance, insulin resistance, dyslipidemia, and liver steatosis. cDC1 deficiency caused adipose tissue inflammation that was preceded by a paucity of NK1.1+ invariant NKT (iNKT) cells. Accordingly, among antigen-presenting cells, cDC1s exhibited notable induction of IFN-γ production by iNKT cells, which plays a metabolically protective role in lean adipose tissue. Flt3L treatment, which expands the dendritic cell (DC) compartment, mitigated diet-induced obesity and hyperlipidemia in a Batf3-dependent manner. This effect was partially mediated by NK1.1+ cells. These results reveal a new critical role for the cDC1-iNKT cell axis in the regulation of adipose tissue homeostasis.

Lysozyme is a ~14 kDa protein present in many mucosal secretions (tears, saliva, and mucus) and tissues of animals and plants, and plays an important role in the innate immunity, providing protection against bacteria, viruses, and fungi. Three main different types of lysozymes are known: the c-type (chicken or conventional type), the g-type (goose type), and the i-type (invertebrate type). It has long been the subject of several applications due to its antimicrobial properties. The problem of antibiotic resistance has stimulated the search for new molecules or new applications of known compounds. The use of lysozyme as an alternative antibiotic is the subject of this review, which covers the results published over the past two decades. This review is focused on the applications of lysozyme in medicine, (the treatment of infectious diseases, wound healing, and anti-biofilm), veterinary, feed, food preservation, and crop protection. It is available from a wide range of sources, in addition to the well-known chicken egg white, and its synergism with other compounds, endowed with antimicrobial activity, are also summarized. An overview of the modified lysozyme applications is provided in the form of tables.

Abstract In the present study, a novel solar still incorporated with fins and phase change material (PCM) based energy storage, was designed. To investigate the influence of fins and energy storage unit, four cases of stills were considered. In case I, a conventional type was considered, whereas square hollow fins were fitted over the basin liner of the still in case II. In addition to fins as in case II, case III employs energy storage unit wherein PCM was packed beneath the basin liner. Case IV was similar to case III except the extension of fins into the storage unit. The addition of fins above the base liner improved the absorber surface area and the extension of the same beneath the basin liner enhanced the storage efficiency. Experiments were carried out on all the four modules with a constant basin water depth of 2 cm. The maximum productivity of the conventional solar still was found to be 3.25 litre/m2/day. On the other hand, the results reveal improvement in productivity of 17.54%, 48.61% and 55.69% with cases II, III and IV, respectively. Although stills with energy storage unit exhibited higher exergy efficiency, the presence of fins in the PCM increases the internal irreversibilities. The cost of water yielded by modified solar still (MSS) used in case IV is proved to be less as compared to conventional solar still (CSS). Further, the payback period of MSS is found to be lesser than that of CSS.

Today transfection of mammalian cell with DNA or RNA construction is the only method for delivering programmed information into the cell nucleus. Electroporation is most commonly used method of transfection in experiments with dendritic cell. The aim of electroporation is to permeabilize the membrane by passing electric impulse through the cell. Due to the increase permeability of the membrane chance DNA or RNA construction getting inside into the cell is increased, wherein survival of the cells is decreased.In the study male mice C57Bl/6 line 2-4 months old were used. From femur bones was isolated 20 × 106 bone marrow cells, which were cultured in 20 mL of complete RPMI-1640 for 7 days. To generate dendritic cells from BM cells, 100 ng/mL of rmFlt3-L was added to culture media at day 0. After 7 days of cultivation, the cell cultures were electroporated with control noncoding plasmids p5 (EP P5) or pmaxCCR9 encoding mouse chemokine receptor CCR9 (EP CCR9). The controls were cell cultures electroporated without any plasmids (mock EP) and cell cultures without electroporation (none EP). 5 × 105 cells were electroporated and resting for 10 minutes. After 10 minutes, cells were harvested and seeded into 24-well plates in 1 mL of culture medium and conditioning medium (1:1). Then, 50 ng/mL of Flt3-L was added to each well. The next day, transfected cells were collected and used for flow cytometry, qRT-PCR analysis.It was found that after electroporation in the groups mock EP, EP P5, EP CCR9 relative amount of live CD11c+ dendritic cells was significantly less than in the non EP group. Moreover, in the EP P5 and EP CCR9 groups the frequency of live CD11c+ dendritic cells was significantly less than in the mock EP group. Expression of the CD86 marker in the EP P5 and EP CCR9 groups was significantly higher than in the non EP and mock EP groups. Expression of the I-AB(MHCII) in the EP CCR9 group on cDC2s was significantly higher than in the non EP group. On plasmacytoid DCs (pDCs) and conventional type 2 DCs (cDC2s) in the EP CCR9 group expression of CCR9 was significantly higher than in the non EP group. Therefore, in this study, we demonstrated the effectiveness of electroporation, accompanied by the decrease in the survival rate and maturation of DCs.

The precise outcome of any indirect casting, depends on the various procedures and materials involved through all its stages. The accurate fit against the prepared surface and the adaptation against the prepared margin depends on how accurately the tooth surface is captured in the impression made and how well it is reproduced in the die This study aims to study and compare the property of fine detail reproduction amongst six die materials.Over a pre-calibrated master die using custom tray, an impression record is made and poured using the six different die materials and the finest line reproduced is visualized under an optical fluorescent microscope at 50X magnification.The data obtained were statistically analysed using one-way ANOVA and subsequently assessed by post-hoc Tukey’s comparison to identify any significant differences between the groups. Epoxy resin die material (1.93 mm) showed a consistently excellent fine detail reproduction, followed by conventional Type V gypsum and Synthetic gypsum (15.91 mm), while Resin modified-Type IV and Type V and conventional Type IV gypsum dies (21.86mm) showed the least accuracy in fine reproduction.

Achieving an optimal cooling of the generator unit is indispensable for high working performance. In this work, computational fluid dynamics analysis on the flow field of the conventional type and silent type of the S688CCS series generator unit is conducted, and design optimization of the generator unit with poor cooling is studied based on flow field analysis results. Flow field simulation results indicate that the total cooling air quantity of the silent type generator unit is lower than that of the conventional type generator unit, and the cooling air quantity of the radiator is also lower than that of the conventional type generator unit, which is not conductive for the cooling of the silent type generator unit. The flow field optimization is achieved by adopting the single variable control method to improve the structure of the fan cover, silent components for silence, adjacent structure, and air inlet grille. The corresponding structure optimization scheme is put forward. After optimization, the air quantity for the cooling radiator of the silent type generator unit is 44.33% higher than that of its original structure, and the total cooling air quantity is higher than that of the conventional type generator unit. The research results in this work can provide a theoretical basis for the design of the cooling air flow path of generator units for achieving an optimal cooling performance.
