Abstract The present study was aimed to evaluate the antioxidant potential and inhibitory effect ofCannabis sativa and Morus nigra against lipid peroxidation in goat brain and liver homogenates. The formation of free radicals, highly reactive oxygen species (ROS) and reactive nitrogen species (RNS) is a normal metabolic process for cellular signaling and countering the antigens. However, they may cause serious damage if they produced at amplified tolls. In addition, metabolic disorders also serve as sources of these reactive species. Although the issue can be addressed through supplements and other phytochemicals. In this study, two plant species were evaluated for their biological potential by employing a spectrum of antioxidant assays. The antioxidant activity was performed by lipid peroxidation assay. The water extract prepared from leaves of Cannabis sativa and Morus nigra showed significant (P<0.05) inhibition as compared to control i.e., 522.6±0.06 and 659.97±0.03 µg/mL against iron-induced lipid peroxidation in goat brain homogenate while the inhibitions were 273.54±0.04 and 309.18±0.05 µg/mL against nitroprusside induced lipid peroxidation of the brain. The iron and nitroprusside induced lipid peroxidation was also significantly inhibited by leaf extracts of Cannabis sativa and Morus nigra in liver homogenates such as 230.63±0.52 and 326.91±0.01 µg/mL (iron-induced) while 300.47±0.07 and 300.47±0.07 µg/mL (nitroprusside induced), respectively. The extracts of Cannabis sativa extract showed promising activity (96.04±0.060%) against DPPH radicals while Morus nigra showed a moderate activity (34.11±0.120%). The results suggest that different accessions ofCannabis sativa and Morus nigra are a potential source of antioxidants and have a therapeutic effect against disease induced by oxidative stress and hence can be used for novel drug discovery and development.

Brain lesions are important cause of morbidity and reduced productivity in goats worldwide. Works on brain lesion in goats are extremely rare in Bangladesh and remain to be investigated. We investigated lesions in goat brains collected from abattoirs in Rajshahi Metropolitan area by gross and microscopic examination. Randomly selected 57 goat brains were investigated of which 24.56% brains showed gross and histopathologic lesions. Prevalence of brain lesions were higher in Jamnapari goats (29.41%) compared to in Black Bengal goats (22.50%) and female goats (26.47%) were more affected than male goats (21.74%). Age-wise prevalence were 31.25%, 21.43%, 12.50%, and 40.00% in the age of up to 2 years, 2-3 years, 3-4 years, and above 4 years old goats, respectively. Goats were more affected in the rainy (35.29%) season in contrast to in the winter (23.53%) and in the summer (10.52%) seasons. The brains collected from goats with Gid disease showed uni or multilocular cysts containing clear fluid and multiple scolices, the rest of the brains were apparently healthy by macroscopic examination. Histopathologically, pyogranuloma, perivascular cuffing, gliosis, neuronal degeneration, hemorrhages, congestion and thrombus were evident. J. Bio-Sci. 29(1): 49-58, 2021 (June)

Hyaluronic acid have a high moisture preservation and biocompatibility characteristic, thus allowing various usage of this substance in pharmaceutical, medicinal, and skin care products. Present manufacturing process of hyaluronic acid requires extraction of animal cells or through other methods utilizing bacteria. The aim of this research is to investigate an alternative source of hyaluronic acid extraction using plant based which is aloe vera (A. Barbadensis).  Three main parts of aloe vera (rind, mesophyll and gel) underwent several steps of extraction process and the result was compared to the sample of hyaluronic acid from goat brain. The evaluation including comparison of total carbohydrates, reducing sugars and degradation using heat treatment. The results of extraction were analyzed using UV-Spectrophotometer at 230 nm and compare to the extraction result of goat brain to ensure the presence of hyaluronic acid. It was found out that the rind part of aloe vera have the highest potential of high content of hyaluronic acid.

The levels of fatty acids, phospholipids and sterols were determined in the brains of she-goat and castrated goat found in Ekiti State, Nigeria by gas chromatography. Results showed that the crude fat levels were 9.98 and 10.2 % in the brains of she-goat and castrated goat respectively. The fatty acid composition of she-goat and castrated goat brain shows that the SFA was 40.6 and 42.7 %, MUFA was 37.1 and 38.7 % and PUFA was 20.9 and 22.3% respectively. The other parameters of she-goat and castrated goat brain were found: PUFA/SFA, 0.490 and 0.548; MUFA/SFA, 0.869 and 0.953; n-6/n-3, 0.775 and 11.7; LA/ALA, 0.876 and 28.0; AA/DGLA, 6.05 and 17.4; EPA/DHA, 1.00 and 5.89 and EPSI (PUFA/MUFA), 0.564 and 0.575 respectively. Phospholipids were present in she-goat and castrated goat with a value range of 2365 and 3047 mg/100g respectively. Among the sterols, only cholesterol was of any significant level with values of 1353 mg/100g (she-goat brain) and 1355 mg/100g (castrated goat brain). Linear correlation at ? = 0.05, df: n-1 showed that no significant difference exists between the crude fats, phospholipids and sterols except in the fatty acids parameters.Bangladesh J. Sci. Ind. Dipeptidylpeptidase-III (DPP-III) from goat brain was purified and characterized using Arginyl-Arginyl-4-methoxy-β-naphthylamide (Arg-Arg-4mβNA) substrate. This enzyme retained its activity in native 10% polyacrylamide gel when stained using Arg-Arg-4mβNA. The activity was significantly increased by 100 mM chloride. Studies for its inhibition with some peptides and chemical inhibitors revealed that Leu-Trp-Met-Arg-Phe-Ala was most potent inhibitor followed by Arg-Phe-Ala and Gly-Phe-Leu. All the studied chemical inhibitors caused 40–50% inhibition at 1 mM. Metal ions helped to regain activity of EDTA pretreated enzyme. ZnCl2 at 50 μM almost completely restored the enzyme activity. Further ZnCl2 and CoCl2 exerted protective effects on EDTA pretreated enzyme for its susceptibility to DTNB inhibition. Therefore, DPP-III is a metalloprotease with the involvement of cysteine residues either located at the catalytic site or involved in regulation.
