ABSTRACT The electrical conductivity test stands out for generating quick responses for lot differentiation; however, studies are needed to improve this practice in safflower seeds. This study aimed to verify the efficiency and establish a methodology of the electrical conductivity test for safflower seeds. Initially, 12 lots were characterized regarding physical properties and physiological potential. For electrical conductivity, the experiment was carried out in three stages with the 12 lots: firstly, the soaking period was determined (2, 4, 6, 8, and 16 hours), then the volume was evaluated (50, 75, 100, 150, and 200 mL), and finally the number of seeds (25 and 50 seeds) was determined. The electrical conductivity test using 25 seeds in 50 mL of distilled water for six hours of soaking enables the differentiation of safflower seed lots. Multivariate analysis of principal components is efficient in discriminating the vigor of safflower seed lots.

Although safflower seed extract exhibits pharmacological activity against various diseases, the effects of its individual compounds on osteoarthritis (OA) have not been elucidated. Here, we evaluated the effects of these extracts and their single compounds on OA. N-(p-Coumaroyl) serotonin and N-feruloyl serotonin, main components of safflower seed extract, were isolated by high-performance liquid chromatography. Under in vitro OA mimic conditions, the expression of the matrix metalloproteinases (MMPs) MMP3/13 and a disintegrin and metalloproteinase with thrombospondin motifs (ADAMTS) ADAMTS5 were reduced in mouse chondrocytes treated with safflower seed extract. Furthermore, the oral administration of safflower seed extract attenuated cartilage destruction in a mouse OA model induced by destabilization of the medial meniscus. N-(p-Coumaroyl) serotonin and N-feruloyl serotonin, but not serotonin, reduced MMP3, MMP13, and ADAMTS5 expression in IL-1β-treated chondrocytes. Additionally, they significantly blocked the nuclear factor-κB (NF-κB) pathway by inhibiting IκB degradation and p65 phosphorylation. Our results suggest that safflower seed extract and its single compounds can attenuate cartilage destruction by suppressing MMP and ADMATS5 expression. The anti-arthritic effects are mediated by NF-κB signaling and involve the inhibition of IκB degradation and p65 phosphorylation. These results indicate that safflower seed extract may serve as a novel therapeutic agent against OA.

Seeds of seven different genotypes of underutilised oil crop Carthamus tinctorius L. (Asteraceae) from alternative oil species collection of the Institute of Field and Vegetable Crops (Novi Sad, Serbia), were analysed for their protein, oil, fatty acids, tocopherols and total phenolics contents, with a view to test their diversity and potential as an alternative source of these valuable compounds. Seeds of the tested safflower genotypes had total protein (determined by Kjeldahl method) content from 11.5 to 16.0%, while total oil content (determined by Soxhlet method) ranged from 16.8 to 24.5% of dry matter, on average. Two main unsaturated fatty acids in safflower seeds, oleic and linoleic acids, represent approximately 90% of the total fatty acid content (determined by gas chromatography). Linoleic acid was the dominant fatty acid in all genotypes (61.2-80.2% of oil), while oleic acid was in a negative correlation with linolenic acid content and ranged from 9.6 to 29.5% of oil. The amount of saturated fatty acids ranged from 5.5 to 6.05% for palmitic, and 2.1 to 3.5% for stearic acid. Safflower seed is a source of a-tocopherol (determined by high performance liquid chromatography method with fluorescence detection) and its amount ranged from 358.8 to 461.8 mg/L of oil. The content of total phenolics (determined spectrophotometrically) ranged from 4.0 to 6.0 mg of gallic acid equivalents/g of dry weight. This comprehensive screening of valuable chemical compounds of safflower seeds shows the importance of this alternative oil seed crop as a good source of important nutrients and bioactive constituents.
