We report 2 novel autosomal translocations in the horse. In Case 1, a breeding stallion with a balanced t(4p;30) had produced normal foals and those with congenital abnormalities. Of his 9 phenotypically normal offspring, 4 had normal karyotypes, 4 had balanced t(4p;30), and 1 carried an unbalanced translocation with tertiary trisomy of 4p. We argue that unbalanced forms of t(4p;30) are more tolerated and result in viable congenital abnormalities, without causing embryonic death like all other known equine autosomal translocations. In Case 2, two stallions produced by somatic cell nuclear transfer from the same donor were karyotyped because of fertility issues. A balanced translocation t(12q;25) was found in one, but not in the other clone. The findings underscore the importance of routine cytogenetic screening of breeding animals and animals produced by assisted reproductive technologies. These cases will contribute to molecular studies of translocation breakpoints and their genetic consequences in the horse.

Obesity has been associated with altered reproductive activity in mares, and may negatively affect fertility. To examine the influence of long-term high-energy (HE) feeding on fertility, Shetland pony mares were fed a diet containing 200% of net energy (NE) requirements during a three-year study. The incidence of hemorrhagic anovulatory follicles (HAF) and annual duration of cyclicity were compared to those in control mares receiving a maintenance diet. Day-7 embryos were flushed and transferred between donor and recipient mares from both groups; the resulting conceptuses were collected 21 days after transfer to assess conceptus development. HE mares became obese, and embryos recovered from HE mares were more likely to succumb to early embryonic death. The period of annual cyclicity was extended in HE compared to control mares in all years. The incidence of HAFs did not consistently differ between HE and control mares. No differences in embryo morphometric parameters were apparent. In conclusion, consuming a HE diet extended the duration of cyclicity, and appeared to increase the likelihood of embryos undergoing early embryonic death following embryo transfer.

Porcine parvovirus (PPV) infection is the primary cause of SMEDI (stillbirth; mummification; embryonic death; infertility) syndrome, which is a global burden for the swine industry. Thus, it is crucial to establish a rapid and efficient detection method against PPV infection. In the present work, we developed a recombinase-aided amplification (RAA) assay, coupled with a lateral flow dipstick (LFD), to achieve an amplification of PPV DNA at 37 °C within 15 min. The detection limits of PPV RAA-LFD assay were 102 copies/μL recombinant plasmid pMD19-T-VP1, 6.38 × 10−7 ng/μL PPV DNA, and 10−1 TCID50/mL virus, respectively. This method was highly specific for PPV detection with no cross-reactivity for other swine pathogens. In contrast to polymerase chain reaction (PCR), the PPV RAA-LFD assay is more sensitive and cost-saving. Hence, the established PPV RAA-LFD assay provided an alternative for PPV detection, especially in resource-limited regions.

The embryo of the sand-dollar (Echinarachnius parma) was exposed to various concentrations of fluorinated pyrimidines immediately after fertilization. FUDR (5-fluorodeoxyuridine) was most active, and a concentration of 2 to 4 mγ/10 cc. (0.8 to 1.6 x 10-6 m.eq./liter) blocked development at the early blastula stage. Larger doses interrupted development at the same stage. This effect was prevented by thymidine (TDR) and thymine (T); and these pyrimidines protected against many times the minimal lethal concentration of FUDR. TDR was active as a protective agent if added just before early blastula formation. The other fluorinated pyrimidines, 5-fluorouracil (FU), 5-fluorouridine (FUR), 5-fluorocytidine (FCR), 5-fluorodeoxycytidine (FCDR), and 5-fluoroorotic acid (FO), were also studied. These drugs produced effects on embryonic development similar to those seen with FUDR. The effective concentrations, however, varied greatly. T and TDR provided protection against these drugs, but in most cases they were not so effective as against FUDR. 5-Bromodeoxyurdine (BrUDR), beginning at the early blastula stage, caused a random pattern of embryonic death up to the pluteus stage. This drug has been shown to be incorporated into bacterial DNA. BrUDR protected embryos against the early lethal effects of FUDR presumably acting as a thymidine substitute, but the embryos died subsequently in a pattern similar to that seen with BrUDR alone. FUDR and BrUDR appear to inhibit the formation and alter the structure of DNA, respectively, distinctive effects whch may provide a means for studying the role of DNA in embryonic development.

ABSTRAK.  Progesteron merupakan hormon yang berperan penting dalam proses pemeliharaan kebuntingan dan dihasilkan oleh corpus luteum. Penelitian ini bertujuan mengetahui hubungan antara kadar hormon progesteron pada fase awal luteal dengan kematian embrio pada sapi Aceh. Dalam penelitian ini digunakan empat ekor sapi betina dewasa berumur 3-5 tahun, bobot badan 150-250 kg, sehat secara klinis, dan memiliki reproduksi normal. Sapi disinkronisasi menggunakan 5 ml prostaglandin F2 alfa (PGF2α) dengan pola penyuntikan ganda berinterval 11 hari. Koleksi sampel darah untuk pengukuran konsentrasi progesteron dilakukan pada hari ke-5, 6, dan 7 pasca inseminasi. Pengukuran konsentrasi progesteron dilakukan menggunakan metode enzymelinked-immunoassay (ELISA), pemeriksaan kebuntingan dan kematian embrio menggunakan metode transrektal ultrasonografi pada hari ke-25 pasca inseminasi. Pemeriksaan diulang setiap 10 hari sampai hari ke-55 pasca inseminasi. Puncak sekresi progesteron pada sapi bunting dengan embrio yang bertahan hidup terdapat pada hari ke-7 (2,082 ng/ml), pada sapi Late Embryonic Mortality (LEM) di hari ke-5 (8,209 ng/ml) dan pada sapi tidak bunting di hari ke-7 (3,051±1,157 ng/ml). Sekresi progesteron sapi LEM pada hari ke-5 sampai dengan ke-7 cenderung menurun sedangkan pada sapi yang bertahan hidup cenderung meningkat.  (Correlation between progesterone levels in early luteal phase and embryonic death  in Aceh cattle) ABSTRACT. Progesterone is an important hormone that functions to maintain pregnancy and is produced by the corpus luteum. The aim of this study was to see a correlation between progesterone and the incidence of embryonic death in Aceh cattle. This study used four adult female cows, 3-5 years old, 150-250 kg body weight, clinically healthy, and have a normal reproduction. The synchronized with 5 ml prostaglandin F2 alfa hormone, and double injection pattern with 11-day intervals. The blood was collected for progesterone measurements on 5th, 6th, 7th day post artificial insemination. Measurement of progesterone concentration was carried out using an enzymelinked-immunoassay (ELISA), while pregnancy and embryo mortality was performed using the trans-rectal ultrasonography method on the 25th day after insemination. The examination was repeated every 10 days until day 55th after insemination. Progesterone secretion peaks in pregnant cows were on day 7th (2.082 ng/ml), in cattle Late Embryonic Mortality (LEM) on day 5th (8.209 ng/ml) and in cattle not pregnant on day 7th (3.051±1.157 ng/ml). The pattern of LEM progesterone secretion on days 5th to 7th tends to decrease while those that survive tend to increase.

Diabetes-induced early embryonic death is accompanied by an increased expression of tumour necrosis factor alpha (TNF-alpha) in the embryonic microenvironment. The aim of the present study was to evaluate whether diabetes-induced embryopathic stress may also alter the expression of TNF-alpha produced by the embryo itself. As a model, whole postimplantation embryos were cultured for 24 h in a medium with high concentrations of glucose, one of the main diabetes-associated teratogenic metabolites. An anomaly such as an open neural tube was used as an end-point characterizing the glucose-induced teratogenic effect and the number of somites was counted to evaluate growth retardation induced by glucose. The expression of TNF-alpha (by immunohistochemistry), apoptosis (by TdT-mediated dUTP nick-end labelling; TUNEL) and the activity of caspases 3 and 8 (by a fluorometric assay) were evaluated in normal and malformed embryos. Ninety-seven per cent of the embryos exposed to 1300 mg glucose dl(-1) exhibited an open neural tube. The percentage of malformed embryos was smaller in media containing 800 and 500 mg glucose dl(-1) (68 and 37%, respectively) but it still exceeded significantly the value registered in embryos developing in a normoglycaemic medium (12%). In addition, a significant decrease in the number of somites was observed in embryos developing in media containing 1300 and 800 mg glucose dl(-1). Malformed embryos exhibited a greater number of nuclei that were positive in the TUNEL assay as well as a higher amount of active caspase 8 compared with normal embryos (with closed neural folds). TNF-alpha expression was detected in the neuroepithelial layer of the neural tube of the malformed embryos, whereas the expression of this cytokine was weak, if detectable, in normal embryos. Together, these findings indicate that TNF-alpha produced by the embryo may be involved in regulating the response of embryos to diabetes-generated embryopathic stress.
