A major application of potentiometric dyes has been the multisite optical recording of electrical activity in excitable systems. After being championed by L.B. Cohen and his colleagues for the past 20 years, the impact of this technology is rapidly being felt and is spreading to an increasing number of neuroscience laboratories. A second class of experiments involves using dyes to image membrane potential distributions in single cells by digital imaging microscopy - a major focus of this lab. These studies usually do not require the temporal resolution of multisite optical recording, being primarily focussed on slow cell biological processes, and therefore can achieve much higher spatial resolution. We have developed 2 methods for quantitative imaging of membrane potential. One method uses dual wavelength imaging of membrane-staining dyes and the other uses quantitative 3D imaging of a fluorescent lipophilic cation; the dyes used in each case were synthesized for this purpose in this laboratory.

ZusammenfassungSeit dem Nachweis boviner spongiformer Enzephalopathie (BSE) unter Rindern in Deutschland im November 2000 ist die Gefahr, an der neuen Variante der Creutzfeld-Jakob-Krankheit (vCJD) zu erkranken, auch hierzulande größer geworden. Diese Übersicht schildert die Differenzialdiagnostik von Demenzerkrankungen mit initialen psychiatrischen Auffälligkeiten im jüngeren und höheren Alter. Besonderer Wert wird auf die Unterschiede zwischen der sporadischen Creutzfeld-Jakob-Krankheit (sCJD) und der vCJD gelegt. Neben den klinischen und initial meist psychiatrisch geprägten Verläufen werden neuere laborchemische, molekulargenetische und neuroradiologische Aspekte dieser beiden Prionkrankheiten dargestellt. Der Liquor ist in den meisten Fällen positiv für Protein 14-3-3, Tau-Protein und Neuron-spezifische Enolase (NSE). Nur bei bestimmten molekulargenetisch am Codon 129 des Prionproteins determinierten Subgruppen der sCJD-Patienten, nicht aber bei vCJD-Patienten, finden sich im EEG periodische scharfe Wellen. In der Frühdiagnostik der sCJD kann vor allem die diffusionsgewichtete MRI eingesetzt werden. Bei den jüngeren vCJD-Patienten findet man neben den psychiatrischen Symptomen Parästhesien, erst später eine Demenz und Ataxie von mehr als 6 Monaten Dauer sowie T2-MRI-Signalhyperintensitäten im Pulvinar. Als weitere Differenzialdiagnosen der verschiedenen CJD-Subtypen wird auch die wenig bekannte Hashimoto-Enzephalopathie näher beschrieben.

AbstractLuminescent quantum dots (QDs) are emerging as a new class of biological labels with unique properties and applications that are not available from traditional organic dyes and fluorescent proteins. Here we report new developments in using semiconductor quantum dots for quantitative imaging and spectroscopy of single cancer cells. We show that both live and fixed cells can be labeled with multicolor QDs, and that single cells can be analyzed by fluorescence imaging and wavelength-resolved spectroscopy. These results raise new possibilities in cancer imaging, molecular profiling, and disease staging.

Dual-energy imaging has emerged as a superior way to recognize materials in X-ray computed tomography. To estimate material properties such as effective atomic number and density, one often generates images in terms of basis functions. This requires decomposition of the dual-energy sinograms into basis sinograms, and subsequently reconstructing the basis images. However, the presence of metal can distort the reconstructed images. In this paper we investigate how photoelectric and Compton basis functions, and synthesized monochromatic basis (SMB) functions behave in the presence of metal and its effect on estimation of effective atomic number and density. Our results indicate that SMB functions, along with edge-preserving total variation regularization, show promise for improved material estimation in the presence of metal. The results are demonstrated using both simulated data as well as data collected from a dualenergy medical CT scanner.

Abstract In this paper the use of a set of nonlinear edge-preserving filters is proposed as a pre-processing stage with the purpose to improve the quality of hyperspectral images before object detection. The capability of each nonlinear filter to improve images, corrupted by spatially and spectrally correlated Gaussian noise, is evaluated in terms of the average Improvement factor in the Peak Signal to Noise Ratio (IPSNR), estimated at the filter output. The simulation results demonstrate that this pre-processing procedure is efficient only in case the spatial and spectral correlation coefficients of noise do not exceed the value of 0.6
