Doxorubicin (DOX) is one of the most potent and widely used chemotherapeutic agents to treat several malignancies. However, the clinical use of DOX is seriously restricted due to its acute and chronic cardiotoxic side effects This study investigated the protective effect of (Ajwa) date aqueous extract (AJDAE) against doxorubicin-induced cardiotoxicity in rats. Sixty Wister albino male rats (150-200 gms.) were comprised in our study and divided into six equal groups: group I (untreated control), group II, group III, rats were orally received AJDAE (0.75 & 1.5 gm/ kg.bw) respectively, for 4 weeks, rats of groups IV, V and VI were intraperitoneally injected with one dose of doxorubicin (5 mg/kg.bw) at the end of the 4th week of the study to induce cardiotoxicity, rats of groups V & VI were orally received AJDAE (0.75 & 1.5 gm/ kg.bw) respectively. Cardiac enzymes, lipid profile, SOD, GR, GST, GPx, CAT and MDA in rats’ hearts homogenate, urinary 8OHdG as well as DNA integrity and histopathological changes were investigated in all studied rats.Oral administration of AJDAE (0.75 & 1.5 gm/ kg.bw) attenuated the cardiotoxicity of DOX, improved the cardiac enzymes, lipid profile, reduced the urinary 8OHdG and prohibited the depletion of endogenous antioxidants and suppressed lipid peroxidation (MDA). Moreover, AJDAE enhanced DNA integrity. Histological findings showed that AJDAE (0.75 & 1.5 gm/ kg.bw) administration reduced cardiomyocytes alterations, congestion, edema and the intense cellular stress exerted on myocardial fibers as well as restored the cardiomyocytes architecture. Our data showed that AJDAE obviously resulted in protective effects against DOX-induced cardiotoxicity in rat’s heart. It can be concluded that Ajwa date offers a considerable protection against DOX-induced cardiotoxicity.

Thyroid hormone stimulates the Na(+)-PO4 symporter but not the Na(+)-SO4 symporter in renal brush border

In our previous studies we established that thyroid hormones [L-thyroxine (T4) or 3,5,3'-triiodothyronine (T3)] elicit an increase in Na(+)-Pi symport in rat and mouse renal brush-border membrane (BBM) vesicles (BBMV), but the Na(+)-coupled symports of other solutes were not influenced. However, a recent report [H. S. Tenenhouse, J. Lee, and N. Harvey. Am. J. Physiol. 261 (Renal Fluid Electrolyte Physiol. 30): F420-F426, 1991]claimed that T3 increases to a similar degree both Na(+)-Pi symport and Na(+)-SO4 symport in murine renal BBM. Adult male rats were fed either normal (0.7% Pi; NPD) or high-phosphate (1.4% Pi; HPD) diet and received T3 (0.2 mg/100 g body wt ip) for 3 days before the kidneys were removed, BBMV were prepared, and the transport rates were determined. Although the Na(+)-Pi symport significantly increased (delta = +35%) in both NPD and HPD rats treated with T3, the Na(+)-35SO4 symport and Na(+)-D-[3H]glucose symports were not influenced by T3. Furthermore, treatment of NPD-fed mice with T3 using a similar protocol as rats resulted in a significant increase (delta = +26%) of Na(+)-(Pi)4 symport, but did not alter Na(+)-SO4 symport or Na(+)-glucose symport. Our findings thus document that T3 regulates selectively the Na(+)-Pi symporter in BBM without having any effect on other major divalent anions, such as SO4.

Introduction The Eurycoma longifolia (EL) root aqueous extract has long been used as an enhancer of male sexual performance. However, data from previous studies in both human males and experimental male animals on the testosterone level in those given the EL extract orally were at best insufficiently conclusive. Materials and Method Sixty-four healthy adult male Sprague Dawley rats were acclimatized, and randomized into six test groups and one control group. All rats where given either the aqueous EL extract or distilled water via metal gavage needle. The first three test groups received the low (50mg/kg bw), medium (100mg/ kg bw) and high (200mg/kg bw) doses respectively of the EL daily for 15 days only. The second three test groups continued receiving the same daily treatment doses for 30 days. The controls were given distilled water only. At the end of each of the study period, blood samples were collected via cardiac puncture and the rats were euthanized. The testicles were obtained, weighed, and processed for histological examination. Results The sera testosterone levels were higher in animals which received the medium and high doses for both treatment duration. Rats which received medium and high oral doses of EL showed an increase of spermatogenesis and mature spermatozoa. Conclusion The optimal enhancing effect on sera testosterone levels and testicular spermatogenesis of EL treatment in adult male rats was observed with the medium dose of 100mg/kg bw given once daily for both 15 and 30 days.
