Introduction The Eurycoma longifolia (EL) root aqueous extract has long been used as an enhancer of male sexual performance. However, data from previous studies in both human males and experimental male animals on the testosterone level in those given the EL extract orally were at best insufficiently conclusive. Materials and Method Sixty-four healthy adult male Sprague Dawley rats were acclimatized, and randomized into six test groups and one control group. All rats where given either the aqueous EL extract or distilled water via metal gavage needle. The first three test groups received the low (50mg/kg bw), medium (100mg/ kg bw) and high (200mg/kg bw) doses respectively of the EL daily for 15 days only. The second three test groups continued receiving the same daily treatment doses for 30 days. The controls were given distilled water only. At the end of each of the study period, blood samples were collected via cardiac puncture and the rats were euthanized. The testicles were obtained, weighed, and processed for histological examination. Results The sera testosterone levels were higher in animals which received the medium and high doses for both treatment duration. Rats which received medium and high oral doses of EL showed an increase of spermatogenesis and mature spermatozoa. Conclusion The optimal enhancing effect on sera testosterone levels and testicular spermatogenesis of EL treatment in adult male rats was observed with the medium dose of 100mg/kg bw given once daily for both 15 and 30 days.

Aim: This study aimed to evaluate the effect of shell supplementation on the regulation of male reproduction in rats. Materials and Methods: The zinc (Zn) level of shell from blood clam (Anadara granosa), green mussel (Perna viridis), and conch shell (Telescopium telescopium) was analyzed. The highest Zn content shell was fed to male Sprague Dawley rats for 0, 9, 30, and 50 days at the dose of either 0.09 mg/200 g BW or 0.18 mg/200 g BW. To determine the testosterone levels, blood was collected through the infraorbitalis sinus just before the rat was sacrificed. Testicular and brain were also collected for Cyp19 aromatase receptor analysis. Results: The Zn level in the shell of blood clam, green mussel, and conch shell 61.55 mg/kg, 2.78 mg/kg, and 3.93 mg/kg, respectively. The testosterone level of T1 group receiving 0.18 mg/200 g BW for 0, 9, 30, and 50 days was 1.42±0.59, 2.15±1.58, 2.98±2.53, and 8.11±2.03 ng/mL, respectively. The testosterone level of T2 group receiving 0.09 mg/200 g BW for 0, 9, 30, and 50 days was 2.50±0.32, 1.25±0.60, 3.87±3.27, and 3.54±0.23 ng/mL, respectively. The T3 group receiving Na-CMC showed the level of testosterone at days 0, 9, 30, and 50 days was 0.77±0.22, 1.99±1.65, 4.12±0.07, and 2.19±1.30 ng/mL, respectively. Finally, the T4 group receiving Zn showed testosterone levels at days 0, 9, 30, and 50 days was 0.51±0.58, 2.24±3.16, 4.58±1.97, and 2.89±0.20 ng/mL, respectively. There was a significant difference (p<0.05) between the T1 group compared to the other groups. However, the absence of expression of Cyp19 aromatase both in Leydig cells and the brain indicated no conversion of testosterone to estradiol. To add, this finding showed the potential use of the shell to boost the testosterone level in male rats. Conclusion: Shell acted as an aromatase blocker to boost the testosterone level in male rats. This also indicates its promising application in birds to manipulate the quality of song and feather.

Abstract: Bisphenol A (BPA) has the structural and physiological activity of estrogen and acts as an endocrine disruptor. This study was aimed to prove that oral BPA could decrease testosterone levels in male Sprague Dawley rats. This was an experimental study with a randomized pretest-posttest control group design using 14 rats (Rattus norvegicus), male, Sprague Dawley strain, 2-3 months old, as subjects. Rats were divided into two groups with 7 rats each. The control group (P0) was treated with placebo for 14 days and the treatment group (P1) was treated with oral bisphenol A (BPA) single dose of 400mg/kg/day for 14 days (P1). At before and after 14-day treatment, blood was drawn through the canthus medial of orbital sinus to obtain the pretest and posttest data. Testosterone levels were examined by using ELISA. The results showed that before treatment (pretest), the mean testosterone level in P0 group was 8.29±4.68 nmol/L, while in P1 group was 7.39±1.45 nmol/L (P > 0.05). After treatment for 14 days, the mean testosterone level in P0 group was 7.40±1.37 nmol/L, while in P1 group was 3.36±1.26 nmol/L (P < 0.01). The statistical analysis showed that there was no difference in testosterone levels in P0 group (P > 0.05), meanwhile there was a very significant decrease of testosterone levels in P1 group (P < 0.01) after the treatment of BPA 400mg/kg/day for 14 days. Conclusion: Oral bisphenol A (BPA) could decrease testosterone levels in male rats (Rattus norvegicus) of Sprague Dawley strain.Keywords: bisphenol A (BPA), testosteroneAbstrak: Bisphenol A (BPA) memiliki aktifitas hormon estrogen sehingga jika masuk ke dalam tubuh dapat menjadi endocrine disruptor. Jenis penelitian ialah eksperimental dengan randomized pretest-postest control group design yang menggunakan 14 ekor tikus (Rattus norvegicus) jantan, galur Sprague Dawley, berumur 2-3 bulan, yang terbagi menjadi 2 (dua) kelompok masing-masing berjumlah 7 ekor tikus. Kelompok kontrol (P0) diberikan plasebo 14 hari dan kelompok perlakuan (P1) diberi BPA dosis tunggal 400mg/kgBB/hari selama 14 hari. Saat sebelum dan sesudah perlakuan selama 14 hari, darah diambil melalui chantus medial orbitalis untuk pemeriksaan pretest dan postest dan kadar testosteron diperiksa dengan metode ELISA. Hasil penelitian menunjukkan bahwa sebelum perlakuan (pretest), rerata kadar testosteron kelompok P0 8,29±4,68 nmol/L, sedangkan pada kelompok P1 7,39±1,45 nmol/L (P > 0,05). Setelah perlakuan selama 14 hari, rerata kadar testosteron kelompok P0 ialah 7,40±1,37 nmol/L dan pada kelompok P1 3,36±1,26 nmol/L (P < 0,01). Analisis efek perlakuan menunjukkan tidak terjadi penurunan kadar hormon testosteron pada kelompok P0 (P > 0,05), sedangkan penurunan bermakna terdapat pada kelompok P1 (P < 0,01). Simpulan: Pemberian bisphenol A (BPA) oral dapat menurunkan kadar testosteron pada tikus (Rattus norvegicus) jantan galur Sprague Dawley.Kata kunci: bisphenol A (BPA), testosteron

Objective: To observe the difference of testosterone levels in adult male Sprague Dawley rats treated with combination of cisplatin and vitamin E compared to those treated with cisplatin only. Material & Methods: We used 24 adult male Sprague Dawley rats weight 200–300 grams and randomly assigned into 4 groups (n=6). Rats in negative control group (NC) were given intraperitoneal normal saline injection, while the positive control (PC) group were injected with cisplatin 5 mg/kgBW at the end of the 3rd week. Two other groups, P1 and P2, were injected with cisplatin 5 mg/kgBW and given vitamin E orally 50 mg/kgBW and 200 mg/kgBW, respectively. Cardiac blood was aspirated at the end of the 7th week and processed for analysis of testosterone levels. Results: We recorded a significantly lower testosterone levels in rats treated only with cisplatin 5 mg/kgBW (CP) compared to those in CN group (p=0.006), and those receiving combination of cisplatin and vitamin E 50 mg/kgBW (p=0.003) and 200 mg/kgBW (p=0.001). Though not significant, testosterone levels were higher in P2 group than in P1 group (p=0.702). Conclusion: Exposure to cisplatin can lower testosterone levels in white rats, and the administration of vitamin E gives protection against such effect.

Spinacia oleracea “Spinach” is a plant rich in antioxidants; it leads to improved male reproduction. For this reason, the objective of this study is to evaluate the effect of the aqueous extract of this plant on the fertility of adult male rabbits after their administration with ethanol. The rabbits were divided into four groups: control group, group treated with 150 mg / kg / day of aqueous Spinacia oleracea extract (Ep), group treated with 40% ethanol (EtOH), and group treated with 150 mg / kg / day of aqueous Spinacia oleracea extract + 40% ethanol (Ep + EtOH), for a period of five successive weeks. Treatment was administered orally. After sacrificing the rabbits, the reproductive organs were removed and weighed. Certain sperm parameters (concentration, mobility, speed, and vitality), the testosterone level and the weight of the testes and epididymis were assessed. The results obtained showed a decrease in the concentration, mobility, speed and vitality of sperm, accompanied by a decrease in testosterone levels, and in the weight of the testes and epididymis in the group treated with ethanol, on the other hand, treatment with aqueous Spinacia oleracea extract revealed an increase in these parameters. In conclusion, treatment with aqueous Spinacia oleracea extract can repair the damage caused by ethanol.

AbstractObjectivesMethotrexate (MTX) is an anticancer drug used in chemotherapy. MTX was known for its toxic effects involving most of the organs including testis. Bee pollen is healthy food for human and has antioxidant effect. We intended to determine protective effect of bee pollen against testicular injury caused by MTX in rats.MethodsThirty-two adult Sprague Dawley male rats were used, and 4 groups were formed: control, MTX, pollen, and MTX + pollen. Rats were given pollen at a dose of 400 mg/kg with intragastric gavage for 10 days. On day 7, MTX was administered a single dose of 30 mg/kg ip. Serum testosterone and LH, tissue MDA level, and SOD and CAT enzyme activities were examined. In addition, spermatological parameters were evaluated.ResultsMDA level and SOD activity increased while testosterone level decreased significantly in the MTX group compared to the control group. In the MTX + pollen group, MDA level and SOD activity decreased while testosterone level increased. There was no significant change in CAT activity and LH values. Abnormal sperm ratio decreased in the MTX + pollen group compared to the MTX group.ConclusionsOur results suggest that bee pollen has a healing effect on reproductive parameters in testicular damage caused by MTX.

The present study investigated the long-term effects of prenatal betamethasone exposure on sperm quality and count, fertility and plasma testosterone levels in adult male rats. Pregnant rats received 0.1 mg kg–1 betamethasone on Days 12, 13, 18 and 19 of pregnancy. This treatment impaired sperm quality, sperm production, fertility and plasma testosterone levels in adult male offspring compared to the control group. Thus, the results of the present study indicate that the long-term effects of prenatal betamethasone exposure may be deleterious to offspring. The consequent decrease in testosterone production during adulthood, in association with damaged semen parameters, may explain for the observed decrease in the capacity of adult male offspring to themselves generate viable descendants.

<p><strong>Objective:</strong><strong>  </strong>To determine the toxic effects of quinolones on serum testosterone level in male albino rats.</p><p><strong>Methods:</strong><strong>  </strong>Eighty male albino rats were randomly divided into A, B, C and D groups each group having 20 albino rats. These groups were further subdivided into A1, A2, B1, B2, C1, C2, D1 and D2 having 10 albino rats each. Ciprofloxcin, ofloxacin and enoxacin dissolved in distilled water were given at 135mg/kg/ day, 72mg/kg/day and 12.5mg/kg/day to groups A, B and C respectively for 12 weeks. Only distilled water was given to group D which was control group for the same time period. Blood samples were drawn for testosterone hormone level estimation at 0, 14<sup>th</sup>, 28<sup>th</sup> and 42<sup>nd</sup> day in subgroups Al, B1, Cl and D1 and then the animals in said groups were sacrificed on 42<sup>nd</sup> day to identify testicular morphological changes. Rats in subgroup A2, B2, C2 and D2 were kept alive till 84<sup>th</sup> day after stopping drugs at 42 days to find out if there is any change in levels of testosterone after discontinuation of the treatment. Blood samples for testosterone hormone estimation were taken at 0, 14<sup>th</sup>, 28<sup>th</sup>, 42<sup>nd, </sup>56<sup>th</sup>, 70<sup>th</sup> and 84<sup>th</sup> day from subgroups A2, B2, C2 and D2. Testosterone hormone estimation assay was performed on the collected samples.</p><p><strong>Results:</strong><strong>  </strong>Testosterone assay showed significant decrease in serum testosterone levels in all experimental groups, when compared with control group. These levels did not return back to normal even after withdrawal of drugs. This study suggests a gonadotoxic potential of quinolones.</p><p><strong>Conclusion:  </strong>Quinolones reduce serum testosterone levels and should be used carefully for long term therapy.<strong></strong></p>

ABSTRACT The study aimed to find out the effect of Eurycoma longifolia Jack on testosterone level and spermatozoa quality of Etawa crossbreed goat. Goats were divided into two treatment groups, three goats each. Group 1 (K1) was control group which given orally 20 ml of distilled water every morning at 9:00 am for six days, and group 2 (K2) was administered with Eurycoma longifolia with the dose of 90 mg/kg bw in 20 ml of distilled water orally every morning at 9:00 am for six days. The concentration of testosterone was measured on day 1, 3, and 6 using the method of enzyme-linked immunosorbent assay (ELISA). The quality of sperm consisted of concentration and percentage of life sperm. The results showed that level of testosterone on K2 increased started from day 1 (9.36 ng/ml) to day 6 (12.43 ng/ml) (P<0.05). The percentage of life spermatozoa increased higher on K2 compare to K1 on day 3 to day 6, that was 88.6 and 89.8% on K1 to 91.0 and 92.7% on K2. In conclusion, the administration of pasak bumi with the dose of 90 mg/kg bw in 20 ml of distilled water for 6 days able to increase testosterone levels and percentage of life sperm of Etawa crossbreed goat.

Abstract Roflumilast (ROF) (3-cyclo-propylmethoxy-4-difuorome-thoxy-N-[3,5-di-chloropyrid-4-yl] benzamide) is a second generation and forcible phosphodiesterase-4 (PDE4) inhibitor. This study aims to investigate the effects of chronic Roflumilast in different doses on testicular tissue and testosterone levels in healthy Sprague-Dawley rats. During the research, the 6 weeks old (180–200 gr), 36 male rats were divided into 4 groups. Roflumilast (ROF) was administered as 0.5 mg/kg and 1 mg/kg by oral gavage for four weeks, once each day. Hematoxylin-Eosin (H&E) staining for histopathological examinations in testicular tissue, immunohistochemical and immunofluorescence examinations for Caspase-3, Apoptosis Inducing Factor (AIF) and Light Chain 3β (LC3B) expression levels, and ELISA method used to determine serum testosterone levels. Data were analyzed using SPSS v.22 with Kruskal-Wallis, Mann Whitney-U, and Wilcoxon tests. Roflumilast group lost weight compared to the control and sham. Shedding in the seminiferous epithelium, degenerations in the interstitial area, separation between cells, desquamation, interstitial edema and degenerative changes in the testicular tissue was observed. While apoptosis and autophagy determined by Caspase-3, AIF and LC3B were close and statistically insignificant in control and sham, there was significantly increased apoptotic and autophagic changes and immunopositivity in the ROF groups. The 1 mg/kg Roflumilast group’s serum testosterone level was lower than control, sham and 0.5 mg/kg Roflumilast groups. When the research data was evaluated, it was determined that the chronic use of the active ingredient Roflumilast, which has a broad-spectrum area such as COPD, arthritis, neurodegenerative diseases, liver and dermatology, had negative effects on the testicular tissue and testosterone level of rats.
