Purification, in vitro antioxidant and in vivo anti-aging activities of soluble polysaccharides by enzyme-assisted extraction from Agaricus bisporus

The aim of this work was designed to investigate the antioxidant, antiaging, and hepatoprotective effects of alkali-extractable polysaccharides (AlAPS) and their three purified fractions (AlAPS-1, AlAPS-2, and AlAPS-3) from Agaricus bisporus in D-galactose induced aging mice. For in vitro antioxidant analysis, both AlAPS and its fractions exhibited moderate reducing power, Fe2+-chelating activities, and potent scavenging activities on hydroxyl and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals. The in vivo results demonstrated that the polysaccharides, especially AlAPS-2, showed potential antiaging and hepatoprotective effects by enhancing the antioxidant status, decreasing serum hepatic enzyme activities, and improving the lipid metabolism. This study suggested that the polysaccharides extracted and purified from A. bisporus could be exploited as a potent dietary supplement to attenuate aging and prevent age-related diseases.

The study was undertaken to determine antifungal potentials of some plant extracts against dry bubble (Verticillium fungicola) disease of white button mushroom (Agaricus bisporus). Twelve botanicals namely, Allium cepa, A. sativum, Saraca asoca, Aloe vera, Azadirachta indica, Lantana camara, Ocimum sanctum, Solanum lycopersicum (Lycopersicon esculentum), Tagetes erecta, Psidium guajava, Catharanthus roseus and Aparagus racemosus were evaluated in-vitro and in-vivo for their efficacy against both A. bisporus and V. fungicola, causing dry bubble disease of mushroom. The efficacy of botanicals was examined by poison food technique in in-vitro. The percent inhibition produced by botanicals against V. Fungicola recorded in-vitro was; A. cepa (25.87%), A. sativum (24.70%), S. asoca (12.35%), A. vera (22.35%), A. indica (35.11%), L. camara (28.48%), O. sanctum (20.59%), S. lycopersicum (20.34%), T. erecta (14.11%), P. guajava (15.11%), C. roseus (18.11%) and A. racemosus (13.52%). Among these plant extracts, A. indica was found best treatment followed by L. Camara and A. Cepa. Plant extracts showing maximum efficacy against V. fungicola and minimum inhibition against mushroom were further evaluated against V. fungicola infection in mushroom crop room (in-vivo test). In in-vivo test, the polybags which receive A. indica show maximum mean increase in yield (43.46%) over control and exhibited minimum mean disease incidence (27.7%).

The orthogonal experimental design was used to optimise Aspergillus niger enzyme-assisted extraction for flavonoids-enriched extract from Dicranopteris. The results indicate that the highest extraction ratio of flavonoids by Aspergillus niger enzyme-assisted extraction could arrive 13.67% using 30ml Aspergillus niger enzyme solution at 70°C,pH5.0 for 2h extraction, which was more efficient than that by without enzymes heating extraction.The flavonoids extract exhibited quite excellent inhibitions of hydroxyl radical. Meanwhile, it was found that the bioactivities appeared to be dose-dependent of flavonoids.

Artichoke by-products are rich in phenolic compounds although they represent a waste for the food industry. This paper examines the application of ultrasound-assisted extraction (UAE) for obtaining organic solvent-free extracts rich in nutraceuticals from artichoke scraps. Application of ultrasounds for 60 minutes on test samples, using water as a solvent, improved recovery of phenolic substances compared with untreated samples. Among the phenols detected by high performance liquid chromatography, 5- O-caffeoylquinic and 1,5-di- O-caffeoylquinic acids were identified. In vivo treatments of tobacco BY-2 cells with ultrasonic extracts consistently enhanced their antioxidant power, making the cells more resistant to heat stress. UAE applied to artichoke by-products, using water as a solvent, appears to be a powerful eco-friendly technique that can provide extracts rich in nutraceuticals and turn waste products into resources. The extracts could be advantageously utilized in the food industry to produce functional foods.
