Methotrexate (MTX) is an anchor drug for the treatment of rheumatoid arthritis (RA); however, long-term and high-dose usage of MTX for patients can cause many side effects and toxic reactions. To address these difficulties, selectively delivering MTX to the inflammatory site of a joint is promising in the treatment of RA. In this study, we prepared MTX-PEI@HA nanoparticles (NPs), composed of hyaluronic acid (HA) as the hydrophilic negative electrical shell, and MTX-linked branched polyethyleneimine (MTX-PEI) NPs as the core. MTX-PEI@HA NPs were prepared in the water phase by a one-pot method. The polymeric NPs were selectively internalized via CD44 receptor-mediated endocytosis in the activated macrophages. In the in vivo mice mode study, treatment with MTX-PEI@HA NPs mitigated inflammatory arthritis with notable safety at a high dose of MTX. We highlight the distinct advantages of aqueous-synthesized NPs coated with HA for arthritis-selective targeted delivery, thus verifying MTX-PEI@HA NPs as a promising MTX-based nanoplatform for treatment of RA.

Extremely small-sized superparamagnetic magnetite nanoparticles of 3Cit). The resulting Cit-coated magnetite nanoparticles exhibited long-term colloidal stability in aqueous media without any surface modification. Regarding the magnetic properties, the nanoparticles were superparamagnetic at room temperature, and might be the potential candidate for MRI contrast agents.

Since the number of bicycles is critical to the sustainable development of dockless PBS, this research practiced the introduction of a machine learning approach to quantity management using OFO bike operation data in Shenzhen. First, two clustering algorithms were used to identify the bicycle gathering area, and the available bike number and coefficient of available bike number variation were analyzed in each bicycle gathering area’s type. Second, five classification algorithms were compared in the accuracy of distinguishing the type of bicycle gathering areas using 25 impact factors. Finally, the application of the knowledge gained from the existing dockless bicycle operation data to guide the number planning and management of public bicycles was explored. We found the following. (1) There were 492 OFO bicycle gathering areas that can be divided into four types: high inefficient, normal inefficient, high efficient, and normal efficient. The high inefficient and normal inefficient areas gathered about 110,000 bicycles with low usage. (2) More types of bicycle gathering area will affect the accuracy of the classification algorithm. The random forest classification had the best performance in identifying bicycle gathering area types in five classification algorithms with an accuracy of more than 75%. (3) There were obvious differences in the characteristics of 25 impact factors in four types of bicycle gathering areas. It is feasible to use these factors to predict area type to optimize the number of available bicycles, reduce operating costs, and improve utilization efficiency. This work helps operators and government understand the characteristics of dockless PBS and contributes to promoting long-term sustainable development of the system through a machine learning approach.

Antimicrobial agents containing symmetrical triiodides complexes with halogen bonding may release free iodine molecules in a controlled manner. This happens due to interactions with the plasma membrane of microorganisms which lead to changes in the structure of the triiodide anion. To verify this hypothesis, the triiodide complex [Na(12-crown-4)2]I3 was prepared by an optimized one-pot synthesis and tested against 18 clinical isolates, 10 reference strains of pathogens and five antibiotics. The antimicrobial activities of this symmetrical triiodide complex were determined by zone of inhibition plate studies through disc- and agar-well-diffusion methods. The triiodide complex proved to be a broad spectrum microbicidal agent. The biological activities were related to the calculated partition coefficient (octanol/water). The microstructural analysis of SEM and EDS undermined the purity of the triiodide complex. The anionic structure consists of isolated, symmetrical triiodide anions [I-I-I]- with halogen bonding. Computational methods were used to calculate the energy required to release iodine from [I-I-I]- and [I-I···I]-. The halogen bonding in the triiodide ion reduces the antibacterial activities in comparison to the inhibitory actions of pure iodine but increases the long term stability of [Na(12-crown-4)2]I3.
