The biosorption performance of both batch and liquid-solid fluidized bed operations of dead fungal biomass type (Agaricusbisporus ) for removal of methylene blue from aqueous solution was investigated. In batch system, the adsorption capacity and removal efficiency of dead fungal biomass were evaluated. In fluidized bed system, the experiments were conducted to study the effects of important parameters such as particle size (701-1400�m), initial dye concentration(10-100 mg/L), bed depth (5-15 cm) and solution flow rate (5-20 ml/min) on breakthrough curves. In batch method, the experimental data was modeled using several models (Langmuir,Freundlich, Temkin and Dubinin-Radushkviechmodels) to study equilibrium isotherms, the experimental data followed Langmuir model and the results showed that the maximum adsorption capacity obtained was (28.90, 24.15, 21.23 mg/g) at mean particle size (0.786, 0.935, 1.280 mm) respectively. In Fluidized-bed method, the results show that the total ion uptake and the overall capacity will be decreased with increasing flow rate and increased with increasing initial concentrations, bed depth and decreasing particle size.

AbstractAntifungal activity of Monothecabuxifolia methanolic extract and its various fractions were assessed against Macrophominaphaseolina, a soil-borne fungal pathogen of more than 500 vegetal species as well as rare and emerging opportunistic human pathogen. Different concentrations of methanolic extract (3.125 to 200 mg mL−1) inhibited fungal biomass by 39–45%. Isolated n-hexane, chloroform and ethyl acetate fractions suppressed fungal biomass by 32–52%, 29–50% and 29–35%, respectively. Triterpenes lupeol and lupeol acetate (1, 2) were isolated from n-hexane while betulin, β-sitosterol, β-amyrin, oleanolic acid (3–6) were isolated from chloroform fraction. Vanillic acid, protocatechuic acid, kaempferol and quercetin (7–10) were isolated from the ethyl acetate fraction and identified using various spectroscopic techniques namely mass spectroscopy and NMR. Antifungal activity of different concentrations (0.0312 to 2 mg mL−1) of the isolated compounds was evaluated and compared with the activity of a broad spectrum fungicide mancozeb. Different concentrations of mencozeb reduced fungal biomass by 83–85%. Among the isolated compounds lupeol acetate (2) was found the highest antifungal against M.phaseolina followed by betulin (3), vanillic acid (7), protocatechuic acid (8), β-amyrin (5) and oleanolic acid (6) resulting in 79–81%, 77–79%, 74–79%, 67–72%, 68–71% and 68–71%, respectively. Rest of the compounds also showed considerable antifungal activity and reduced M.phaseolina biomass by 41–64%.

AbstractShort chain chitooligosaccharides (COs) are chitin derivative molecules involved in plant-fungus signaling during arbuscular mycorrhizal (AM) interactions. In host plants, COs activate a symbiotic signalling pathway that regulates AM-related gene expression. Furthermore, exogenous CO application was shown to promote AM establishment, with a major interest for agricultural applications of AM fungi as biofertilizers. Currently, the main source of commercial COs is from the shrimp processing industry, but purification costs and environmental concerns limit the convenience of this approach. In an attempt to find a low cost and low impact alternative, this work aimed to isolate, characterize and test the bioactivity of COs from selected strains of phylogenetically distant filamentous fungi: Pleurotus ostreatus, Cunninghamella bertholletiae and Trichoderma viride. Our optimized protocol successfully isolated short chain COs from lyophilized fungal biomass. Fungal COs were more acetylated and displayed a higher biological activity compared to shrimp-derived COs, a feature that—alongside low production costs—opens promising perspectives for the large scale use of COs in agriculture.

Abstract Aim Wildfire is a natural pulsed disturbance in landscapes of the Savannah Biome. This study evaluates short-term post-fire effects on leaf litter breakdown, the invertebrate community and fungal biomass of litter from three different vegetal species in a tropical stream. Methods Senescent leaves of Inga laurina, Protium spruceanum and Rircheria grandis (2 ± 0.1 g dry mass) were individually placed in litter bags (30 × 30 cm: 10 mm coarse mesh and 0.5 mm fine mesh) and submerged in the study stream before and after fire. Replicate bags (n = 4; individually for each species, sampling time, fire event and mesh size) were then retrieved after 20 and 40 days and washed to separate the invertebrates before fire event and again immediately after fire. Disks were cut from leaves to determine ash-free dry mass, while the remaining material was oven-dried to determine dry mass. Results The pre-fire mean decomposition coefficient (k = -0.012 day-1) was intermediate compared to that reported for other savannah streams, but post-fire it was lower (k = -0.007 day-1), due to decreased allochthonous litter input and increased autochthones production. Intermediate k values for all qualities of litter post-fire may indicate that fire is equalizing litter quality in the stream ecosystem. The abundance of scrapers was found to be more important than fungal biomass or shredder abundance, probably due to their functioning in leaf fragmentation while consuming periphyton growing on leaf litter. Conclusions Theses results indicate that fire can modify the relationships within decomposer communities in tropical stream ecosystems.

AbstractChromatographic separation of extracts from the fungal biomass of a plant pathogenic fungus, Myrothecium roridum, yielded 8 trichothecene toxins including 6 type D trichothecenes (1–6) and 2 type A trichothecenes (7–8). 6′,12′-Epoxymyrotoxin A (1) and 7′-hydroxymytoxin B (2) were new macrocyclic trichothecenes, while the other trichothecenes were identified as myrotoxin B (3), myrotoxin D hydrate (4), 2′,3′-epoxymyrothecine A (5), miotoxin A (6), and 2 trichothecenes lacking the macrocyclic lactone system, roridin L-2 (7) and trichoverritone (8). The structures of these mycotoxins were characterized using spectroscopic methods. The absolute configurations of 1 and 2 were determined by NOESY and a comparison of their experimental and calculated ECD spectra. Most of these mycotoxins (1–4 and 6) exhibited highly potent antimalarial activity against Plasmodium falciparum. They also showed strong cytotoxicity towards KB and NCI-H187 cell lines (IC50 0.60 – 112.28 nM), as well as the Vero cell line (IC50 1.50 – 46.51 nM).

To develop efficient control measures against fungal plant pathogens, the dynamics of host plant colonization during disease development and the interactions among fungi within host plant tissues need to be clarified. These studies require accurate quantitative estimation of specific fungal biomass in plant tissues. This has been approached by direct-microscopic methods, cultural methods, chemical determinations of fungal components, serological methods, and molecular methods. Among these methods, serological and molecular methods provide rapid, specific, and sensitive quantitative measures of fungal biomass in host plant tissues. Therefore, studies on fungal dynamics of host plant colonization using these two methods are presented. Some examples of species interactions among pathogenic fungi within host plants, such as synergism and competition, are reviewed and the usefulness of serological and molecular methods for studies on these interactions is presented. These quantitative methods will provide helpful information for understanding the ecology of plant pathogenic fungi, such as the dynamics of host plant colonization and species interactions. Abstract Malic acid production from the biodiesel coproduct crude glycerol by Aspergillus niger ATCC 9142, ATCC 10577 and ATCC 12846 was observed to occur with the highest malic acid level acid being produced by A. niger ATCC 12846. Fungal biomass production from crude glycerol was similar, but ATCC 10577 produced the highest biomass. Fungal biotransformation of crude glycerol into the commercially valuable organic acid malic acid appeared feasible.

Clearcutting Newfoundland boreal forests significantly reduced organic layer fungal and total microbial biomass in clearcut areas with and without slash cover, compared with forested plots. However, aerobically incubated respiration rates were highest in organic layers from clearcut areas under slash, intermediate under forests, and lowest from clearcut areas without slash.
