Simultaneous bacterial inactivation and microcontaminant removal by solar photo-Fenton mediated by Fe3+-NTA in WWTP secondary effluents

Fe (VI) was evaluated to treat municipal secondary effluents derived from wastewater treatment plants. Organic matter and reactive phosphate compounds in wastewater was oxidized by Fe (VI) l, and the formation of adsorbable organic haloids (AOX) as by-products in this study. Additionally, the most probable number (MPN) of cultivable heterotrophic bacteria was also determined. In every case for 100% bacterial inactivation a low Fe (VI) concentration (5 mg L.1) was enough and by this concentration 40% chemical oxygen demand (COD) and 20% total organic carbon (TOC) removal could be obtained on average. Higher Fe (VI) concentration (715 mg L.1) resulted in an even higher organics reduction (depending on the water matrix up to 79% COD and 45% TOC removal). Monitoring AOX concentration during Fe (VI)-treatment, it could be stated that AOX formation was in linear relationship with the applied Fe (VI) concentration, but at higher than about 10 mg L-1Fe (VI) concentration this increase became much less than at lower Fe (VI) concentration.

Indoor surface contamination by microbes is a major public health concern. A damp environment is one potential sources for microbe proliferation. Smart photocatalytic coatings on building surfaces using semiconductors like titania (TiO<sub>2</sub>) can effectively curb this growing threat.<b> </b>Metal-doped titania in anatase phase has been proved as a promising candidate for energy and environmental applications. In this present work, the antimicrobial efficacy of copper (Cu) doped TiO<sub>2 </sub>(Cu-TiO<sub>2</sub>) was evaluated against <i>Escherichia coli</i> (Gram-negative) and <i>Staphylococcus aureus</i> (Gram-positive) under visible light irradiation. Doping of a minute fraction of Cu (0.5 mol %) in TiO<sub>2 </sub>was carried out <i>via</i> sol-gel technique. Cu-TiO<sub>2</sub> further calcined at various temperatures (in the range of 500 °C – 700 °C) to evaluate the thermal stability of TiO<sub>2</sub> anatase phase. The physico-chemical properties of the samples were characterised through X-ray diffraction (XRD), Raman spectroscopy, X-ray photo-electron spectroscopy (XPS) and UV-visible spectroscopy techniques. XRD results revealed that the anatase phase of TiO<sub>2</sub> was maintained well, up to 650 °C, by the Cu dopant. UV-DRS results suggested that the visible light absorption property of Cu-TiO<sub>2 </sub>was enhanced and the band gap is reduced to 2.8 eV. Density functional theory (DFT) studies emphasises the introduction of Cu<sup>+</sup> and Cu<sup>2+</sup> ions by replacing Ti<sup>4+</sup> ions in the TiO<sub>2</sub> lattice, creating oxygen vacancies. These further promoted the photocatalytic efficiency. A significantly high bacterial inactivation (99.9%) was attained in 30 mins of visible light irradiation by Cu-TiO<sub>2</sub>.

Staphylococccus aureus is a ubiquitous and opportunistic bacteria associated with high mortality rates. Antimicrobial photodynamic therapy (aPDT) is based on the application of a light source and a photosensitizer that can interact with molecular oxygen, forming Reactive Oxygen Species (ROS) that result in bacterial inactivation. This study aimed to analyze, in vitro, the action of aPDT with Photodithazine® (PDZ) in methicillin-sensitive Staphylococcus aureus (MSSA) and methicillin-resistant Staphylococcus aureus (MRSA) strains. The strains were incubated with PDZ at 25, 50, 75, and 100 mg/L for 15 min and irradiated with fluences of 25, 50, and 100 J/cm2. The internalization of PDZ was evaluated by confocal microscopy, the bacterial growth by counting the number of colony-forming units, as well as the bacterial metabolic activity post-aPDT and the production of ROS. In both strains, the photosensitizer was internalized; the production of ROS increased when the aPDT was applied; there was a bacterial reduction compared to the control at all the evaluated fluences and concentrations; and, in most parameters, it was obtained complete inactivation with significant difference (p < 0.05). The implementation of aPDT with PDZ in clinical strains of S. aureus has resulted in its complete inactivation, including the MRSA strains.

Plasma-activated water (PAW) is generated by treating water with cold atmospheric plasma (CAP) using controllable parameters, such as plasma-forming voltage, carrier gas, temperature, pulses, or frequency as required. PAW is reported to have lower pH, higher conductivity, and higher oxygen reduction potential when compared with untreated water due to the presence of reactive species. PAW has received significant attention from researchers over the last decade due to its non-thermal and non-toxic mode of action especially for bacterial inactivation. The objective of the current review is to develop a summary of the effect of PAW on bacterial strains in foods as well as model systems such as buffers, with a specific focus on fruit and vegetables. The review elaborated the properties of PAW, the effect of various treatment parameters on its efficiency in bacterial inactivation along with its usage as a standalone technology as well as a hurdle approach with mild thermal treatments. A section highlighting different models that can be employed to generate PAW alongside a direct comparison of the PAW characteristics on the inactivation potential and the existing research gaps are also included. The mechanism of action of PAW on the bacterial cells and any reported effects on the sensory qualities and shelf life of food has been evaluated. Based on the literature, it can be concluded that PAW offers a significant potential as a non-chemical and non-thermal intervention for bacterial inactivation, especially on food. However, the applicability and usage of PAW depend on the effect of environmental and bacterial strain-based conditions and cost-effectiveness.

This work presents the comparison of four advanced oxidation processes driven by UVC-LED radiation (278 nm—2 W/m2) for simultaneous bacteria inactivation (Escherichia coli—106 CFU/mL) and microcontaminant removal (imidacloprid—50 µg/L) in simulated wastewater secondary effluent. To this end, the activation of H2O2 and S2O82− as precursors of HO• and SO4•−, respectively, by UVC-LED and UVC-LED/Fe3+–NTA (ferric nitrilotriacetate at 0.1 mM) has been studied at different oxidant concentrations. For the purpose of comparison, conventional chlorination was used as the baseline along with bacterial regrowth 24 h after treatment. Disinfection was achieved within the first 30 min in all of the processes, mainly due to the bactericidal effect of UVC-LED radiation. UVC-LED/H2O2 did not substantially affect imidacloprid removal due to the low HO• generation by UVC irradiation at 278 nm, while more than 80% imidacloprid removal was achieved by the UVC-LED/S2O82−, UVC-LED/Fe3+–NTA/S2O82−, and UVC-LED/Fe3+–NTA/H2O2 processes. The most efficient concentration of both oxidants for the simultaneous disinfection and microcontaminant removal was 1.47 mM. Chlorination was the most effective treatment for bacterial inactivation without imidacloprid removal. These findings are relevant for scaling up UVC-LED photoreactors for tertiary wastewater treatment aimed at removing bacteria and microcontaminants.

AbstractA growing body of literature has recognized the non-thermal effect of pulsed microwave radiation (PMR) on bacterial systems. However, its mode of action in deactivating bacteria has not yet been extensively investigated. Nevertheless, it is highly important to advance the applications of PMR from simple to complex biological systems. In this study, we first optimized the conditions of the PMR device and we assessed the results by simulations, using ANSYS HFSS (High Frequency Structure Simulator) and a 3D particle-in-cell code for the electron behavior, to provide a better overview of the bacterial cell exposure to microwave radiation. To determine the sensitivity of PMR, Escherichia coli and Staphylococcus aureus cultures were exposed to PMR (pulse duration: 60 ns, peak frequency: 3.5 GHz) with power density of 17 kW/cm2 at the free space of sample position, which would induce electric field of 8.0 kV/cm inside the PBS solution of falcon tube in this experiment at 25 °C. At various discharges (D) of microwaves, the colony forming unit curves were analyzed. The highest ratios of viable count reductions were observed when the doses were increased from 20D to 80D, which resulted in an approximate 6 log reduction in E. coli and 4 log reduction in S. aureus. Moreover, scanning electron microscopy also revealed surface damage in both bacterial strains after PMR exposure. The bacterial inactivation was attributed to the deactivation of oxidation-regulating genes and DNA damage.
