Lipid extracts of Listeria monocytogenes, capable of producing monocytosis and lymphopenia in mice, were fractionated by column chromatography on acid-treated Florisil. The biological activity was associated with a phospholipid fraction and further separation of this fraction by thin-layer chromatography indicated that most of the activity was in the slower running components which gave a positive reaction with ninhydrin. Water-soluble ninhydrin-positive material was separated from either the crude lipids or the phospholipid fractions by techniques such as a Folch wash, chromatography on Sephadex, or dialysis of a chloroform solution of the lipids against water. These water-soluble materials were also able to produce monocytosis and lymphopenia in mice, but the remaining phospholipid was still ninhydrin-positive and biologically active. Most of the water-soluble material was dialyzable, but the biological activity appeared to be concentrated largely in the non-dialyzable fraction. This fraction contained protein, and digestion with Pronase appeared to enhance the biological activity and to make the active material more readily dialyzable. Extraction of the lipid-extracted bacterial residue with saline yielded additional non-dialyzable water-soluble material with activity comparable to that shown by the lipid extracts.

ABSTRACT A new method for preparation of Human Menopausal Gonadotrophin involves successively alcoholic precipitation, kaolin adsorption and chromatography on ion exchangers. A highly active material is obtained which corresponds to 1 mg per litre of urine and has an activity of 1 mouse uterus unit at a dose of 0.003 mg. This gonadotrophin possesses both follicle stimulating and luteinizing activities in hypophysectomized female rats, by histological study. It contains 13 % hexose, 10% hexosamine and 8.5 % sialic acid. A further purification, by zone electrophoresis on starch, gives a final product, biologically active at 0.001 mg, which behaves as an homogenous substance in free electrophoresis with mobility −4.76 × 10−5 at pH 8.6.

Holoptelea integrifolia Roxb. Planch (HI) has been used to treat various ailments including obesity, osteoarthritis, arthritis, inflammation, anemia, diabetes etc. To review the major phytochemicals and medicinal properties of HI, exhaustive bibliographic research was designed by means of various scientific search engines and databases. Only 12 phytochemicals have been reported including biologically active compounds like betulin, betulinic acid, epifriedlin, octacosanol, Friedlin, Holoptelin-A and Holoptelin-B. Analytical methods including the Thin Layer Chromatography (TLC), High-Performance Thin Layer Chromatography (HPTLC), High-Performance Liquid Chromatography (HPLC) and Liquid Chromatography With Mass Spectral (LC-MS) analysis have been used to analyze the HI. From medicinal potency point of view, these phytochemicals have a wide range of pharmacological activities such as antioxidant, antibacterial, anti-inflammatory, and anti-tumor. In the current review, it has been noticed that the mechanism of action of HI with biomolecules has not been fully explored. Pharmacology and toxicological studies are very few. This seems a huge literature gap to be fulfilled through the detailed in-vivo and in-vitro studies.

SummaryIt is widely reported that persistent anti-Xa activity follows administration of low molecular weight heparins. To identify the effectors of this activity we have injected 125I-labelled Enoxaparin sodium into rabbits and subsequently analysed the circulating radiolabelled material and anti-Xa activity by affinity and size exclusion chromatography. Antithrombin III-binding material derived from the injected drug was responsible for all the anti-Xa amidolytic activity. At early times after injection additional anticoagulant activity which was largely attributable to tissue factor pathway inhibitor was measured by the Heptest clotting assay after removal of glycosaminoglycans from plasma samples. Small radiolabelled fragments, including penta/hexasaccharide with affinity for antithrombin III, were detectable in the circulation 1 week later, and sulphated oligosaccharides persisted for 3-4 weeks. Significant quantities of radiolabel remained in the liver and kidney several weeks post-injection; these organs may sequester some of the injected drug and give rise to circulating biologically active material by degradation and secretion of catabolic products into the plasma.

Plants are a rich source of effective non-toxic biologically active substances. Various physicochemical methods of analysis are used for evaluation of plant antioxidant activity. Composition of ethanol extracts of red, yellow and white onion husks, dried rosemary, basil, and chaga were studied by high performance thin layer chromatography (HPTLC) method. The antioxidant activity of the obtained fractions on a chromatographic plate was assessed by subsequent DPPH screening. The extracts red and yellow onion husk and rosemary demonstrated the highest antioxidant activity, variability of the qualitative composition and similarity of antioxidant profiles, while extract of white onion husks did not contain any antioxidant classes. Intensive spots with Rf of 0.13-0.97 were observed along the whole chromatogram track corresponding to red onion husks. It was also found that all tested extract, excepting white onion husk and chaga, contained spots with varying degrees of intensity in the Rf range of 0.96-0.98, which corresponded quercetin Rf value.

The State Pharmacopoeia of the XIV edition defines the approach for the assessment of the quality of medicinal plant materials, it deals with the identification of the main groups of biologically active substances by thin layer chromatography. According to this approach, the analysis of some types of medicinal plant materials as a part of some objects of plant origin was carried out. The article presents the quality assessment algorithm as examplified by peppermint leaves (Mentha piperita L.), which are part of a variety of plant object.

Bioassays with soybeans and lentils were used to compare the persistence of 2,4,5-T, triclopyr, and picloram in soil after applying 0.038, 0.38, and 3.8 kg ae/ha under field conditions. Soil samples were collected from 1 to 269 days after spraying and were kept frozen until growth room bioassays were conducted. Triclopyr was slightly less persistent than 2,4,5-T, but neither herbicide was biologically active during the next season. At least 90% of picloram disappeared within 7 months, but its biological activity was more persistent than that of either triclopyr or 2,4,5-T. Nine months after treatment, neither lentils nor soybeans could be grown in soil treated with picloram at 3.8 kg/ka. Triclopyr and 2,4-D had smiilar soil thin layer chromatographic mobilities in four different scils. Picloram was significantly more mobile than either triclopyr or 2,4,5-T, primarily due to its lower adsorption in the soils examined.
