ABSTRACT A role for the Plasmodium falciparum cyclic GMP (cGMP)-dependent protein kinase (PfPKG) in gametogenesis in the malaria parasite was elucidated previously. In the present study we examined the role of PfPKG in the asexual blood-stage of the parasite life cycle, the stage that causes malaria pathology. A specific PKG inhibitor (compound 1, a trisubstituted pyrrole) prevented the progression of P. falciparum schizonts through to ring stages in erythrocyte invasion assays. Addition of compound 1 to ring-stage parasites allowed normal development up to 30 h postinvasion, and segmented schizonts were able to form. However, synchronized schizonts treated with compound 1 for ≥6 h became large and dysmorphic and were unable to rupture or liberate merozoites. To conclusively demonstrate that the effect of compound 1 on schizogony was due to its selective action on PfPKG, we utilized genetically manipulated P. falciparum parasites expressing a compound 1-insensitive PfPKG. The mutant parasites were able to complete schizogony in the presence of compound 1 but not in the presence of the broad-spectrum protein kinase inhibitor staurosporine. This shows that PfPKG is the primary target of compound 1 during schizogony and provides direct evidence of a role for PfPKG in this process. Discovery of essential roles for the P. falciparum PKG in both asexual and sexual development demonstrates that cGMP signaling is a key regulator of both of these crucial life cycle phases and defines this molecule as an exciting potential drug target for both therapeutic and transmission blocking action against malaria.

The single-celled apicomplexan parasite Plasmodium falciparum is responsible for the majority of deaths due to malaria each year. The selection of drug resistance has been a recurring theme over the decades with each new drug that is developed. It is therefore crucial that future generations of drugs are explored to tackle this major public health problem. Cyclic GMP (cGMP) signaling is one of the biochemical pathways that is being explored as a potential target for new antimalarial drugs. It has been shown that this pathway is essential for all of the key developmental stages of the complex malaria parasite life cycle. This gives hope that targeting cGMP signaling might give rise to drugs that treat disease, block its transmission and even prevent the establishment of infection. Here we review previous work that has been carried out to develop and optimize inhibitors of the cGMP-dependent protein kinase (PKG) which is a critical regulator of the malaria parasite life cycle.

Darkling beetle Zophobas atratus (Coleoptera, Tenebrionidae) is recorded here for the first time as a new species for the fauna of Ukraine. Detailed study on morphology of preimaginal stages and biology of this species recently introduced to Ukraine, is given here. Zophobas atratus is an important species being easily reared in laboratory cultures and widely distributed in North and South America, Europe, and Asia. Detailed descriptions of all life stages, including egg, young and older larvae, pupa and adult of Z. atratus are required for further taxonomical study of the genus Zophobas, which isn’t yet definitively established. New identification keys for adults and larvae of the genera of tribe Tenebrionini are presented here, based on a comparative analysis of the taxonomic characters of adults and larvae of Z. atratus and species from the genera Tenebrio and Neatus. Comparative analysis of morphology of larva of Z. atratus and larvae of the tribe Cteniopodini of close subfamily Alleculinae was conducted here. The subfamily Alleculinae previously had the rank of family Alleculidae, but our analysis confirmed the reliability of its current taxonomic position as subfamily. The study of morphology of larvae of 1st and 2nd instars of Z. atratus revealed that they are characterized by special taxonomic structures that are not characteristic for oldest instars of larvae of Z. atratus. These characters include absence of spines on caudal segment, presence of a set of 4 setae at posterior margin of tergites of prothorax, metathorax, and 1st to 8th abdominal segments, strongly convex 2nd antennal segment and sensory zone in the form of an open ring on its apex, and etc. Moreover, structure of antenna of larvae of Z. atratus is similar to that of oldest larvae of most species of darkling beetles of the fauna of Ukraine. The most distinctive features of Z. atratus are: sexual dimorphism in structure of clypeus of adults; filiform sclerotized antenna of larva with a continuous sensory zone at apex of 2nd segment, weakly developed 3rd segment; fusion of sclerotized pleurites of 1st–8th abdominal segments with their tergites; sexual dimorphism in structure of 9th abdominal segment of pupa, and presence of two hooks on apex of its appendages. The data of original study of features of life cycle of Z. atratus are given. The pictures and photos of details of morphology of egg, larvae, pupa and adult of Z. atratus are presented. It was recored that life cycle of Z. atratus from laying of egg to the emergence of adult continues from 169 up to 181 days. Adults lived maximum up to 206 days. Maturation of eggs in female after copulation continues 10-11 days. Stage of egg continues 7 days, larva – up to 151 days, including pre-pupal period from 6 to 22 days, pupa – from 8 to 21 days. Twelve larval stages of Z. atratus were recorded in laboratory culture.

Subject This article deals with the issues of determining the level of financial literacy, identifying problems, and comparing the financial literacy rates of the population of different areas. Objectives The article aims to identify territorial differences in the overall level of financial literacy of the population of the Northwestern Federal District regions and study the prevalence of problems related to the family budget, using financial products, and risk. Methods For the study, we used a sociological survey and an original methodology for indexing the level of financial literacy of the population based on the methodology of the Organization for Economic Cooperation and Development, taking into account the adults' financial competence system proposed by the Ministry of Finance of the Russian Federation. Results The article determines the level of financial literacy of the Northwestern Federal District population and identifies similar problems for the District's regions. Conclusions A comparative analysis of the components of financial literacy of the four regions reveals the strengths and weaknesses of each of them. This will help adjust financial literacy programmes being implemented in some regions and developed in others.
