A note on the effect of amino acid or urea supplements to barley diets for the finishing pig

SUMMARYIn trials 1 and 2, supplements of urea and cottonseed meal increased the protein content of the basal diets from 12·2 to 14·0–14·6%, but had no significant effects on growth rate or carcass characteristics. In trial 3 the effect of cottonseed meal on growth rate was almost significant but urea had less effect. In trial 4, the basal diet contained 11·0% protein; soya bean meal increased this to 13·1% and had an almost significant effect on growth rate. Lysine and lysine + urea had no effect.

1. Three growth trials were done using male broiler chicks. In the first two trials, groundnut meal was used, with and without supplementary methionine and lysine. In the third trial, soya-bean meal was used with and without supplementary methionine. Protein levels ranged in the first trial from 120 to 420 g/kg diet and in the third trial from 120 to 300 g/kg diet. Thus the assumed minimal amino acid requirements of the chick were supplied by high levels of low-quality dietary protein.2. Diets based on cereals and groundnut meal did not support maximum live-weight gain or maximum efficiency of food utilization at any level of dietary protein. When the principal deficiencies of lysine and methionine were corrected, this protein mixture was capable of supporting the same growth rate as a control diet of cereals and herring meal.3. Diets based on maize and soya-bean meal did not support quite the same growth rate as similar diets supplemented with methionine, even though the protein level in the unsupplemented diets was sufficient to meet the assumed methionine requirements.4. These results are interpreted as examples of amino acid imbalance in diets composed of familiar feeding-stuffs. It is concluded that one cannot assume that the poor quality of a protein source can always be offset by increasing the concentration of dietary protein.

ABSTRACTNitrogen balance was measured in a series of experiments to determine the optimum balance between lysine, threonine and methionine plus cystine by successive additions of the free amino acids to a basal diet of barley, soya bean meal and tallow. The basal diet had a calculated digestible energy (DE) value of 14·5 MJ/kg and a crude protein content of 146 g/kg. The ranges of total lysine; threonine and methionine plus cystine tested were (g/kg) 6·6 to 11·8, 4·9 to 6·9 and 4·3 to 71, respectively. Latin-square designs were used involving a total of 33 pigs. Nitrogen utilization was optimized in pigs of 3 to 9 weeks of age when each kg of diet contained 10·5 g lysine, 6·4 g threonine and 5·0 g methionine plus cystine, corresponding to a ratio of 100: 61: 48. Thereafter, five diets were formulated in which the same ratio was maintained, but at different protein levels (129 to 192 g/kg). Again the diets were based on barley, soya bean meal and tallow supplemented with free amino acids. These diets were compared, in both nitrogen balance and performance experiments, with a conventional early-weaning diet with a protein content of 240 g/kg. All six diets had the same calculated DE value (14·5 MJ/kg). The N balance experiment was of Latin-square design and involved 18 pigs, and the performance experiment used 36 pigs fed individually from 3 to 9 weeks of age. A diet containing 0·99 g lysine per MJ DE (14·4 g lysine and 192 g balanced protein per kg) was considered to be adequate on the basis of the growth rate and food conversion efficiency results obtained.

SummaryA 4 x 4 latin-square design experiment was carried out to determine the effects of increasing nitrogen (N) intake by feeding diets containing increasing amounts of soyabean meal upon the digestion of organic matter (OM) and N by cattle equipped with rumen and duodenal cannulae. A basal diet (B) containing 600 g ground barley and 400 g grass silage/kg diet and three diets (BS1, BS2 and BS3) in which increasing amounts of the barley were replaced by soya-bean meal were fed. The mean daily intakes of OM and N when each diet was fed were 4·56, 4·55, 4·30 and 4·52 kg OM and 920, 114·7, 138·3 and 164·1 g N for diets B, BSl, BS2 and BS3 respectively.Neither the amounts of OM entering the small intestine nor those voided in the faeces were altered by the diets fed. Thus the mean apparent OM digestibility for all the diets fed was 0·74 + 0·013 and the proportion of digestible OM intake apparently digested in the rumen was 0·77 + 0048.Mean daily concentrations of ammonia N in the rumen were significantly (P < 0·001) increased from 38 mg N/l (diet B) to 129 mg N/l (diet BS3) as N intake increased.The quantities of non-ammonia N and of amino acid N entering the small intestine were not significantly (P >0·05) increased as more soya-bean meal was added to the diet, although diet BSl supported the greatest flows of N to the small intestine. Thus as more soya-bean meal was added to the diet there were increasing net losses of nonammoniaN(P < 0·01) and amino acid N (P < 0·01) prior to the small intestine. Faecal N excretion was not increased (P > 0·05) as soya-bean meal intake increased and thus apparent N digestibility was significantly (P < 0·01) increased by increasing soyabean intake.Both the quantities of microbial N entering the small intestine daily and the apparent efficiency of microbial N synthesis within the rumen were increased when diet BSl was fed in comparison with the basal diet (B) and then declined when diets BS2 and BS3 were fed; these increases were not significant. The quantities of feed N entering the small intestine daily were not significantly (P > 0·05) increased as soya-bean meal intake increased; thus apparent feed N degradability in the rumen was significantly (P < 0·01) increased as soya-bean meal intake increased. In contrast, the rates of disappearance of N from each of the components of the diets fed, when incubated in the rumens of the cattle in porous synthetic fibre-bags, were not increased (P > 0·05) as soya-bean meal intake increased.

1. In a 4 x 4 Latin square experiment four cows were given, twice daily, diets consisting of (g/kg dry matter (DM)) 500 barley, 400 grass silage and 100 soya-bean meal. The diets were given at either 1.15 (L) or 2.3 (H) times maintenance energy requirements and the soya-bean meal was either untreated (U) or formaldehyde (HCH0)-treated (T).2. The passage of digesta to the duodenum was estimated using chromic oxide as a flow marker;35S was used to estimate the amount of microbial protein entering the small intestine. A microbial fraction was prepared by differential centrifugation from duodenal digesta. Samples of bacteria and of protozoa from rumen digesta were also prepared.3. The total amino acid contents of feedingstuffs, duodenal digesta, duodenal microbial material, rumen bacteria and rumen protozoa were determined by ion-exchange chromatography. The D-alanine and D-glutamic acid contents of the samples were determined by gas–liquid chromatography.4. The quantity of each amino acid entering the small intestine was significantly (P < 0,001) increased by increasing DM intake and tended to be increased by formaldehyde-treatment of the soya-bean meal. There were net losses of all amino acids across the forestomachs except for lysine, methione, o-alanine and D-glutamic acid for which there were net gains.5. There were significant (P < 0.05) differences in amino acid composition between rumen bacteria and duodenal microbial material; differences in amino acid composition between rumen bacteria and rumen protozoa were also observed.6. D-Alanine and D-glutamic acid were present in the silage but not in the barley or either of the soya-bean meals. All samples of microbes and digesta contained D-alanine and D-glutamic acid.7. The use of D-ahine and D-glUtamiC acid as markers for microbial nitrogen entering the small intestine was assessed. Estimates of the quantities of microbial N entering the small intestine based on the D-alanine or D-glutamic acid contents of rumen bacteria or duodenal microbes were significantly higher than those determined using 35S as a marker.

1. Young male rats were used in five experiments to study the utilization for growth of methionine sulphoxide, and the relationship between the sulphoxide content in the diet and the level of microbiologically determined methionine activity in blood or blood plasma. In one nitrogen-balance experiment methionine and methionine sulphoxide were compared as supplements to a casein diet and a fish-meal diet.2. Methionine sulphoxide was poorly utilized for growth when tested as the sole sulphur amino acid in an amino acid diet. Substitution of one-third of the sulphoxide with cystine improved utilization so that it approached that of methionine.3. Methionine alone and in combination with methionine sulphoxide were added to a soya-bean-meal diet. The sulphoxide showed no adverse effect on growth.4. Fish meal in which methionine had been oxidized to methionine sulphoxide was tested alone and in combinations with unoxidized fish meal. Only when the oxidized meal was given alone was there an appreciable effect on growth. The fish meals used were low in cystine.5. Whereas both methionine and methionine sulphoxide improved the N balance when a casein diet was given, there was no effect when a fish-meal diet was given.6. There was a linear relationship between methionine sulphoxide content in the amino acid diets and the methionine activity in the blood plasma. Methionine sulphoxide added to a soya-bean-meal diet or present in oxidized fish meal gave a curvilinear relationship, and the observed activities were lower than with the amino acid diets. Methionine activity in blood could not be used as an indicator of moderate amounts of methionine sulphoxide in protein-containing diets.

SUMMARY1. Nitrogen balance studies were made on 8 Friesian steers given allconcentrate diets containing fish meal, soya bean meal, groundnut meal or cottonseed meal.2. There were no differences in nitrogen retention but voluntary feed intake was significantly lower on the fish meal diet.3. Adjustment of nitrogen retention to constant dry matter intake resulted in significantly higher values for the diets containing fish meal and cottonseed meal than for the others.4. Rumen pH, rumen ammonia and blood urea were all much lower on the fish meal diet.

ABSTRACTSix diets based on maize and soya-bean meal were prepared. The basal diet had a crude protein (CP) concentration of 171 g/kg. The same ingredients, together with maize starch, were used to give a diet with CP 131 g/kg. This diet was supplemented with all (positive control) or with three of the four amino acids lysine, threonine, tryptophan and methionine. Each diet was given at two rates of intake. From the results, the optimum ratio between these four amino acids at different planes of intake was calculated. Amino acid digestibility in the basal diet was measured at three intakes, using pigs fitted with T cannulas at the terminal ileum.The results showed that the optimum ratios between the four amino acids (expressed in terms of digestible amino acids) were not affected by the rate of food intake. The optimum ratio between the four amino acids (i.e. when they are equally limiting) was lysine 1·00, methionine + cystine 0-61, threonine 0·64 and tryptophan 0·20. Digestibility was not affected by the level of intake.

AbstractTwo experiments were carried out with weaned piglets from 8 to 25 kg live weight in order to determine the dietary amino acid content necessary for maximum growth. Six diets based on wheat, soya-bean meal, soya-bean oil and free amino acids were compared in each experiment. Essential amino acids were in the same relative proportion for all diets: (methionine + cystine)/lysine = 0·60 to 0·65; threonine/lysine = 0·65; tryptophan/lysine = 0·19. The range of amino acid content was 9·53 to 12·52 g lysine per kg in the first experiment and 11·34 to 15·94 g lysine per kg in the second experiment. The number of piglets used per diet was 136 (20 pens) and 106 (16 pens) in respectively the first and the second experiment. The relationship between either dietary lysine content or daily lysine intake and growth rate was quadratic and significant. Dietary lysine content and daily lysine intake which enable maximum growth were calculated according to this model. Dietary lysine contents were 15·5 and 14·9 g/kg for the first 3 weeks (8 to 17 kg) and for the overall post-weaning period (8 to 25 kg) respectively. Daily lysine intakes were 10·6 and 13·3 g/day respectively for the same two periods. Reasons for these values being higher than those currently cited are discussed.

AbstractAn experiment was carried out in growing chickens to study the effect of supplementation of a semi-synthetic diet containing soya-bean meal as the sole protein source with DL-methionine, to improve its biological value, on the excretion of endogenous protein and amino acids measured in lower ileum and total tract using traditional methods. Thirty-two White Rock male broilers (10 days old) were randomly divided into eight groups each of four birds, of similar body weight (mean live weight: 142·8 (s.e. 0·68) g), and individually housed in metabolism cages. Following a paired-feeding design based on metabolic body weight (kgM0·75), each group of birds was given, for an experimental period of 20 days, each of four levels of protein (60, 120, 180 or 240 g/kg; 5 days each) in two groups of isoenergetic (14·5 kJ metabolizable energy per g dry matter) and semi-synthetic diets based on soya-bean meal, either not supplemented or supplemented with 2 g/kg DL-methionine (diets S and SM, respectively). After 3 days of each treatment excreta were collected for 48 h, frozen and stored at –20ºC. At the end of the fourth treatment three chickens of each group were killed and their lower ileal contents collected. The remaining chick of each treatment was fasted for 24 h and given a protein-free diet for 8 days and excreta were collected for the last 4 days. Then (day 39 of age), chickens were killed and lower ileum contents removed and stored at –20ºC. Samples of excreta and lower ileum contents were subjected to nitrogen (N) analysis by Kjeldahl procedure and amino acid (AA) analysis by high-performance liquid chromatography. Supplementation with DL-methionine of the soya-bean meal-based diets halved total tract endogenous AA losses. Regression analysis produced a higher estimation of ileal and faecal endogenous AA excretion than feeding a protein-free diet. Endogenous AA excretion determined in the lower ileum was higher than in excreta no matter which estimation procedure was utilized. In conclusion, supplementation of dietary protein with the first limiting AA to improve its protein quality, causes an important drop in endogenous AA losses, that may have an important effect on the N economy and energy requirements in poultry. The use of regression analysis on excreta data where graded amounts of protein are given to growing chickens, seems a suitable method for determining endogenous AA losses provided that good quality proteins are used.

Two experiments were conducted to determine the utilization of ileal digestible isoleucine by growing pigs. In the first, the apparent ileal digestibility of amino acids in cottonseed meal, lupin-seed meal and soya-bean meal was determined in pigs fitted with‘T‘-shaped cannulas. In the second, three isoleucine-deficient diets were formulated to 0.23 g ileal digestible isoleucine/MJ digestible energy (DE) with the three protein concentrates contributing the only source of isoleucine in sucrose-based diets. An additional three diets were formulated with supplements of isoleucine to confirm that isoleucine was limiting in the first three diets. The growth performance and retention of isoleucine by pigs given the six diets over the 20–45 kg growth phase were then determined. The apparent ileal digestibility of isoleucine in the three protein concentrates (proportion of total) was: cottonseed meal 0.68, lupin-seed meal 0.86, soya-bean meal 0.86. There were no significant differences (P > 0.05) in growth rates (g/d) and crude protein deposition rates (g/d) of the pigs given the three diets formulated to 0.23 g ileal digestible isoleucine/MJ DE: cottonseed meal 590, 84; lupin-seed meal 613, 87; soya-bean meal 594, 91 (SEM 13.0, 2.9) respectively. The response of pigs to the addition of isoleucine confirmed that isoleucine was limiting in these diets. The proportion of ileal digestible isoleucine retained by pigs given the cottonseed meal (0.65) was slightly lower than that retained by pigs given soya-bean meal (0.73; P < 0.05). These results indicate that values for the ileal digestibility of isoleucine in protein concentrates more closely reflect the proportion of isoleucine that can be utilized by the pig than occurs for other amino acids such as lysine, threonine and methionine.
