The level of free amino acids in Diclidophora merlangi is high, comprising over 500 μmoles/g ethanol extracted dry weight. A single amino acid, proline, constitutes some 70% of the total pool. Analysis of parasite protein and host blood and mucus revealed low proline levels, suggesting that the high free pool content was not related to a requirement for protein synthesis or to its abundance in the diet of the worm. Experiments revealed that proline was not involved specifically in osmoregulation, and the reasons for the large amounts present in Diclidophora remain unknown.

Concentrations of the following Ninhydrin-reacting substances (NRS) were determined in the unhydrolyzed protein-free fraction of mouse liver by column chromatography: phosphoethanolamine, taurine, urea, aspartic acid, threonine, serine, glutamine, proline, glutamic acid, glycine, alanine, valine, cystine, methionine, isoleucine, leucine, tyrosine, phenylalanine, ß-alanine, ß-aminoisobutyric acid, α-aminobutyric acid, ornithine, ethanolamine, lysine, histidine, and arginine. The NRS present in highest concentration was taurine. Adrenalectomy, fasting for 24 hr, and cortisol administration had little effect on the sum of NRS or individual amino acids. Administration of cortisol did, however, decrease the concentration of amino acids in fasted adrenalectomized animals but increased their concentration in nonfasted adrenalectomized animals. Since the concentration of amino acids was lowered or raised under circumstances known to increase protein synthesis, it is concluded that increased protein synthesis promoted by cortisol is independent of the total pool of amino acids in liver.

Observations have been made on the specific free amino acids (chromatographic analysis) and other nitrogenous constituents in the serum and urine of normal and arthritic men under controlled dietary conditions, before and after a glycine load and adrenocorticotropin administered separately and together.Differences in the metabolism of amino acids between normal individuals and arthritics, particularly of alanine, proline, glutamic acid, taurine, and possibly tyrosine (and/or tryptophan) and cystine, were apparent. The differences were largely confined to the non-essential amino acids. Concomitant increases seen in the serum levels and in the excretion of several amino acids, in response to a load of a single amino acid, indicate that the increases in excretion are more than a matter of competition for reabsorption in the kidney. A prerenal phenomenon appears to be involved, possibly interconversion of amino acids. This concept is supported by the evidence that the increases in the serum levels were restricted to the non-essential amino acids.

1. The catabolism of [14C]phenylalanine was used to indicate the effects of varying the dietary level of lysine and threonine on the retention of dietary amino acids by 2-week-old pigs receiving diets containing skim milk and a mixture of free amino acids.2. Reducing the dietary level of lysine from 16 to 12 g/kg had no influence on phenylalanine oxidation, reducing the lysine level from 12 to 11 then to 10 g/kg caused an almost linear increase in phenylalanine oxidation whereas further reduction to 9 or 8 g/kg resulted in a less-marked increase in phenylalanine oxidation. This showed that 12 g lysine/kg was required to maximize amino acid retention and indicated that lysine was conserved more effectively at low dietary concentrations than at dietary concentrations approaching the requirement.3. Reducing the dietary level of threonine from 8 to 6 g/kg had no influence on phenylalanine oxidation, whereas further reduction to 4 g/kg caused a linear increase in phenylalanine catabolism showing that 6 g threonine/kg was required to maximize amino acid retention.4. Reduction of the levels of lysine, threonine and methionine from the generous levels characteristic of a diet containing 240 g protein from skim milk/kg, to the requirement levels determined separately in the presence of the generous levels of all the other amino acids, resulted in a twofold increase in phenylalanine catabolism. This shows that the pig seems able to conserve limiting intakes of a single amino acid, but not if the intakes of two or three amino acids are limiting.

Free amino acid pools were examined for cultures of vegetative cells, gametes, and mature zygotes of the unicellular green alga Chlamydomonas reinhardtii (Dangeard). The total pool of amino acids found in premature gametes of strains 137c+ (10.0 pmol∙μg protein−1) and 137c− (10.8 pmol∙μg protein−1) decreased to levels about half that seen in vegetative 137c− cells (19.8 pmol∙μg protein−1). Following light activation, amino acid pools in these gametes increased to 18.7 pmol∙μg protein−1 in 137c+ cells and 20.0 pmol∙μg protein−1 in 137c− cells. With the exception of cystine, individual amino acid pools in these cells had increased once more to levels similar to those seen in vegetative cells grown in liquid medium. Levels of cystine remained one to two orders of magnitude lower than that seen in vegetative cells. Mature 137c + and 137c− gametes mixed in solutions of either 2 mM cystine or 2 mM cysteine (half-cystine) suffered a 52–64% reduction, respectively, in the number of vis-à-vis conjugative pairs formed. This suggests that pools of endogenous cystine may play a role in the onset of mating. In zygotes levels of all amino acid pools, except histidine, were depressed; levels of cystine, valine, and phenylalanine were nondetectable in these cells.Key words: Chlamydomonas reinhardtii, amino acid pools, gametes, zygotes, cystine.

AbstractThe effect of a 2-month infestation by the cassava mealybug (Phenacoccus manihoti Mat. Ferr.) on the metabolism of nitrogen (amino acids) and carbon (carbohydrates), leaf area and total dry weight of five cassava varieties (Manihot esculenta Crantz), faux-caoutchouc (a hybrid of M. esculenta and M. glaziovii Muell, Arg.), poinsettia (Euphorbia pulcherrima Wild) and talinum (Talinum triangulare Jack) was studied. Free amino acid and free sugar contents as well as relative free amino acid composition in the leaf extracts, although found to be very different from one plant to another, were not significantly modified by P. manihoti infestation, except for the total amino acid contents of the cassava variety 30M7. Variations in one particular amino acid induced by mealybug infestation were not linked to the antibiotic resistance of these plants. Infestation by the cassava mealybug did not modify the total dry weight but reduced the total leaf area although this reduction was only significant in cassava varieties 59M2, 30M7 and M'pembe, and in faux-caoutchouc. Within the genus Manihot, the reduction in leaf area was strongly correlated (r= -0.878, P≤0.05) to the degree of antibiotic resistance and was coupled to an increase in the ratio of sugars to amino acids, suggesting a similarity between the effects of water stress and those of mealybug infestation.

Abstract We report for the first time the concentrations of free amino acids in human intestinal biopsies obtained by routinely performed endoscopy. We studied 15 medical patients with no changes of the mucosa and six HIV-infected persons with duodenitis. The mean (and SD) sum of all amino acids, taurine excepted, was 61.9 (5.4) mmol/kg dry weight in duodenal biopsies of HIV-negative subjects (n = 11) and 82.9 (0.6) mmol/kg in colonic specimens: 50% (44%) of the total (minus taurine) consisted of aspartate and glutamate and 14% (12%), of the essential amino acids. The relative amino acid pattern in duodenum and colon differed completely from that for muscle: aspartate was fourfold higher; glutamate, phenylalanine, glycine, valine, leucine, and isoleucine were about twofold higher. In contrast, glutamine amounted only to 4% (duodenum) to 14% (colon) of muscle glutamine. In duodenal biopsies of the HIV-infected persons, we found significantly (P less than 0.01, except glutamine: P less than 0.025) increased concentrations of glutamate (24.1 vs 17 mmol/kg dry weight), ornithine (1.4 vs 0.4), valine (2.2 vs 1.7), and glutamine.

It is not appropriate to compare ratio-based expressions for different cultivars or treatments if a plot of the denominator versus the numerator of a ratio-based expression has a nonzero y-intercept and the values for either the denominators or numerators differ with cultivars or treatments. Whenever nonzero y-intercepts are encountered, the value for a ratio-based expression will be dependent on both the denominator and numerator. The “ratio problem” is demonstrated with shoot N concentration in blueberries (Vaccinium corymbosum L.) and amino acid accumulation in almonds [Prunis dulcis (Mill.) D.A. Webb]. Data were collected from the first and second growth flush of blueberry shoots on plants that were at two in-row spacings and two rates of N fertilizer. Free amino acid:total amino acid ratios were measured in dormant almond trees fertilized at different rates with and without foliar N supplements. Functions describing the relationship between dry weight and total N content in blueberry tissues have positive y-intercepts for both N fertilizer application rates. Functions describing the relationship between total amino acids and free amino acids in almond trees have a negative y-intercept. Differences attributable to fertilization rate in blueberries probably were the result of differences in N uptake and N utilization, but the effects of spacing and growth flush are indirect and can be accounted for by differences in dry weight. Likewise, effects of fertilization rate and foliar N supplement in almonds are indirect and can be accounted for by differences in the total amino acids in dormant trees. With regression one can determine if the relationship between the denominator and numerator differs for the groups or treatments being studied. When an analysis of covariance is used to account for differences in the denominators of ratio-based expressions, results are consistent with the regression analysis. When a conclusion is based on statistical differences of a ratio-based expression, it is the researcher's responsibility to determine whether these effects are direct or indirect.

SummaryHaemolymph free amino acids (HFAAs) in adult Tenebrio molitor varied in concentration with beetle age. Between 9 and 30 days post-emergence the concentration range was 34–94 mM with a peak occurring in 15-day-old insects. Much of this age-related variation was due to changes in the concentration of a single amino acid, proline. An analysis of concentrations of individual HFAAs in 15-day-old beetles, following infection with metacestodes of Hymenolepis diminuta, revealed that changes in female insects are more marked than those occurring in males. In the former, changes occurred in the concentration of 9 amino acids (isoleucine, leucine, arginine, serine, threonine, tyrosine, phenylalanine, proline and alanine/citrulline) whereas in the latter only 4 amino acids were affected (threonine, glycine, histidine and arginine). It is suggested that H. diminuta may interact with the endocrine system of its beetle host.

Abstract Objectives The rate of protein digestion and amino acid absorption determines the postprandial rise in circulating amino acids and, as such, modulates postprandial muscle protein synthesis rates. This study compares protein digestion and amino acid absorption kinetics and the subsequent muscle protein synthetic response following ingestion of intact protein versus an equivalent amount of free, crystalline amino acids. Methods Twenty-four healthy, young subjects (age: 22 ± 3 y, BMI: 23 ± 2 kg·m−2, sex: 12 M/12F) ingested 30 g intrinsically L-[1–13C]-phenylalanine and L-[1–13C]-leucine labeled milk protein (PROT; n = 12) or an equivalent amount of free amino acids (AA; n = 12). In addition, subjects received primed continuous L-[ring-2H5]-phenylalanine, L-[ring-3,5–2H2]-tyrosine, and L-[1–13C]-leucine infusions. Blood samples and muscle biopsies were obtained frequently to assess protein digestion and amino acid absorption kinetics and subsequent muscle protein synthesis rates over a 6 h postprandial period. An unpaired t-test was used to compare overall exogenous phenylalanine release in plasma. For other parameters repeated measures ANOVA were applied to determine differences between groups over time (time as within, and group as between-subjects factor). Data are expressed as mean ± SD. Results Postprandial plasma amino acid concentrations and exogenous phenylalanine appearance rates increased after ingestion of PROT and AA (both, P < 0.001), with a greater increase following ingestion of AA when compared to PROT (time*group interaction P < 0.001). Exogenous phenylalanine release in plasma assessed over the 6 h postprandial period, was greater in AA (76 ± 9%) compared with PROT (59 ± 10%; P < 0.001). Ingestion of AA and PROT strongly increased muscle protein synthesis rates based upon L-[ring-2H5]-phenylalanine (time effect P < 0.001), with no differences between groups (from 0.037 ± 0.015 to 0.053 ± 0.014%·h−1 and from 0.039 ± 0.016 to 0.051 ± 0.010%·h−1, respectively; time*group interaction P = 0.629). Conclusions Ingestion of free amino acids as opposed to intact milk protein is followed by more rapid amino acid absorption and greater postprandial plasma amino acid availability, but this does not further augment postprandial muscle protein synthesis rates.
