AbstractThe plasma concentrations of cortisol and progesterone during reproductive maturation in the heifer calf were examined. Six beef heifer calves were handled and bled every 2 weeks (control), 30 were left unhandled (naive). At 13, 21, 30, 39 and 47 weeks of age, a different group of naive heifers and the control heifers were bled every day for 5 days (puberty seen at 57·4 (s.e. 1·3) weeks). Thirty-nine dairy heifer calves were allocated to seven groups to receive adrenocorticotropic hormone (ACTH) at either 4, 7, 9, 16, 26, 36 or 46 weeks of age (puberty expected around 43 weeks of age). Plasma concentrations of cortisol increased at 21, 30 and 39 weeks of age in frequently handled and naive beef heifers; the increase was greater at these ages in the naive beef heifers (age and treatment P < 0·01). No age trend was apparent for plasma concentrations of cortisol and progesterone prior to ACTH injections (t = 0) in frequently handled dairy heifers. Adrenal progesterone secretion did not change with age in beef heifers, but naive beef heifers had greater plasma concentrations than frequently handled beef heifers. ACTH induced cortisol release in dairy heifers as early as 4 weeks of age (P < 0·05), but the response was greater in dairy heifers 16 weeks old and older (P < 0·05). A progesterone response to ACTH in dairy heifers was not seen until animals were 9 weeks old. It is concluded that as heifers mature reproductively, there is a parallel increase in the sensitivity of the adrenal gland to ACTH and handling stress.

Abstract The development of replacement heifers is at the core of cow-calf beef production systems. In 2020, the USDA, National Agricultural Statistics Service reported 5.771 million beef heifers, 500 pounds and over, are under development for cow replacement. A compilation of data from several studies indicate that between 85% and 95% of these heifers will become pregnant in their first breeding season. Several thousands of heifers being raised for replacement may not deliver a calf on their first breeding season and result in economic losses to cow-calf producers. Many management procedures have been developed to maximize the reproductive potential of beef heifers. Such approaches include, but are not limited to the following: nutritional management for controlled weight gain, identification of reproductive maturity by physiological and morphological indicators, and the implementation of an estrous synchronization program. The implementation of management strategies has important positive impact(s) on the reproductive efficiency of heifers. There are limitations, however, because some heifers deemed ready to enter their first breeding season do not become pregnant. In parallel, genetic selection for fertility-related traits in beef heifers have not promoted major genetic gains on this particular area, most likely due to low heritability of female fertility traits in cattle. Technologies such as antral follicle counting, DNA genotyping and RNA profiling are being investigated as a means to aid in the identification of heifers of low fertility potential. To date, many polymorphisms have been associated with heifer fertility, but no DNA markers have been identified across herds. Antral follicle count is an indication of the ovarian reserve and is an indicator of the reproductive health of a heifer. We have been working on the identification of transcriptome profiles in heifers associated with pregnancy outcome. Our current investigations integrating protein-coding transcript abundance and artificial intelligence have identified the potential for bloodborne transcript abundance to be used as indicators of fertility potential in beef heifers. In summary, there is an ongoing pressure for reducing costs and increasing efficiency in cow-calf production systems, and new technologies can help reduce the long-standing limitations in beef heifer fertility.

SummaryThirty-two patients who had responded to amitriptyline (150 mg daily) when suffering from a depressive illness were allocated either to receive placebo or to remain on the same medication for one year.Plasma concentrations of the drug were regularly estimated. There was no correlation between plasma concentration and subsequent residual affective morbidity. In spite of considerable encouragement, three of the patients did not take the prescribed amitriptyline and they all relapsed. Five out of sixteen patients who received placebo relapsed. None of the patients who continued to take amitriptyline relapsed.It is emphasized that the patients studied were selected, inasmuch as they were apparent responders to amitriptyline. It is concluded that this group of patients should continue to be treated with antidepressant medication for eight months after apparent recovery, and care should be taken to ensure the patients' compliance.

Abstract Option 1, Fetal Programming Only: Non-lactating, pregnant, multiparous Angus × Hereford cows (n = 104) were ranked by pregnancy sire (1 of 2 sires), body weight (BW), and body condition score (BCS) on d -15 of the experiment (d 180 of gestation). Cows were then assigned to receive: (1) 195 g/cow daily of CSSO (n = 52) or 2) 170 g/cow daily of CON (n = 52). Cows received treatments individually from d 0 to calving. Calves were weaned on d 290 of the experiment, preconditioned for 35 d, and placed in a feedyard until slaughter (d 514). Cows receiving CSSO and their calves had greater (P &lt; 0.01) plasma concentrations ω-6 PUFA compared with CON after calving. Colostrum IgG and calf plasma IgG concentrations 24 h after birth were greater (P ≤ 0.02) in CSSO vs. CON cattle. Calves from CSSO cows had greater (P ≤ 0.05) expression of adipogenic and myogenic genes in the longissimus muscle compared with CON. No treatments differences in birth BW, weaning BW, and final preconditioning BW were noted (P ≥ 0.36). Average daily gain and final BW in the feedyard were greater (P ≤ 0.05) in steers from CSSO cows compared with CON. The incidence of calves diagnosed with BRD that required a second antimicrobial treatment was less (P = 0.03) in calves from CSSO cows, resulting in reduced (P = 0.05) need of treatments to regain health compared with CON. Upon slaughter, LM area was greater (P = 0.03) in calves from CSSO cows compared with CON. Collectively, these results are indicative of programming effects on postnatal offspring growth and health resultant from CSSO supplementation to late-gestating cows. Hence, supplementing CSSO to beef cows during pregnancy might be a feasible alternative to optimize offspring productivity and welfare. Option 2, Fetal Programming and Fat Tubs: Two experiments were conducted to evaluate: (1) effects of calcium salts of soybean oil (CSSO) supplementation to late gestating beef cows on the performance of the offspring; (2) the viability of utilizing low-moisture molasses-based blocks (LMB) as a delivery method for CSSO supplementation. In experiment 1, non-lactating, pregnant, multiparous Angus × Hereford cows were assigned to receive: (1) 195 g/cow daily of CSSO (n = 52) or (2) 170 g/cow daily of CON (n = 52) from d 195 of gestation until calving. CSSO cattle had greater (P &lt; 0.01) plasma concentrations ω-6 PUFA; greater (P ≤ 0.02) colostrum and calf plasma IgG concentrations; greater (P ≤ 0.05) expression of adipogenic and myogenic genes in the longissimus muscle (LM); required fewer microbial treatments for BRD (P = 0.05) and had greater LM area compared to CON cohorts. In experiment 2, non-lactating, non-pregnant, multiparous beef cows were allocated to 1 of 9 pens (n = 9 pens; 4 cows/pen) and each pen was assigned to receive: 1) NOSUPP, hay only diet; 2) LMB, self-fed LMB supplement containing 24.7% DM CSSO; 3) CONC, hand-fed supplement identical to LMB. The design was a replicated 3 x 2 Latin square with 2 periods of 42 d, and a 21-d washout interval. Supplement DM intake did not differ (P = 0.39) between LMB and CONC cows from d 14 to 42 as designed, despite a greater variation in daily intake of LMB vs. CONC (treatment × day interaction; P &lt; 0.01). No treatments effects were noted (P ≥ 0.40) for hay intake, BCS, and BW. From d 14 to 42, plasma concentrations of, ω-6 PUFA (P &lt; 0.01) in CONC and LMB vs. NOSUPP cows. Hence, the use of self-fed LMB appears to be a valid strategy to provide CSSO to forage-fed beef cattle.

Purpose To test the benefits of Solumedrol treatment in sepsis patients with a blunted adrenocorticotropic hormone (ACTH)-cortisol response (delta <13 µg/dL) with regard to the number of days on ventilator, days on intravenous blood pressure support, length of time in an intensive care unit (ICU), 14-day mortality, and 28-day mortality. The trial was prospective, randomized, and double-blind. As part of a larger sepsis trial, 54 patients with sepsis had an intravenous ACTH stimulation test using 250 µg of ACTH, and serum cortisol was measured at times 0, 30, and 60 min. Eleven patients failed to increase their cortisol concentration above 19.9 µg/dL and were excluded from the clinical trial as they were considered to have adrenal insufficiency. The remaining 43 patients had a baseline cortisol of 32 ± 1 µg/dL increased to 38 ± 3 µg/dL at 30 min and 40 ± 3 at 60 min. All cortisol responses were <12.9 µg/dL between time 0 and time 60, which is defined as a blunted cortisol response to intravenous ACTH administration. Twenty-one were randomized to receive 20 mg of intravenous Solumedrol and 22 were randomized to receive a matching placebo every 8 h for 7-days. There was no significant difference between the two randomized groups. Data analysis was carried out bya two-tailed test and P < .05 as significant. Results Results: The mean age was 51 ± 2 (mean ± SEM) with 61% female. Groups were well matched with regard to APACHE III score in Solumedrol versus placebo (59 ± 6 vs 59 ± 6), white blood cell count (18.8 ± 2.2 vs 18.6 ± 2.6), and incidence of bacteremia (29 vs 39%). The 28-day mortality rate was reduced in the Solumedrol treated arm (43 ± 11 vs 73 ± 10%; P < .05). There was no change in days in ICU, days on blood pressure agents, or days on ventilator. Seven days of high-dose intravenous Solumedrol treatment (20 mg every 8 h) in patients with a blunted cortisol response to ACTH was associated with an improved 28-day survival. This small study suggests that an inability to increase endogenous cortisol production in patients with sepsis who are then provided steroid treatment could improve survival.

One hundred-one crossbred beef heifers (average weight 256.1 kg) were used to evaluate the effects of diet during an 84-day period, on growth and reproductive performance. The four treatments were: 1) grass hay; 2) grass hay + 0.9 kg/day barley; 3) grass hay + 1.8 kg/day barley; and 4) alfalfa hay. Heifers were assigned randomly to 12 pens by weight (three pens/treatment). Prior to feeding, barley was cracked through a roller-mill. Heifers consuming alfalfa hay or grass hay + 1.8 kg barley gained 39% faster (0.68 and 0.74 kg/day, respectively; P = 0.01) than heifers consuming grass hay alone (0.51 kg/day). Heifers consuming grass hay + 0.9 kg barley were intermediate in average daily gain (ADG; 0.63 kg/day). Hay intake was 16% greater (P < 0.10) by heifers consuming alfalfa hay, grass hay, and grass hay + 0.9 kg barley compared with those consuming grass hay + 1.8 kg barley (average 8.1 vs 7.0 kg/day). Using costs (USD) of $0.066/kg, $0.099/kg, and $0.0935/kg for grass hay, alfalfa hay, and barley, respectively, feed cost/day was greatest (P < 0.01) for alfalfa hay ($0.81/day), intermediate for the grass hay + barley diets (average $0.63/day), and lowest for grass hay ($0.53/day). Cost/kg gain was less (P < 0.10) for grass hay + 1.8 kg barley compared with alfalfa hay. No diet differences (P > 0.70) were found in reproductive performance of the heifers. Feeding replacement heifers alfalfa hay resulted in performance equivalent to feeding grass hay + 1.8 kg barley, but the alfalfa hay diet had a higher cost.

Objective: Adrenocorticotropic hormone (ACTH)-dependent Cushing’s syndrome is biochemically characterized by increased plasma concentrations of ACTH inducing hypersecretion of cortisol. Somatostatin is known to inhibit ACTH secretion, and in vitro data have shown the inhibition of ACTH secretion by agonists activating sst2 and sst5 receptors. The present study aimed to determine the inhibitory effect of the multireceptor ligand SOM230, compared with the sst2-preferring agonist octreotide, on corticotropin-releasing hormone (CRH)-stimulated secretion of ACTH and corticosterone in rats. Methods: Secretion of ACTH and corticosterone was induced by i.v. application of CRH (0.5 μg/kg) in rats pretreated 1 h before by i.v. application of SOM230 (1, 3, or 10 μg/kg), octreotide (10 μg/kg) or NaCl 0.9%. Results: SOM230 (3 and 10 μg/kg) inhibited CRH-induced ACTH release by 45±3% and 51±2%, respectively, and corticosterone release by 43±5% and 27±16%, respectively. 10 μg/kg of octreotide tended to be less potent at inhibiting ACTH release (34±6% inhibition) and did not alter the secretion of corticosterone. Conclusion: SOM230 has a stronger inhibitory effect on ACTH and corticosterone secretion than octreotide in rats. This difference can be explained by its higher affinity to sst1, sst3 and especially sst5 receptors compared with octreotide.

Abstract Previous studies evaluating single and multiple GnRH injections at time of CIDR removal have reported decreased interval to estrus and increased expression of estrus, and increased conception rates, respectively. Thus, the objective of this study was to compare the effects of supplementing GnRH at CIDR removal on expression of estrus and conception rates in beef heifers following the 7-d CO-Synch + CIDR protocol. Heifers (n = 1,482) from 11 herds over two years were synchronized using the 7-d CO-Synch + CIDR fixed time AI (FTAI) protocol and were randomly assigned to receive one of three treatments at CIDR removal [0 µg GnRH (0); 5 µg GnRH (5); or 5 µg at CIDR removal and 5 µg 12 h later (5 + 5)]. Expression of estrus and conception rates were analyzed using the GLIMMIX procedure of SAS with treatment as an independent variable and herd as a random effect. There was no effect of treatment on expression of estrus (P = 0.38), or AI conception rates (P = 0.85). There was an effect of estrus (P < 0.01) and a treatment × estrus interaction (P = 0.02). Animals that exhibited estrus prior to FTAI had increased (P < 0.01) conception rates compared with those that did not. Animals that exhibited estrus had increased (P < 0.01) conception rates compared with non-estrus animals in the 0 and 5 + 5 µg groups (54 ± 3% vs 34 ± 3%, and 52 ± 3% vs 36 ± 3%, respectively), but in the 5 µg group, there was no difference (P = 0.53) in conception rates between estrus and non-estrus animals (44 ± 3% vs 41 ± 3%, respectively). In conclusion, estrus expression increased pregnancy success, but supplementation with 5 or 5 + 5 µg of GnRH at CIDR removal did not improve estrus response or conception rates in beef heifers.

ABSTRACT BAL8557 is the water-soluble prodrug of a novel antifungal triazole, BAL4815. BAL4815 is active against a broad spectrum of major opportunistic and pathogenic fungi, including strains that are resistant to other azoles. Cohorts of healthy male subjects received single-ascending oral (p.o.) doses of BAL8557 that were equivalent to 100, 200, or 400 mg of BAL4815 or single-ascending, 1-h constant-rate intravenous (i.v.) infusions of BAL8557 which were equivalent to 50, 100, or 200 mg of BAL4815. In each cohort, six subjects were randomly assigned to receive active drug and two subjects were assigned to receive the placebo. All doses were well tolerated, and no severe or serious adverse events occurred. Maximum plasma concentrations of BAL4815 were observed 1.5 to 3 h after p.o. drug intake or at the end of the 1-h infusion. After both routes of administration, values for maximum drug concentration observed in plasma and area under the concentration-time curve increased slightly more than proportionally to the administered dose. Mean elimination half-lives were particularly long (56 to 77 h after p.o. administration and 76 to 104 h after i.v. administration). The volume of distribution was large (155 to 292 liters after p.o. administration and 304 to 494 liters after i.v. administration) and systemic clearance was low (1.9 to 2.8 liter/h after p.o. administration and 2.8 to 5.0 liter/h after i.v. administration). Urinary recovery of BAL4815 was less than 0.4% of the infused dose. Based on the exposure data, oral bioavailability of BAL4815 is assumed to be very high. The pharmacokinetics of BAL4815 are well suited to maintaining concentrations of BAL4815 for a long period of time in the body and to enabling an effective treatment of systemic mycoses.

An oestradiol-based protocol (named J-Synch) has been developed recently with the aim of providing for a longer proestrus, enhancing the development of the dominant follicle before ovulation, and increasing pregnancy rates following fixed-time AI (FTAI) in heifers (de la Matta and Bo 2012 Taurus 55, 17–23). Two experiments were performed in Uruguay with 1953 Angus × Hereford crossbred heifers to compare pregnancy rates obtained with the J-Synch protocol with the conventional 7-day oestradiol-based protocol. A secondary objective was to determine the effects of the time of FTAI and the addition of eCG at the time progestin device removal on pregnancy rates. In Experiment 1, 966 heifers received a DIB device (0.5 g of progesterone, Syntex, Buenos Aires, Argentina) plus 2 mg of oestradiol benzoate IM (Syntex) on Day 0. Heifers in the conventional treatment group (n = 485) received cloprostenol (500 μg, Ciclase DL, Syntex) and oestradiol cypionate (0.5 mg, Cipiosyn, Syntex) IM and had their DIB removed on Day 7 a.m. Heifers in this group were then subdivided to be FTAI on Day 9 a.m. or p.m. (i.e. 48 or 56 h after DIB removal). Heifers in the J-Synch treatment group (n = 481) received cloprostenol IM and DIB removal on Day 6 p.m. and received gonadotropin-releasing hormone (100 μg gonadorelin acetate, Gonasyn Gdr, Syntex) on Day 9 a.m.; heifers were then FTAI on Day 9 a.m. or p.m. (i.e. 60 or 72 h after DIB removal). All heifers in this experiment were also treated with 300 IU of eCG (Novormon, Syntex) at DIB removal. In Experiment 2, 987 heifers were treated with the J-Synch protocol as described in Experiment 1. At device removal (Day 6 p.m.), heifers were divided to receive (n = 488) or not (n = 499) 300 IU of eCG IM at that time and were further subdivided to receive gonadotropin-releasing hormone and FTAI on Day 9 a.m. or p.m. (i.e. 60 or 72 h after DIB removal). Pregnancy rates were determined by ultrasonography 30 days after FTAI. Data were analysed using logistic regression, and results are shown in Table 1. J-Synch-treated heifers tended (P < 0.1) to have higher pregnancy rates following FTAI, whereas time of FTAI only affected pregnancy rates following the conventional 7-day treatment (P < 0.05). However, removal of eCG from the J-Synch protocol in Experiment 2 resulted in reduced pregnancy rates when inseminations were done on Day 9 p.m. (P < 0.05). In summary, the addition of eCG to the J-Synch protocol provided for a wider window of insemination times facilitating FTAI in large groups of beef heifers. Two experiments were designed to evaluate the effect of the timing of fixed-time AI (FTAI) in relation to the removal of an intravaginal progesterone-releasing device (1 g of progesterone, DIB, Syntex SA, Buenos Aires, Argentina) on pregnancy rates in Bos indicus × Bos taurus cross-bred heifers. In experiment 1, 285 Bonsmara × zebu cross-bred heifers, between 18 and 24 months of age and with a body condition score (BCS) between 3.0 and 3.5 (1-5 scale) were used. On the day of initiation of treatment (Day 0), the heifers’ ovaries were palpated (92% of them had a CL) and they received a new DIB plus 2 mg of estradiol benzoate (EB; Syntex SA) and 250 μg of cloprostenol (Ciclase DL, Syntex SA). On Day 8, DIB devices were removed and all heifers received 250 μg of Ciclase plus 0.5 mg of estradiol cypionate (ECP; Cipiosyn, Syntex SA). At that time the heifers were randomly divided to receive FTAI between 48 to 49 h, 53 to 54 h, or 58 to 59 h after DIB removal. The heifers underwent FTAI with semen from 4 bulls by 2 inseminators. In experiment 2, 260 heifers from the same group as those used in experiment 1 (87% with a CL) were treated exactly as those in experiment 1, except that previously used DIB was inserted on Day 0. Pregnancy diagnosis was performed 30 days post-fixed-time AI by ultrasonography. The data were analyzed by logistic regression, taking into account the effect of time of FTAI, semen, and inseminator on pregnancy rates. In experiment 1, pregnancy rates were lower (P = 0.04) in the heifers undergoing FTAI between 48 and 49 h after DIB removal (46/95, 48.4%) than those undergoing FTAI 53 to 54 h (61/99, 61.6%) or 58 to 60 h (57/91, 62.6%) after DIB removal. However, no differences in pregnancy rates were found (P = 0.72) in experiment 2 between the 3 treatment groups, with 39/91 (42.9%) for the 48 to 49 h group, 45/89 (50.6%) for the 53 to 54 h group, and 35/89 (43.8%) for the 58 to 59 h group. There was no effect of the semen or inseminator (P > 0.2) in either experiment. We conclude that when Bos indicus × Bos taurus beef heifers are synchronized with new DIB devices and ECP, higher pregnancy rates are obtained in heifers undergoing FTAI late (between 53 to 60 h after DIB removal) than in those undergoing FTAI early (48 to 49 h after DIB removal). However, time of insemination does not apparently affect pregnancy rates when Bos indicus × Bos taurus beef heifers are synchronized with previously used DIB devices and ECP.
