The use of fecal inoculums for in vitro fermentation models requires a viable gut microbiota, capable of fermenting the unabsorbed nutrients. Fresh samples from human donors are used; however, the availability of fresh fecal inoculum and its inherent variability is often a problem. This study aimed to optimize a method of preserving pooled human fecal samples for in vitro fermentation studies. Different conditions and times of storage at −20 °C were tested. In vitro fermentation experiments were carried out for both fresh and frozen inoculums, and the metabolic profile compared. In comparison with the fresh, the inoculum frozen in a PBS and 30% glycerol solution, had a significantly lower (p < 0.05) bacterial count (<1 log CFU/mL). However, no significant differences (p < 0.05) were found between the metabolic profiles after 48 h. Hence, a PBS and 30% glycerol solution can be used to maintain the gut microbiota viability during storage at −20 °C for at least 3 months, without interfering with the normal course of colonic fermentation.

A wide range of compounds has been described which have the potential to improve animal production by manipulating the rumen fermentation. Prominent among these rumen modifiers are the ionophores. Ionophores, such as monensin and tetronasin, improve feed efficiency, partly by increasing the flow of amino-N from the rumen and partly by stimulating the production of propionate in the rumen with an associated reduction in the production of methane (Russell and Strobel, 1988). Recently there has been increasing interest in the use of yeast culture (YC) and other fungal preparation to modify the rumen fermentation. These products have been shown to increase bacterial numbers within the rumen with an associated increase in the breakdown of fibre and supply of microbial protein (Williams and Newbold, 1990). YC has also been reported to increase the production of propionate in the rumen. Little appears to be known about the effect a combination YC and an ionophore would have on the rumen fermentation. This study describes the effects of the ionophores monensin and tetronasin on the fermentation of hay by rumen fluid from sheep fed a basal diet with or without YC.

The effect of extruding white lupin (Lupinus albus 'Lublanc') seeds (WLS) at 110, 130, 150 or 180 °C on the in vitro solubility of crude protein (CP) and in sacco rumen degradation and intestinal digestion of rumen escape CP and fiber was determined. Rumen degradation was estimated by incubating nylon bags in the rumen of cows for 2, 4, 8, 16, 24 and 48 h. Extruding WLS at 110, 130, 150 and 180 °C reduced the CP-solubility by 32.8, 47.7, 58.4 and 67.5%, respectively. The effective ruminal degradabilities of CP, acid detergent fiber (ADF) and neutral detergent fiber (NDF) were evaluated assuming a ruminal outflow rate of 0.06 h−1. Heating WLS at 110, 130, 150 and 180 °C decreased the ruminal degradability of CP value: 89.8, 79.9, 65.1, 61.8 vs. 93.4% (raw), respectively; the corresponding values for ADF and NDF were: 38.4, 35.3, 34.2, 27.6 vs. 43.6% (raw) and 37.7, 33.1, 32.2, 26.5 vs. 39.4% (raw). Postruminal digestion was estimated using a sequence of ruminal in situ incubation for 4, 8 and 16 h, in vitro incubation in an acid-pepsin bath for 3 h and a mobile nylon bag technique distal to the abomasum. Extrusion of WLS decreased the degradability of CP, ADF and NDF in the rumen with a corresponding increase in the amounts digested in the postruminal sections; the whole-tract digestibility was generally unchanged. Therefore, the processing shifted the digestion of these components from rumen to the lower gastrointestinal tract. ABSTRACT Choline is a water-soluble nutrient essential for human life. Gut microbial metabolism of choline results in the production of trimethylamine (TMA), which upon absorption by the host is converted in the liver to trimethylamine-N-oxide (TMAO). Recent studies revealed that TMAO exacerbates atherosclerosis in mice and positively correlates with the severity of this disease in humans. However, which microbes contribute to TMA production in the human gut, the extent to which host factors (e.g., genotype) and diet affect TMA production and colonization of these microbes, and the effects TMA-producing microbes have on the bioavailability of dietary choline remain largely unknown. We screened a collection of 79 sequenced human intestinal isolates encompassing the major phyla found in the human gut and identified nine strains capable of producing TMA from choline in vitro. Gnotobiotic mouse studies showed that TMAO accumulates in the serum of animals colonized with TMA-producing species, but not in the serum of animals colonized with intestinal isolates that do not generate TMA from choline in vitro. Remarkably, low levels of colonization by TMA-producing bacteria significantly reduced choline levels available to the host. This effect was more pronounced as the abundance of TMA-producing bacteria increased. Our findings provide a framework for designing strategies aimed at changing the representation or activity of TMA-producing bacteria in the human gut and suggest that the TMA-producing status of the gut microbiota should be considered when making recommendations about choline intake requirements for humans. IMPORTANCE Cardiovascular disease (CVD) is the leading cause of death and disability worldwide, and increased trimethylamine N-oxide (TMAO) levels have been causally linked with CVD development. This work identifies members of the human gut microbiota responsible for both the accumulation of trimethylamine (TMA), the precursor of the proatherogenic compound TMAO, and subsequent decreased choline bioavailability to the host. Understanding how to manipulate the representation and function of choline-consuming, TMA-producing species in the intestinal microbiota could potentially lead to novel means for preventing or treating atherosclerosis and choline deficiency-associated diseases.
