Exogenously supplied sucrose improves the postharvest quality of Grevillea ‘Sylvia’ inflorescences

Summary. Short vase life (e.g. 3–7 days) is a problem with inflorescences of Grevillea used as cut flowers and may be associated with depletion of respirable substrates. A series of 4 experiments addressing this hypothesis were conducted with Grevillea hybrid ‘Sylvia’ inflorescences. In experiment 1, the effect of leaf retention on inflorescence stems and a range of sucrose concentrations (0–6% w/v) in vase solutions was investigated. In experiment 2, the relationship between inflorescence maturity (4 stages) and sucrose supply was studied. In experiment 3, the effect of sucrose concentration as a short period pulse and pulsing duration was determined. In experiment 4, simulated handling and transport was undertaken to gauge the practical usefulness of sucrose pulsing to the cut flower industry. These vase life experiments were conducted in a constant temperature room (22 ± 2°C) at 50–80% relative humidity. All vase solutions contained 10 mg chlorine/L to suppress microbial growth. Three percent sucrose (w/v) was found in experiment 1 to be the best vase solution. This treatment gave a mean vase life of 10.1 ± 0.2 days, which was 44% longer than the control (no sucrose) treatment. Inflorescences picked at the ‘early looping’ stage and provided with 3% (w/v) sucrose had the best overall vase life (12.0 ± 0 days) and visual appeal in experiment 2. Sucrose pulsing (10% w/v) for 6–24 h extended vase life by at least 1 day. The presence of leaves shortened the vase life of both sucrose-pulsed and non-pulsed inflorescences by about 1 day in experiment 3. Storage for 5 days at 5°C as handling/transport simulation in experiment 4 did not shorten the vase life of Grevillea ‘Sylvia’ inflorescences. Sucrose pulsing (10% w/v) before simulated handling/transport did not result in a significant increase in the vase life of Grevillea ‘Sylvia’ inflorescences, and is therefore not recommended. To achieve the best floral display, Grevillea ‘Sylvia’ inflorescences should be picked at the ‘early looping’ stage of maturity and then placed in a vase solution containing 3% (w/v) sucrose.

The plant growth regulator abscisic acid induces stomatal closure, which can reduce transpiration rate and extend vase life of cut flowers by maintaining a positive water balance. Analytical-reagent grade abscisic acid is, however, expensive. In this work, the effects of 2 potentially inexpensive synthetic analogues of abscisic acid were studied as to their effects on cut Baccara roses. Abscisic acid and its analogues PBI-365 and PBI-429 reduced transpiration and helped to increase fresh weight during the first few days of vase life. However, the reduction in transpiration rate brought about by abscisic acid and the analogues did not alter vase life. Sucrose in the vase solution at concentrations of 2 and 4% increased vase life of cut Baccara roses. However, the highest sucrose concentration (4%) caused leaf crisping, thereby reducing the quality of the flower stems. Abscisic acid and the analogue PBI-365 reduced this phytotoxicity by induction of stomatal closure and reduction of vase solution sucrose flux into the leaves.

The effect of STS and 1-MCP on the postharvest quality of carnation and rose cut flowers was studied. Cut flowers of Dianthus c..aryophyllus L. cv. Asso and Rosa hybritia cv. Baroness were treated with silver thiosulfate (STS) at 0.4 mM with sucrose at 50 g 1-t and 1-methylcyclopropene ( I -MCP) at 0.5 g m-3 for 611. Pretreatment with STS and 1-MCP significantly extended the vase life and minimized the % loss of initial weight of carnation and rose cut flowers comparing to the untreated control. The two chemicals applied inhibited the chlorophyll degradation and carbohydrate loss and hence, significantly improved the postharvest quality of carnation and rose cut flowers comparing to the control. Ethylene production by cut flowers was inhibited as a result of using these chemicals. In general, there were no differences between STS and (-MCP but the later does not have the heavy metal implications of STS treatment, and hence, using 1-MCP pretreatment for extending the vase life of carnation and rose cut flowers was recommended.

Low root zone temperatures in summer stimulate generative development of Alstroemeria, resulting in a higher flower production in winter. The effects of greenhouse soil and air temperatures on vase life and ornamental value were evaluated. Preharvest treatments were two locations (field stations), four air temperatures (9, 12, 15, 18C), four root zone temperatures (11, 14, 17C, uncontrolled) and three varieties ('Flamengo', 'Jubilee', 'Wilhelmina') in a factorial design. The flowers were placed in a commercial pretreatment solution for 24 hours immediately after harvest. After a two day transport simulation and rehydration for 3 hours at 5C, flowers were kept at 1.5 W.m-2 PAR (12hr/day), 20C and 60% RH. The experiment was done three times. Results from the first harvest showed that lowering the soil temperature increased the number of stems that had two whirls of flowers opening after harvest. Average vase life was two weeks. Low air temperatures increased whirl opening as well, and increased vase life by one or two days. Ornamental value and number of flower branches per stem were not affected.

Lily (Lilium L. hybrids) is sub-tropical cut flower that can be grown in open place or in green house and has consumer demand in local as well as in offshore markets. Short vase life, postharvest decay of flowering petals, activities of oxidative enzymes, malondialdehyde contents, electrolyte leakage and low water ratio are the main problems in marketing of cut lilies. This study was carried out to investigate the effect of oxalic acid (OA) treatments on vase life (12 days) and postharvest quality of ‘Mero Star’ cut lily flowers. The flowering stems were placed in 2, 4 and 6% aqueous solution of OA for 12 days at 20±2 °C with 70±5% relative humidity. Flowers placed in 6% OA solution exhibited longest vase life and flower diameter with higher relative water content and increased fresh weight and reduced flower decay. Reduced malondialdehyde contents and electrolyte leakage with higher activities of catalase, superoxide dismutase and peroxidase enzymes were also noted in 6% OA-treated flowers. In conclusion, exogenous postharvest application of 6% OA extended vase life and maintained postharvest quality of cut flowers by reducing electrolyte leakage and malondialdehyde contents with higher antioxidative enzymes activities.

Abstract Postharvest senescence is one of the crucial challenges limiting the marketability of cut flowers. Pertinently, recent investigations implicate extensive role of polyamines in regulation of flower senescence. The present study was envisaged to test the efficacy of poylamines in preserving the postharvest quality of Consolida ajacis (C. ajacis) cut spikes. The cut spikes of C. ajacis were subjected to various treatments of polyamines viz, 4 mM Spermine (SPM), 6mM Putrescine (PUT) and 6 mM Spermidine (SPD). A separate set of spikes held in distilled water represented the control. Our results authenticate a significant improvement in vase life of cut spikes of C. ajacis as compared to control. The increment in vase life was commensurate with the higher concentration of sugars, proteins and phenols in the tepal tissues. Polyamines amplified the activity of various antioxidant enzymes viz, superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) to overcome the deleterious effects of reactive oxygen species (ROS). The membrane outflow of tepal tissues was profoundly reduced due to attenuated lipoxygenase (LOX) activity. These findings reveal conspicuous role of polyamines particularly SPM in modulation of flower senescence in cut spikes of C. ajacis.

Abstract Quality of cut flowers is an important issue at postharvest as well as an important factor contributing to marketing of and profitability from the tuberose. In this study, the effects of citric acid (CA) and hydrogen peroxide (H2O2) added to the vase water on postharvest quality of tuberose cut flowers were investigated. CA was applied in concentrations of 50, 100, 200, 400 mg·dm−3 and H2O2 in concentrations of 10, 20, 40 and 80 mg·dm−3 and distilled water as control treatment. Results showed that both compounds had significant positive effects on solution uptake, wilting and abscission of florets, relative water content, chlorophyll content, and vase life duration. The effects of 100 and 200 mg·dm−3 of CA and 20 and 40 mg·dm−3 of H2O2 proved to be more effective than other treatments. Both compounds increased the vase life of tuberose cut flowers and CA at concentrations 100 and 200 mg dm−3 and H2O2 at concentrations 20 and 40 mg dm−3 doubled this time up to 14-17 days.

Abstract Rose is an ornamental plant, which constitutes one of the leading cut flowers in the world. The vase life of cut Rosa hybrida L. flowers is truly short. We examined the effect of various temperatures and different time duration factors on postharvest longevity, and some other physic-biochemical characteristics and antioxidants activity. Fresh cut Rosa hybrida L. flowers were treated by water with various temperatures (23º, 42º, 47º, and 52°C) and different time duration (5, 10, 15, and 20 min). Maximum vase life (7.52 days) was related to cut flowers that were treated with 52°C for 15 min. Minimum vase life (4.46 days) was observed in cut flowers that were treated with 42°C for 5 min. Treatment of cut flowers by water with the temperature of 52°C for 15 min induced the highest water absorption, petal’s anthocyanin, and leaf’s chlorophyll. The lowest ethylene production was also obtained in cut flowers treated with 52°C for 15 min. The use of warm water (52°C) for 15 min clearly extended the vase life and most traits pertaining to the vase life of cut Rosa hybrida L. flowers.

The use of methyl jasmonate has demonstrated its efficiency to extend the vase life of cut flowers. The objective of this study was to evaluate the effect of methyl jasmonate associated with storage at low temperatures on the postharvest quality of Rosa cv. Avalanche stems. The treatments consisted of 125, 250, 500 and 1000 μM of methyl jasmonate, besides the control with distilled water. The flower buds were sprayed with 4 mL of the solution, according to the treatments, and then kept in a cold chamber (1 °C) for periods of 2 and 6 days. Subsequently, the stems were taken to the postharvest laboratory at a temperature of 16 °C. Better quality, higher fresh weight and water absorption were observed in flower stems stored for 2 days. The application of methyl jasmonate caused less turgescence and greater darkening of roses. It was possible to conclude that two days is the best storage time at 1 °C and the use of methyl jasmonate does not maintain the quality of roses cv. Avalanche after harvest.

Abstract Occlusion of the stem vasculature by microorganisms that proliferate in the vase water, or the plant vessels, leads to water stress symptoms that reduce postharvest quality of cut flowers. This study aimed to determine the effects of pulsing and wet-cold storage on the microbial profiles in cut Gladiolus grandiflorus L. cv. Fado. Pulsing treatments of 600-ppm 8-hydroxyquinoline sulfate plus 5% sucrose solution versus distilled water were administered before wet cold storage periods of 0-5 days in cut Gladiolus, previously grown from corms under open field. A two-by-six factorial experiment embedded in a completely randomized design with four replicates was accomplished. Proc GLM in two-way Anova was adopted, and the means were separated using Tukey’s test at a 5% level of significance. The pulsing treatment of 600 ppm 8-HQS plus 5% sucrose, the wet cold storage duration and their interactive effects significantly (P˂0.0209; ˂0.0001 and ˂0.0001 respectively) affected the means of the colony-forming units in the vase water of cut Gladiolus at senescence. The prolonged vase life of cut gladioli spikes was associated with decreased microbial proliferation as influenced by pulsing and wet storage duration of up to 4 days. Data generated from this study will improve existing technologies related to the quality and market value of this Gladiolus cultivar.

Ethylene is the most important factor playing roles in senescence and deterioration of harvested crops including cut flowers. Brassinosteroids (BRs), as natural phytohormones, have been reported to differently modulate ethylene production and related senescence processes in different crops. This study was carried out to determine the effects of different levels of 24-epibrassinolide (EBL) on ACC oxidase enzyme activity, the final enzyme in ethylene biosynthesis pathway, vase life, and senescence rate in lisianthus cut flowers. Harvested flowers were treated with EBL (at 0, 3, 6, and 9 µmol/L) and kept at 25 °C for 15 days. The ACC oxidase activity, water absorption, malondialdehyde (MDA) production and vase solution absorption rates, chlorophyll and anthocyanin contents, and the vase life of the flowers were evaluated during and at the end of storage. EBL at 3 µmol/L significantly (p ≤ 0.01) enhanced the flower vase life by decreasing the ACC oxidase activity, MDA production and senescence rates, and enhancing chlorophyll and anthocyanin biosynthesis and accumulation, relative water content, and vase solution absorption rates. By increasing the concentration, EBL negatively affected the flower vase life and postharvest quality probably via enhancing the ACC oxidase enzyme activity and subsequent ethylene production. EBL at 6 and 9 µmol/L and in a concentration dependent manner, enhanced the ACC oxidase activity and MDA production rate and decreased chlorophyll and anthocyanin accumulation and water absorption rate. The results indicate that the effects of brassinosteroids on ethylene production and physiology of lisianthus cut flowers is highly dose dependent.
