Strategies for control of Phoma black stem in annual Medicago species

Strategies for control of Phoma black stem disease in annual Medicago species through selection of cultivars with increased resistance to Phoma medicaginis, fungicidal spray applications to swards, and fungicidal control of seedborne infection, were investigated. Fiftyseven annual Medicago cultivars and lines were screened for resistance in the field in 1 m rows over 2 consecutive seasons. There were significant differences in resistance among species and also between lines and cultivars of any particular species. Three M. rugosa cultivars were very highly resistant and most cultivars and lines showed some resistance. In a field trial, the fungicides benomyl, carbendazim, flutriafol, propiconazole, thiabendazole and triadimefon were tested for their efficacy in controlling Phoma black stem disease. All fungicides reduced disease severity in the sward and, except for thiabendazole, the percentage burrs with Phoma lesions. The role and control of seed-borne P. medicaginis in causing Phoma black stem disease in Medicago truncatula and M. polymorpha var. brevispina at a field site was also investigated. Seed-borne P. medicaginis caused subsequent development of Phoma black stem disease in swards sown with infected seed. Disease appeared earlier, developed faster and became much more severe in M. truncatula cv. Cyprus than in M. polymorpha cv. Serena. Application of benomyl seed treatments (0.1 and 0.5% w/w) resulted in only a 4-5 week delay in the onset of Phoma black stem symptoms.

An improved technique for successful artificial hybridization in annual medic (Medicago spp.) is described. Using a previously reported method, only four out of seven species were successfully crossed, with the percentage of success ranging from 3 to 22%. Initial modifications to this technique gave a 7-8 fold increase in the successful crossing rate in M. murex and M. polymorpha medic, from 9 to 64% with M. murex and from 10 to 82% with M. polymorpha. Further modifications to the technique resulted in a success rate of 100% in both species. The numbers of seeds per pod obtained from crosses in both species were also increased by using the modified techniques compared to the established method. Selection of larger, more mature flowers, differences in flower cutting position, as well as post pollination position were the main modifications which accounted for the greatly improved success rate. The modified technique was subsequently applied successfully to obtain for the first time inter-specific crosses involving M. polymorphax M. murex, M. polymorphax M. sphaerocarpos, M.murexx M. sphaerocarpos, M. solerolii x M. littoralis/M.truncatula hybrid, M. solerolii x M, tornata, and M. littoralis/M.truncatula hybrid x M. sphaerocarpos.

On a moderately acid soil (pH 4.6 in 115 suspension of soil in 0.01M calcium chloride), nodulation of Medicago truncatula cv. Cyprus responded markedly to increasing applications of calcium carbonate, which increased soil pH. Since the effect of increasing soil pH on the percentage nodulation could be replaced to a large extent by increasing the inoculation level, it appeared that nodulation was restricted by the inability of Rhizobium meliloti to survive or multiply in the acid soil. The growth of R, meliloti appeared more sensitive to soil acidity than growth of the host plant of annual Medicago species. It is suggested that more acid-tolerant strains of R. meliloti would permit annual Medicago species to be grown successfully on moderately acid soils, thus extending the range of soils suitable for the growth of these species.

Phoma black stem and leaf spot disease of annual Medicago spp., caused by Phoma medicaginis, not only can devastate forage and seed yield, but also reduces herbage quality by inducing production of phytoestrogens, particularly coumestrol and 4'-O-methylcoumestrol, that can reduce ovulation rates of animals grazing infected forage. We determined the consequent phytoestrogen levels on three different annual Medicago species/cultivars (M. truncatula cv. Cyprus, M. polymorpha var. brevispina cv. Serena and M. murex cv. Zodiac), following inoculation with 35 isolates of P. medicaginis. Across the isolate x cultivar combinations, leaf disease incidence (%LDI), petiole disease incidence (%PDI), leaf disease severity (%LDS), petiole disease severity (%PDS), and leaf yellowing severity (%LYS) ranged up to 100, 89.4, 100, 58.1 and 61.2%, respectively. Cultivars Cyprus and Serena were most susceptible and cv. Zodiac the most resistant to P. medicaginis. Isolates WAC3653, WAC3658 and WAC4252 produced the most severe disease. Levels of phytoestrogens in stems ranged from 25 to 1995 mg/kg for coumestrol and from 0 to 418 mg/kg for 4'-O-methylcoumestrol. There was a significant positive relationship of disease incidence and severity parameters with both coumestrol and 4'-O-methylcoumestrol contents, as noted across individual cultivars, and as across the three cultivars overall where r=0.39 and 0.37 for coumestrol and 4'-O-methylcoumestrol, respectively (P<0.05). Although, cv. Serena was most susceptible to P. medicaginis and produced the highest levels of phytoestrogens in the presence of P. medicaginis, cv. Zodiac contained the highest levels of phytoestrogens in comparison with other cultivars in the absence of P. medicaginis. There was a 15-fold increase in coumestrol in cv. Serena but only a 7-fold increase in cv. Zodiac from infection of P. medicaginis. The study highlights that the intrinsic ability of a particular cultivar to produce phytoestrogens in the absence of the pathogen, and its comparative ability to produce phytoestrogens in the presence of the P. medicaginis, are both important and highly relevant to developing new annual Medicago spp. cultivars that offer improved disease resistance and better animal reproductive outcomes.

Six strains of Rhizobium meliloti varied greatly in their ability to colonize two moderately acid sandy soils (pH 5.0-5.1 in 115 suspension of soil in 0.01M calcium chloride) in the year of sowing inoculated lime-coated seeds of Medicago truncatula. The strains also varied in their ability to nodulate M. truncatula in the year after sowing. With all rhizobial strains and on both soils, calcium carbonate applied in the year of sowing strongly promoted colonization in the year of sowing. It also greatly increased nodulation and growth of M. truncatula in the year after sowing. Apparently calcium carbonate increased second year nodulation by promoting the colonization of the acid soils by R. meliloti in the year of sowing. Differences among strains in second year nodulation appeared to be related to differences in the ability of the strains to colonize the soils in the year of sowing. Strains isolated from moderately acid sandy soils were markedly superior in both properties, and on both soils, to strain SU47 from commercial inoculum, but were generally no better than strain U45 from commercial inoculum. Even the most acid-tolerant strains colonized the acid soils relatively slowly. It is suggested that R. meliloti strains in general have a poor ability to colonize acid soils and that this property is a major factor limiting the growth and persistence of annual Medicago species on acid soils in the field.

The article presents the results of mathematical experiments with the system «Social potential of childhood in the Russian regions». In the structure of system divided into three subsystems – the «Reproduction of children in the region», «Children’s health» and «Education of children», for each defined its target factor (output parameter). The groups of infrastructure factors (education, health, culture and sport, transport), socio-economic, territorial-settlement, demographic and en-vironmental factors are designated as the factors that control the system (input parameters). The aim of the study is to build a model îf «Social potential of childhood in the Russian regions», as well as to conduct experiments to find the optimal ratio of the values of target and control factors. Three waves of experiments were conducted. The first wave is related to the analysis of the dynam-ics of indicators for 6 years. The second – with the selection of optimal values of control factors at fixed ideal values of target factors. The third wave allowed us to calculate the values of the target factors based on the selected optimal values of the control factors of the previous wave.

We introduce a method that permits to co-evolve the body and the control properties of robots. It can be used to adapt the morphological traits of robots with a hand-designed morphological bauplan or to evolve the morphological bauplan as well. Our results indicate that robots with co-adapted body and control traits outperform robots with fixed hand-designed morphologies. Interestingly, the advantage is not due to the selection of better morphologies but rather to the mutual scaffolding process that results from the possibility to co-adapt the morphological traits to the control traits and vice versa. Our results also demonstrate that morphological variations do not necessarily have destructive effects on robots’ skills.

The Covid-19 restrictions have limited the access of face-to face therapies for many people and continues to effect how Dramatherapists operate. The following article offers reflections around adapting to an on-line medium, focusing more specifically around the software of ProReal. Limitations and considerations are acknowledged, including technological difficulties, computer efficacy, ambiguity tolerance and the need for careful contracting and reassurance of autonomy and control when using on-line platforms. The article ends with a short selection of vignettes from ProReal sessions.

Since application of integrated energy systems (IESs) has formed a markedly increasing trend recently, selecting an appropriate integrated energy system construction scheme becomes essential to the energy supplier. This paper aims to develop a multi-criteria decision-making model for the evaluation and selection of an IES construction scheme equipped with smart energy management and control platform. Firstly, a comprehensive evaluation criteria system including economy, energy, environment, technology and service is established. The evaluation criteria system is divided into quantitative criteria denoted by interval numbers and qualitative criteria. Secondly, single-valued neutrosophic numbers are adopted to denote the qualitative criteria in the evaluation criteria system. Thirdly, in order to accommodate mixed data types consisting of both interval numbers and single-valued neutrosophic numbers, the TOPSIS (Technique for Order Preference by Similarity to an Ideal Solution) method is extended into a three-stage technique by introducing a fusion coefficient μ. Then, a real case in China is evaluated through applying the proposed method. Furthermore, a comprehensive discussion is made to analyze the evaluation result and verify the reliability and stability of the method. In short, this study provides a useful tool for the energy supplier to evaluate and select a preferred IES construction scheme.

Because the behavior of the condition number can have highly steep and multi-modal structure, optimal control and monitoring problems based on the condition number cannot be easily solved. In this paper, a minimization problem is formulated for κ2(P), the condition number of an eigensystem (P) of a matrix in terms of the L2 norm. A new non-normality measure is shown to exist that guarantees small values for the condition number. In addition, this measure can be minimized by proper selection of controller and observer gains. Application to the design of well-conditioned controller and observer-based monitors is illustrated.

AbstractTo understand the mechanisms that mediate germline genetic leukemia predisposition, we studied the inherited ribosomopathy Shwachman-Diamond syndrome (SDS), a bone marrow failure disorder with high risk of myeloid malignancies at an early age. To define the mechanistic basis of clonal hematopoiesis in SDS, we investigate somatic mutations acquired by patients with SDS followed longitudinally. Here we report that multiple independent somatic hematopoietic clones arise early in life, most commonly harboring heterozygous mutations in EIF6 or TP53. We show that germline SBDS deficiency establishes a fitness constraint that drives selection of somatic clones via two distinct mechanisms with different clinical consequences. EIF6 inactivation mediates a compensatory pathway with limited leukemic potential by ameliorating the underlying SDS ribosome defect and enhancing clone fitness. TP53 mutations define a maladaptive pathway with enhanced leukemic potential by inactivating tumor suppressor checkpoints without correcting the ribosome defect. Subsequent development of leukemia was associated with acquisition of biallelic TP53 alterations. These results mechanistically link leukemia predisposition to germline genetic constraints on cellular fitness, and provide a rational framework for clinical surveillance strategies.
