The use of assisted reproductive techniques for endangered species is a major goal for conservation. One of these techniques, testis tissue xenografting, allows for the development of spermatozoa from animals that die before reaching sexual maturity. To assess the potential use of this technique with endangered species, testis tissue from six Iberian lynxes (one fetus, two perinatal cubs, two 6-month-old and one 2-year-old lynx), two Cuvier’s gazelle fetuses and one 8-month-old Mohor gazelle were transplanted ectopically into nude mice. Tissue from the lynx fetus, perinatal cubs and 2-year-old donors degenerated, whereas spermatogonia were present in 15% of seminiferous tubules more than 70 weeks after grafting in transplanted testis tissue from 6-month-old donors. Seminal vesicle weights (indicative of testosterone production) increased over time in mice transplanted with tissue from 6-month-old lynxes. Progression of spermatogenesis was observed in xenografts from gazelles and was donor age dependent. Tissue from Cuvier’s gazelle fetuses contained spermatocytes 40 weeks after grafting. Finally, round spermatids were found 28 weeks after transplantation in grafts from the 8-month-old Mohor gazelle. This is the first time that xenotransplantation of testicular tissue has been performed with an endangered felid and the first successful xenotransplantation in an endangered species. Our results open important options for the preservation of biological diversity.

SUMMARY Concentrations of oestradiol-17β and testosterone were estimated in testicular tissue from intact and hypophysectomized rats. Within 30 min after intravenous injection of human chorionic gonadotrophin (HCG) or follicle-stimulating hormone (FSH) to intact animals the tissue concentrations of both steroids were not significantly changed. Prolonged s.c. administration of HCG (5 days) caused an increase in the tissue levels of both steroids, which was further increased when the prolonged treatment was followed by an intravenous injection with this trophic hormone. FSH had no influence on tissue concentrations of oestradiol-17β or testosterone in hypophysectomized rats. Assay of separated seminiferous tubules and interstitial tissue indicated that oestradiol-17β and testosterone were mainly localized in the interstitial tissue. Incubations of these constituents showed that oestradiol-17β was produced in the seminiferous tubules, while testosterone was produced in the interstitial compartment.

The aim of this study was to clarify the role of the protein kinase suppressor of Ras1 (KSR1) in spermatogenesis. Spermatogenesis in ksr1−/− mice was studied in testicular tissue and epididymal spermatozoa by light and transmission electron microscopy and by immunofluorescence using antibodies to ghrelin and 3β-hydroxysteroid dehydrogenase (3β-HSD). Blood testosterone levels were also assessed. ksr1−/− mice showed reduced epididymal sperm concentration and motility as compared with wild-type (wt) mice. Testis tissue from ksr1−/− mice revealed a prevalent spermatogenetic arrest at the spermatocyte stage; the interstitial tissue was hypertrophic and the cytoplasm of the Leydig cells was full of lipid droplets. Ghrelin signal was present in the seminiferous tubules and, particularly, in the interstitial tissue of wt mice; however, in ksr1−/− mice ghrelin expression was very weak in both the interstitial tissue and tubules. On the contrary, the signal of 3β-HSD was weak in the interstitial tissue of wt and strong in ksr1−/− mice. Testosterone levels were significantly increased in the blood of ksr1−/− mice (P<0.05) as compared with wt. The results obtained reveal the importance of the KSR scaffold proteins in the spermatogenetic process. The study of the molecular mechanisms associated with spermatogenetic defects in a mouse model is essential to understand the factors involved in human spermatogenesis.

AbstractTesticular tissue was studied in domesticated yak and in wild yak x domesticated yak (F1 and F2) bulls at 6, 12, 18 and 24 months of age by stereology for quantitative histology and by comparative studies on the lactate dehydrogenase (LDH) isoenzyme spectrum, LDH activity and the percentage content of LDH isoenzymes in testes. The results indicated that all three types of yak were similar in both the characteristics of ultrastructure of testicular tissue and the degree of germ cell development. The process of spermatоgenesis was initiated and sperm were produced for the first time at the age of 12 months in all three yak types. All yak types exhibited similar age-related increases in weight of testes, volume density of both the seminiferous tubules and seminiferous epithelium and height of seminiferous epithelium but all these indexes were slightly affected by season. The number of LDH isoenzyme bands after electrophoresis varied with age as follows: four bands were present at 6 months of age, five bands at 12 and 18 months and six bands at 24 months of age. The sixth band was LDH-x, which appeared between the LDH4 and LDH5 band. These results indicated that yak bulls reach sexual maturity at 24 months of age and that puberty is not affected by infusion of wild strains.

Spermatogenesis from many mammalian species occurs in fragments of normal testis tissue xenografted to mice. Here we apply xenografting to the study of testicular pathology. Using the horse model, we investigated whether exposure to a permissive extratesticular environment in the mouse host would rescue spermatogenesis in cryptorchid testicular tissue or in tissue affected by idiopathic testicular degeneration (ITD). In cryptorchid tissue, where the extratesticular environment is abnormal, xenografting induced spermatogenesis up to meiosis in a subpopulation of seminiferous tubules. Thus, spermatogonia survive and partially retain their potential to differentiate in cryptorchid horse testes. In contrast, the primary defect in equine ITD is hypothesised to be tissue autologous. In support of this, xenografting did not restore spermatogenesis to tissue affected by ITD, thus confirming that the testis itself is primarily diseased. This outcome was not affected by supplementation of exogenous gonadotropins to the mouse host or by reconstitution of a normal reproductive regulatory axis supplied by functional porcine testicular xenografts. These studies demonstrate the usefulness of xenografting for the study of testicular pathology.

The Iberian lynx is an endangered felid that has been subject to an intensive, ongoing conservation program to save it from extinction. Identification of dental pathologies could play an important role in the survival of this endangered species. The prevalence of periodontal bone loss, tooth resorption, and oral neoplasia was evaluated in this species. Skulls of 88 adult specimens of the Iberian lynx ( Lynx pardinus) from the Doñana Biological Station (EBD-CSIC), Seville, Spain, were examined macroscopically and full-mouth dental radiographs of all specimens were performed. Different stages of periodontal disease were identified in 81.3% of teeth. Approximately two-thirds (64.8%) of the specimens examined had at least 1 tooth affected by stage 3 or 4 periodontal disease. Maxillary and mandibular incisor teeth accounted for a majority (59.4%) of teeth affected by stage 3 or 4 periodontal disease. Tooth resorption affected 12 teeth (0.63% of evaluated teeth) in 6 skull specimens (6.8% of skulls evaluated). Five of the teeth with resorptive lesions were detected on 1 skull specimen. Six teeth (50%) of those affected by tooth resorption were incisor teeth. Only 1 (1.1%) adult specimen showed signs of oral neoplasia. Our study concluded that there is a high prevalence of periodontal disease that could lead to impaired hunting ability, as well as may represent a threat to overall health and subsequent survival of this endangered species. The extremely low prevalence of tooth resorption in L pardinus could open new pathways in the discovery on the pathogenesis of tooth resorption in felids.

Retrieval, extracorporal storage and autotransplantation of testicular tissue could become an important strategy for preserving male gonadal function. The present study used syngeneic and immunodeficient nude mice as hosts, and immature and adult mice, neonatal and adult photoregressed Djungarian hamsters and neonatal marmosets to identify the potential of testicular tissue grafting to maintain the morphological and functional integrity of the testis. Testicular tissue was grafted s.c. either as fresh tissue or after cryopreservation into adult, orchidectomized hosts. The mice that received rodent testis tissue were autopsied 50 days later, and blood samples were collected. Sixty-five per cent of mouse isografts contained morphologically normal testicular tissue and seminiferous tubules with some degree of spermatogenic recovery. Mature spermatozoa were recovered after enzymatic disaggregation. Although the recovery of spermatogenesis was limited in adult mouse and hamster tissue, complete spermatogenesis was observed in grafts from immature rodents. Testicular tissue from neonatal marmosets developed up to the stage of spermatocytes at day 135 after xenografting. Androgen concentrations were comparable in intact control mice and in mice receiving fresh mouse and hamster grafts, slightly lower in mice receiving cryopreserved grafts and adult photoregressed hamster tissue, and low in castrated control mice and in mice receiving marmoset tissue. These results show that isografts and xenografts of immature and adult testicular tissue become functionally active as a s.c. graft in the mouse and that this approach might be useful in combination with cryopreservation as a tool for storage and activation of the male germ line and androgen replacement therapy in patients.

The Iberian lynx is an endangered felid that has been subject to an intensive ongoing conservation program in an attempt to save it from extinction. Identification of dental pathologies could play an important role in the survival of this endangered species. The objective of this study is to evaluate the dental pathologies (congenital, developmental, and traumatic abnormalities) of this species of felid. Skulls of 88 adult specimens of the Iberian lynx ( Lynx pardinus) from the Doñana Biological Station [Estación Biológica de Doñana-Consejo Superior de Investigaciones Científicas] (EBD-CSIC), Seville, Spain, were examined macroscopically, and full-mouth dental radiographs of all specimens were performed. Presence, absence, form of teeth, number of roots, supernumerary teeth, and persistent deciduous teeth were evaluated. The presence of attrition/abrasion, tooth fractures, enamel hypoplasia/hypomineralization, endodontic disease, intrinsically stained teeth, and other traumatic findings were also evaluated. Abnormal grooves and dilacerations were the most common abnormalities seen in the roots of teeth. Two hundred and fifteen fractured teeth (11.3% of evaluated teeth) were detected, and mainly complicated fractures were noted in maxillary canine teeth (24.2% of fractured teeth) and mandibular canine teeth (16.7% of fractured teeth). Endodontic disease was present in 3.9% of the teeth examined. Intrinsic tooth staining was assessed in 8.5% of evaluated teeth. Of all teeth examined, 831 teeth (43.7% of evaluated teeth) exhibited some type of attrition/abrasion. Our study concluded that there is a high prevalence of dental fractures and attrition/abrasion with associated endodontic disease that could lead to impaired hunting ability as well as a threat to overall health and subsequent survival of this endangered species.

Впервые в мире создана популяция зубров на территории 60 градусов северной широты. В новых климатических условиях разведения и сохранения зубров определены и проанализированы факторы существования вида на севере Европейской части РФ. Выявлены признаки, динамика численности, которые являются составной частью системы, предназначенной для управления биоразнообразием. Интродукция, являясь процессом введения в экосистему нехарактерных для нее видов, может усиливать изменения биоценозов как положительно, так и отрицательно. Насколько быстро и успешно проходит процесс адаптации заселенного вида, и усматривается его влияние на окружающую среду зависит дальнейшее существование зубров и в целом биоразнообразия. В статье обсуждаются вопросы взаимоотношения зубров с другими видами копытных и хозяйственной деятельностью человека, а также дальнейшим использованием зубров в сельскохозяйственном производстве. Пластичность зубров, выявление изменений и их анализ при вселении видов в новые условия обитания необходимы не только для определения развития или деградации биоценозов и в целом экосистемы, но и прогноза социально-экономических последствий интродукции как одного из методов сохранения редких и исчезающих видов фауны.For the first time in the world, a bison population has been created in an area of 60 degrees north latitude. In the new climatic conditions of breeding and preservation of bison, the factors of the species existence in the north of the European part of the Russian Federation are identified and analyzed. The signs, dynamics of abundance, which are an integral part of the system designed to manage biodiversity are identified, since the preservation of biological diversity on the planet is one of the main problems of our time. Introduction, being the process of introducing non-typical species into an ecosystem, can enhance changes in biocenoses, both positively and negatively. The question posing sounds especially when it comes to such a large hoofed animal as the European bison. How quickly and successfully the process of adaptation of the universe takes place and its environmental impact is seen depends on the continued existence of bison and biodiversity in general. The article discusses the relationship of bison with other types of ungulates and human activities, as well as the further use of bison in agricultural production. How these issues will be resolved positively depends on the future of these animals. Thus, the plasticity of bison, the identification of changes and their analysis, with the introduction of species into new habitat conditions is necessary not only to determine the development or degradation of biocenoses and the ecosystem as a whole, but also to predict the socio-economic consequences due to the introduction as one of the methods of preserving rare and endangered species of fauna.

Abstract: The Resplendent Shrub Frog, Raorchestes resplendens Biju, Shouche, Dubois, Dutta, & Bossuyt, 2010 is a Critically Endangered species endemic to the Western Ghats and was considered to be restricted to a three-square kilometer patch atop Anamudi summit.  In this study, we report 36 new locations of the species from the Anamalai massif of the southern Western Ghats.  Niche-based prediction modelling suggests that the species is restricted to Anamalai massif.  The call description of this frog is also provided for the first time. The preferred microhabitat of the frog is Chrysopogon grass clumps in the marshy/swampy montane grassland ecosystem. Restricted to a small area with controlled burning management practiced in its habitat, R. resplendens needs immediate attention.
