Abstract SHOX (short stature homeobox-containing gene) mutations causing haploinsufficiency have been reported in some individuals with idiopathic short stature and in many patients with Leri-Weill-dyschondrosteosis. Around 80% of SHOX mutations are complete gene deletions, whereas diverse point mutations account for the rest. The aim of this study was to estimate the prevalence of SHOX mutations in children with idiopathic short stature and to give an unbiased characterization of the haploinsufficiency phenotype of such children. We recruited 140 children (61 girls), in our clinic, with idiopathic short stature, which was defined by the presence of normal IGF-I and free T4; a normal karyotype in females; the absence of endomysium antibodies, of chronic organic, psychological, or syndromatic disease; and by the lack of clear signs of any osteodysplasia. Height, arm span, and sitting height were recorded, and subischial leg length was calculated. Two highly polymorphic microsatellite markers located around the SHOX coding region (CA-SHOX repeat and DXYS233) were PCR-amplified with fluorescent primers and separated in an automatic sequencing machine. Analysis of parental DNA was performed in the probands who had only one fragment size of each of both markers. SHOX haploinsufficiency caused by a SHOX deletion was confirmed in three probands (2%), all females, who carried a de novo deletion through loss of the paternal allele. Their auxological data revealed a significant shortening of arms and legs in the presence of a low-normal sitting height, when compared with the other 137 children tested. Therefore, the extremities-trunk ratio (sum of leg length and arm span, divided by sitting height) for total height was significantly lower in the three SHOX haploinsufficient probands, in comparison with the whole group. This observation was confirmed with the auxological data of five additional patients (four females) previously diagnosed with SHOX haploinsufficiency; all but the youngest girl had height-adjusted extremities-trunk ratios more than 1 sd below the mean. All children with SHOX haploinsufficiency exhibited at least one characteristic radiological sign of Leri-Weill-dyschondrosteosis in their left-hand radiography, namely triangularization of the distal radial epiphysis, pyramidalization of the distal carpal row, or lucency of the distal ulnar border of the radius. Our observations suggest that it is rational to limit SHOX mutation screening to children with an extremities-trunk ratio less than 1.95 + 1/2 height (m) and to add a critical judgment of the hand radiography.

AbstractEarly-life environmental and nutritional exposures are considered to contribute to the differences in cardiovascular disease (CVD) burden. Among sub-Saharan African populations, the association between markers of early-life exposures such as leg length and sitting height and CVD risk is yet to be investigated. This study assessed the association between leg length, sitting height, and estimated 10-year atherosclerotic cardiovascular disease (ASCVD) risk among Ghanaian-born populations in Europe and Ghana. We constructed sex-specific quintiles for sitting height and leg length for 3250 participants aged 40–70 years (mean age 52 years; men 39.6%; women 60.4%) in the cross-sectional multicenter Research on Diabetes and Obesity among African Migrants study. Ten-year risk of ASCVD was estimated using the Pooled Cohort Equations; risk ≥7.5% was defined as “elevated” CVD risk. Prevalence ratios (PR) were estimated to determine the associations between sitting height, leg length, and estimated 10-year ASCVD risk. For both men and women, mean sitting height and leg length were highest in Europe and lowest in rural Ghana. Sitting height was inversely associated with 10-year ASCVD risk among all women (PR for 1 standard deviation increase of sitting height: 0.75; 95% confidence interval: 0.67, 0.85). Among men, an inverse association between sitting height and 10-year ASCVD risk was significant on adjustment for study site, adult, and parental education but attenuated when further adjusted for height. No association was found between leg length and estimated 10-year ASCVD risk. Early-life and childhood exposures that influence sitting height could be the important determinants of ASCVD risk in this adult population.

BACKGROUND: Previous research has shown that the collection and analysis of crane operators’ anthropometric characteristics is very important for operators’ comfort, health and working ability and also from the aspect of performance, productivity and safety. OBJECTIVE: The first aim of this survey is to collect up-to-date anthropometric data of crane operators in Serbia and to collect those data for the first time in Libya. The second aim of this survey is to compare the collected data. The third aim is to model the interior space necessary to accommodate operators in the cabins they operate in Serbia and Libya. METHODS: Standing height, sitting height, lower leg length, upper leg length, shoulder width, hip breadth, arm length, foot length and weight data were collected of 83 Serbian and 50 Libyan crane operators. Descriptive statistics, correlation analysis and tests for differences between variables were conducted to examine differences between Serbian and Libyan crane operators and enable further modeling. The modeling of the cabin interior was done using both univariate and multivariate operators’ models from both samples. RESULTS: There are only four common correlations between variables for both samples. The only measurement without statistical differences is shoulder width. Serbian crane operators have statistically higher values of almost all measurements in comparison to Libyans. The minimal cabin interior space dimensions are 1207×1080×1884 mm for Serbian operators and 1106×1040×1790 mm for Libyan operators when using univariate approach, while multivariate approach provides more precise and comfortable accommodation within 1327×1123×1926 mm for Serbian operators and 1203×1090×1830 mm for Libyan operators. Accordingly, our survey shows that percentile models include less than the intended population proportion in the design problem with few dimensions and depends on correlations among them. CONCLUSIONS: A number of problems are solved through modeling the crane operator workspace and it has been shown that cabins with different dimensions have to be offered to Serbian and Libyan markets.

There is evidence of a reduction in children's physical activity in India in the last decade. Our objective was to assess whether size and body composition at birth are associated with physical activity in school-aged children. Children from a prospective observational cohort study born in Mysore, South India between 1997 and 1998 (n = 663) had neonatal anthropometric measurements made within 72 h of delivery [weight, mid-upper arm circumference (MUAC), chest, abdomen and head circumference, crown–heel, crown–buttock and leg length, triceps and subscapular skinfolds]. At 6–10 years, children (n = 449) were asked to wear AM7164 or GT1M Actigraph accelerometers for 7 days. Body composition was measured within 6 months of activity monitoring. Arm muscle area at birth and time of activity monitoring was calculated from MUAC and skinfold measurements.Activity outcome measures were: mean accelerometer counts per minute (cpm); counts per day and proportion of time spent in moderate and vigorous activity. The mean (s.d.) number of days with ⩾500 min of recorded accelerometer data was 7.0 (1.1). Linear regression models showed no significant associations between any of the neonatal anthropometric measures and the activity variables. Body fat percentage at 7.5 years was negatively associated with all activity variables (B = −4.69, CI: −7.31, −2.07 for mean cpm).In conclusion, this study showed no associations between body size and skinfold thickness at birth and objectively measured physical activity in childhood.

Background: Anthropometric indexes are powerful indicators of the environment and the plasticity of the human body. This study aimed at exploring the anthropometric indexes that are associated with late-life cognition impairment among the elderly Chinese in the Shanghai Aging Study. Methods: The height, weight, and sitting height of 3,741 participants were measured. Participants were diagnosed with ‘dementia', ‘mild cognitive impairment', or ‘cognitive normal' by neurologists using DSM-IV and Petersen criteria. Logistic regression was used to evaluate the association between height, sitting height, leg length or relative sitting height and cognitive function. Results: Participants with dementia had the shortest body height (mean 157.2 cm, SD 9.1), the shortest sitting height (mean 81.8 cm, SD 5.6), and the lowest relative sitting height (mean 52.0 cm, SD 1.9). After adjustment for age, gender, education, lifestyles, medical history, apolipoprotein genotype and weight, shorter sitting height (OR 1.08, 95% CI 1.01-1.16 per cm), longer leg length (OR 0.93, 95% CI 0.88-0.99 per cm), and lower relative sitting height (OR 1.17, 95% CI 1.04-1.31 per 1%) were found to be significantly associated with dementia in older women. Conclusions: The potential risks for late-life severe cognitive impairment may be related to health problems in childhood and slow growth during puberty in women.

Abstract The aim of this study was to compare the characteristics of somatic maturation, anthropometric and physical performance (vertical jump and aerobic power) in young basketball players of different playing positions (under 13 years) and analyze these relationships using Peak Height Velocity (PHV) as a measure of somatic maturation. For this, 26 male athletes were evaluated. Anthropometric variables were: body mass, standing and sitting height, and length of lower limbs. Maturation was determined by age at PHV. Physical performance was determined by lower limb power (counter movement jump - CMJ) and aerobic power (Intermittent Recovery Test) tests. MANOVA reported significant differences (p<0.05) among playing positions regarding variables Maturity Offset, estimated PHV age, standing height, sitting height, estimated leg length, body mass and Yo-Yo IR1. In addition, it was identified that point guards reached estimated PHV at later age than their peers who act as small forwards and centers. Regarding CMJ, no significant differences were identified among playing positions, but in relation to aerobic power, point guards and small forwards presented higher performance. These findings confirm that maturation has great effect on growth and physical performance measures and the estimated PHV age is an applicable tool in young athletes, mainly aiding professionals in structuring the teaching-learning- training process in this age group.

AbstractBackgroundCentral African hunter-gatherer Pygmy populations have reduced body size compared with their often much larger agricultural non-Pygmy neighbors, potentially reflecting adaptation to the anatomical and physiological constraints of their lifestyle in tropical rainforests. Earlier studies investigating the genetics of the pygmy phenotype have focused on standing height, one aspect of this complex phenotype that is itself a composite of skeletal components with different growth patterns. Here, we extend the investigations of standing height to the variability and genetic architecture of sitting height and subischial leg length as well as body mass index (BMI) in a sample of 406 unrelated West Central African Pygmies and non-Pygmies.ResultsIn addition to their significantly reduced standing height compared with non-Pygmies, we find Pygmies to have significantly shorter sitting heights and subischial leg lengths as well as higher sitting/standing height ratios than non-Pygmies. However, while male Pygmies had significantly lower BMI compared with male non-Pygmies, the BMI of females were instead similar. Consistent with prior observations with standing height, sitting height and subischial leg length were strongly correlated with inferred levels of non-Pygmy genetic admixture while BMI was instead weakly correlated, likely reflecting the greater contribution of non-genetic factors to the determination of body weight compared with height. Using 196,725 SNPs on the Illumina Cardio-MetaboChip with genotypes on 358 Pygmy and 169 non-Pygmy individuals together with single-and multi-marker association approaches, we identified a single genomic region and seven genes associated with Pygmy/non-Pygmy categorization as well as 9, 10, 9, and 10 genes associated with standing and sitting height, sitting/standing height ratio, and subischial leg length, respectively. Many of the genes identified have putative functions consistent with a role in determining their associated trait as well as the complex Central African pygmy phenotype.ConclusionsThese findings highlight the potential of modestly sized datasets of Pygmies and non-Pygmies to detect biologically meaningful associations with traits contributing to the Central African pygmy phenotype. Moreover, they provide new insights into the phenotypic and genetic bases of the complex pygmy phenotype and offer new opportunities to facilitate our understanding of its complex evolutionary origins.

The purpose of the study was to evaluate predicted maturity offset (time before age at PHV) and age at PHV (chronological age [CA] minus maturity offset) in a longitudinal sample of 58 under-13 club level soccer players in central Portugal for whom ages at PHV were estimated with the SITAR model. Two maturity offset prediction equations were applied: the original equation which requires CA sitting height, estimated leg length, height and weight, and a modified equation which requires CA and height. Predicted maturity offset increased, on average, with CA at prediction throughout the age range considered, while variation in predicted maturity offset and ages at PHV within CA groups was considerably reduced compared to variation in observed ages at offset and at PHV. Predicted maturity offset and ages at PHV were consistently later than observed maturity offset and age at PHV among early maturing players, and earlier than observed in late maturing players. Both predicted offset and ages at PHV with the two equations were, on average, later than observed among players maturing on time. Intra-individual variation in predicted ages at PHV with each equation was considerable. The results for soccer players were consistent with similar studies in the general population and two recent longitudinal studies of soccer players. The results question the utility of predicted maturity offset and age at PHV as valid indicators of maturity timing and status.
