AbstractThe causative agent of amoebic gill disease, Neoparamoeba perurans is reported to lose virulence during prolonged in vitro maintenance. In this study, the impact of prolonged culture on N. perurans virulence and its proteome was investigated. Two isolates, attenuated and virulent, had their virulence assessed in an experimental trial using Atlantic salmon smolts and their bacterial community composition was evaluated by 16S rRNA Illumina MiSeq sequencing. Soluble proteins were isolated from three isolates: a newly acquired, virulent and attenuated N. perurans culture. Proteins were analysed using two-dimensional electrophoresis coupled with liquid chromatography tandem mass spectrometry (LC–MS/MS). The challenge trial using naïve smolts confirmed a loss in virulence in the attenuated N. perurans culture. A greater diversity of bacterial communities was found in the microbiome of the virulent isolate in contrast to a reduction in microbial community richness in the attenuated microbiome. A collated proteome database of N. perurans, Amoebozoa and four bacterial genera resulted in 24 proteins differentially expressed between the three cultures. The present LC–MS/MS results indicate protein synthesis, oxidative stress and immunomodulation are upregulated in a newly acquired N. perurans culture and future studies may exploit these protein identifications for therapeutic purposes in infected farmed fish.

In this study the transgenic lines (TLs) of tobacco (Nicotianatabacum L.), which overexpress the heterologous gene encoding the bacterial enzyme choline oxidase were evaluated. The goal of our work is to study the effect of choline oxidase gene expression on the sensitivity of plant tissues to the action of NaCl. The regenerative capacity, rhizogenesis, the amount of photosynthetic pigments and osmotically active compounds (proline and glycine betaine) were assessed by in vitro cell culture methods using biochemical and morphological parameters. Transgenic lines with confirmed expression were characterized by high regeneration capacity from callus in the presence of 200 mmol NaCl, partial retention of viability at 400 mmol NaCl. These data correlated with the implicit response of regenerants and whole plants to the harmful effects of salinity. They turned out to be less sensitive to the presence of 200 mmol NaCl in the cultivation medium, in contrast to the WT plants.

AbstrakJeruk Nipis (Citrus aurantifolia S.) merupakan salah satu tanaman obat keluarga yang banyak terdapat ditengah masyarkat dan banyak digunakan sebagai ramuan tradisional. Bagian yang sering digunakan adalah air perasannya, dengan salah satu manfaat dapat digunakan untuk menghilangkan jerawat serta penyembuhan luka agar tidak terjadi abses. Jerawat dan abses pada luka merupakan salah satu infeksi yang disebabkan oleh bakteri Staphylococcus aureus.Tujuan Penelitian ini adalah untuk mengetahui daya hambat air perasan buah jeruk nipis (Citrus aurantifolia S.) terhadap pertumbuhan bakteri Staphylococcus aureus secara invitro. Penelitian dilakukan dengan metoda eksperimental laboratorium dengan desain postest only control group design yang dilakukan di Laboratorium Mikrobiologi Fakultas Kedokteran Universitas Andalas.Hasil penelitian menunjukan bahwa air perasan buah jeruk nipis memiliki daya hambat terhadap pertumbuhan bakteri Staphylococcus aureus dengan berbagai konsentrasi yaitu 25%, 50%, 75%, dan 100% dan terdapat pengaruh lama kontak terhadap pertumbuhan bakteri dimana bakteri tidak tumbuh seteleh kontak 5 menit pertama dan diikuti menit-menit berikutnya dengan air perasan buah jeruk nipis konsentrasi 100%. Jadi, semakin tinggi konsentrasi air perasan buah jeruk nipis dan semakin lama kontak dengan bakteri Staphylococcus aureus maka daya hambatnya semakin baik.Kata kunci: Uji Daya Hambat, Air Perasan Buah Jeruk Nipis, Staphylococcus aureus.Abstract Lime (Citrus aurantifolia S.) is kind of family’s herbal medicine, most using in the community is widely used as a traditional herb. The most common used part is the lime fruit squeeze with one of the function is used for removing acne and wound healing to prevent the form of abscess. Pimples and abscesses of the wound is one of the infections caused by the bacterium Staphylococcus aureus.The purpose of this study was to determine the inhibition of lime fruit (Citrus aurantifolia S.) squeeze towards the growth of the bacteria Staphylococcus aureus in vitro condition. The study was conducted with laboratory experimental methods to the design of control group design postest only performed at the Laboratory of Microbiology Faculty of Medicine, University of Andalas.The results showed that the lime fruit (Citrus aurantifolia S.) squeeze has the ability to inhibite the bacterial growth of Staphylococcus aureus with various concentrations of 25%, 50%, 75%, and 100% and there is the effect of contact time on the growth of bacteria which the bacteria do not grow after contact the first 5 minutes and the next minute followed by lime fruit squeeze with 100% concentration lime fruit squeeze. Thus, the higher the concentration of lime fruit squeeze and the longer the contact with the bacteria Staphylococcus aureus is the better towards.Keywords:Inhibition test, The Lime Fruit Squeeze, Staphylococcus Aureus.

The active transport of calcium from the mucosal to the serosal surfaces of everted gut-sacs of the rat is dependent on the dietary vitamin D. The active transfer in vitro is significantly increased one hour following the administration of calciferol by gastric tube to rats depleted of the vitamin. Vitamins D2 and D3 are approximately equally effective, whereas dihydrotachysterol (A.T. 10) is somewhat more effective than either of the vitamins D. Ultraviolet irradiation of intact rats also increases the active transport of calcium in vitro. Maintenance on a low calcium diet increases the active transfer, whereas thyroparathyroidectomy decreases it. Vitamin D is required to demonstrate both of these effects clearly. The results support the hypothesis that the active transfer is an adaptive mechanism which ensures adequate absorption when the requirement for calcium is increased, or when the diet is low in calcium.

Foliar explants of Vanda testacea (Lindl.) Reichb. f. were cultured on Mitra (M) medium with 1.0 mg/l BAP, Kn  each and 1.0 mg/l NAA individually and in combination for initiation of regeneration response, proliferation of regenerants and subsequent development of plantlets. Juvenility of the tissues and chemical stimulus were important factors in initiating the regeneration response in the explants. The relatively older leaf explants (>1cm in length) remained recalcitrant to regeneration the representing younger ones (<1cm in length) responded to certain chemical regimes. BAP, Kn individually in the medium should direct PLB regeneration whereas when used with NAA, the explants showed callus proliferation and further differentiated into PLBs. An individual treatment with NAA (1.0 mg/l) impaired the response frequency and delayed further morphogenetic processes leading to plantlet development. The best response in the explants (in terms of high regeneration frequency, early initiation, PLB proliferations, and plantlet development) was observed in 1.0 mg/l BAP alone/with 1.0 mg/l NAA + activated charcoal. Plantlets were transferred to pots containing epiphytic compost (1 charcoal : 1 brick pices : 1 bats).
