The α-ketoacid–hydroxylamine (KAHA) ligation allows the coupling of unprotected peptide segments. The most widely used variant employs a 5-membered cyclic hydroxylamine that forms a homoserine ester as the primary ligation product. While very effective, monomers that give canonical amino acid residues are in high demand. In order to preserve the stability and reactivity of cyclic hydroxylamines, but form a canonical amino acid residue upon ligation, we sought to prepare cyclic derivatives of serine hydroxylamine. An evaluation of several cyclization strategies led to cyclobutanone ketals as the leading structures. The preparation, stability, and amide-forming ligation of these serine-derived ketals are described.

Syntheses are described of four peptides with modified leucine-enkephalin sequences in which the native glycylglycyl segment is replaced by an ω-amino acid residue. o-Nitrophenylthio-ω-amino acids were used as intermediates, and it was found that the derivatives of 4-aminobutyric and 5-amino-valeric acids undergo facile intramolecular cyclization under the influence of N,N'-dicyclohexylcarbodiimide.

Samples of various grain crops (wheat, barley, oats) of agricultural enterprises of the Krasnoyarsk Territory in the central and western zones (LLC UCHHOZ MINDERLINSKOE "Sukhobuzimsky District, LLC ELEVATOR Uzhursky District, the composition of OJSC Iskra. It was determined that compared with wheat and barley oats are characterized by the highest content of the essential amino acid lysine, all crops have a low level of tryptophan. It was found that during storage of grain there was no significant change in the amino acid residue. acid composition, which indicates a low rate of the processes leading to a change in biological value. A previous paper described the complete amino acid sequences of sarcotoxins IA, IB and IC, which are a group of potent antibacterial proteins with almost identical primary structures produced by Sarcophaga peregrina (fleshfly) larvae [Okada & Natori (1985) J. Biol. Chem. 260, 7174-7177]. The present paper describes the cDNA cloning and complete nucleotide sequencing of a cDNA clone for sarcotoxin IA. The C-terminal amino acid residue of sarcotoxin IA deduced from the nucleotide sequence was glycine, whereas it was found to be arginine by amino acid sequencing of purified sarcotoxin IA. Analysis of the elution profiles on h.p.l.c. of the synthetic derivatives of sarcotoxin IA showed that the C-terminal amino acid residue of authentic sarcotoxin IA is amidated arginine, which is probably produced by enzymic cleavage of terminal glycine.
