The Aggregation Pheromone of Metamasius spinolae (Coleoptera: Dryophthoridae) Revisited: Less is More

Abstract The weevil Metamasius spinolae (Gyllenhal) is the most important insect pest of cultivated prickly pear in Mexico. A previous work reported that the pheromone of this weevil species was composed by three components. In this study, we reinvestigated the aggregation pheromone of M. spinolae using gas chromatography-electroantennography and gas chromatography–mass spectrometry to locate and identify new potential pheromonal compounds. The behavioral activity of identified compounds was evaluated in laboratory and field trials. Metamasius spinolae males released four compounds: 2-methyl-4-heptanone, 6-methyl-2-hepten-4-one, 2-methyl-4-octanone, and 2-hydroxy-2-methyl-4-heptanone. In the laboratory assays, depending on the concentration, the compounds were attractive, neutral, or repellent to M. spinolae. Field evaluation showed that traps baited with 2-hydroxy-2-methyl-4-heptanone singly or in most of the binary or tertiary blends where this compound was present captured a higher number of M. spinolae compared to live males and the other compounds identified. In conclusion, our results indicate that 2-hydroxy-2-methyl-4-heptanone is the main component of the aggregation pheromone of M. spinolae. We suggest that this compound should be used for developing a monitoring or a mass-trapping system for M. spinolae.

Thymus bovei Benth. (TB) is an important plant in the traditional medicine of the Mediterranean region. This study investigates the health-promoting properties of TB essential oil (TB-EO) for its possible use in clinical practice with regards to its cytotoxic, anti-herpes simplex virus type 2 (HSV-2), and antihypertensive (through inhibition of human angiotensin-converting enzyme; ACE) properties. The phytochemical profile of EO (99.9%) was analyzed by Gas Chromatography with Flame-Ionization Detection (GC-FID) and Gas Chromatography-Mass Spectrometry (GC-MS). In this study, all biological methods were performed at the level of in vitro studies. The results showed that TB-EO exerted remarked cytotoxic properties against human cervical carcinoma cells, colon cancer cells, and lung adenocarcinoma cells with the half-maximal inhibitory concentration (IC50) values of 7.22, 9.30, and 8.62 µg/mL, respectively, in comparison with that of standard anticancer drug cisplatin with IC50 values of 4.24, 5.21, and 5.43 µg/mL, respectively. Fascinatingly, TB-EO showed very weak cytotoxicity on the healthy human fetal lung fibroblast cells with an IC50 value of 118.34 µg/mL compared with that of cisplatin (IC50 = 10.08 µg/mL). TB-EO, its main component geraniol, TB-EO combined with acyclovir (ACV) along with standard ACV, have displayed pronounced inhibitory properties against the replication of HSV-2 with the half-maximal effective concentration (EC50) values of 2.13, 1.92, 0.81 and 1.94 µg/mL, respectively, with corresponding selectivity indices (SI) 98.59, 109.38, 259.26 and 108.25, respectively. TB-EO and geraniol at a concentration of 15 µg/mL showed prominent inhibitory activities against ACE with % of inhibition 95.4% and 92.2%, respectively, compared with that of standard inhibitor captopril (99.8%; 15 µg/mL). Molecular docking studies were performed to unveil the mechanism of action of geraniol as well as structural parameters necessary for anti-HSV-2 activity (through the inhibition of HSV-2 protease) and ACE inhibition. This is the first report on the chemical composition of Egyptian TB-EO along with the above-mentioned biological activities. Our results may be considered as novel findings in the course of a search for new and active anticancer, anti-HSV-2 and antihypertensive agents, and expand the medicinal value of this plant and its phytochemicals in clinical practice.

The essential oils from Magnolia grandiflora and Chrysactinia mexicana leaves, and from Schinus molle leaves and fruit, were characterized by gas chromatography/flame-ionization detection and gas chromatography/mass spectrometry. Twenty-eight compounds from M. grandiflora leaves were identified (representing 93.6% of the total area of the gas chromatogram), with the major component being bornyl acetate (20.9%). Colorless and yellow oils were obtained from the C. mexicana leaves with 18 (86.7%) and 11 (100%) compounds identified, respectively. In both fractions, the principal component was sylvestrene (36.8% and 41.1%, respectively). The essential oils of S. molle leaves and fruit were each separated into colorless and yellow fractions, in which 14 (98.2) and 20 (99.8%) compounds were identified. The main component was α-phellandrene in all fractions (between 32.8% and 45.0%). The M. grandiflora oil displayed antifungal activity against five dermatophyte strains. The oils from S. molle and M. grandiflora leaves had antimicrobial activity against Staphylococcus aureus and Streptococcus pyogenes, which cause skin infections that potentially may lead to sepsis. However, the antioxidant activities of all oils were small (half maximal effective concentration values >250 μg/mL).

Conifers are well represented in Vietnam where a new pine species has been recently discovered in Son La province: Pinus cernua, synonym P. armandii ssp. xuannhaensis. The compositions of needle, cone, and branch oils have been investigated by gas chromatography (retention index), gas chromatography-mass spectrometry, and 13C nuclear magnetic resonance. Myrcene (47.0%) was the main component of needle oil, followed by β-pinene (28.4%) and α-pinene (12.5%). Branch oil also contained myrcene (32.8%), α-pinene (17.9%), β-pinene (9.8%), and a high content of limonene (20.0%). Finally, cone oil displayed α-pinene (44.1%) beside myrcene (11.5%), β-pinene (8.1%), and limonene (5.8%).

The hydro-distilled essential oil of Citrus reticulata Blanco var. kinnow was analyzed by Gas chromatography-mass spectrometry (GC-MS). Five constituents out of fifteen constituents were identified from seeded C. reticulata oil representing 74.66% of the oil. The major constituent of the oil was ?- phellandrene (62.00%). ?-pinene(6.53%), ?-myrcene(2.81%), limonene(2.81%) and caryophyllene(0.51%) were present in considerable amount. From the low seeded C. reticulata oil, six components out of seventeen compounds were identified constituting 54.74% of the oil and the main component was ?-phellandrene (37.35%). ?-pinene(2.79%), ?-pinene(3.26%), ?-myrcene(4.16%), limonene(5.77%), caryophyllene(1.41%) were present in considerable amount. Extracts of female sex pheromone glands of the sand Salix carpenterworm moth, Holcocerus arenicola, a serious pest of desert thicket, were analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Based on comparison of retention times and mass spectra of synthetic standards, four compounds were identifi ed as cis-7-tetradecen-1-ol (Z7 - 14:OH), cis-5-tetradecen-1-yl acetate (Z5 - 14:OAc), cis-7-tetradecen- 1-yl acetate (Z7 - 14:OAc), and cis-9-hexadecen-1-yl acetate (Z9 - 16:OAc) with the ratio of 24:39:100:43. Electroantennographic (EAG) analyses of these standard chemicals and their analogues showed that Z7 - 14:OAc elicited the largest male EAG response, followed by Z5 - 14:OAc and Z9 - 16:OAc. In fi eld trials, traps baited with either Z7 - 14:OAc or Z5 - 14:OAc captured males while Z7 - 14:OH-, Z9 - 16:OAc- or solvent-baited traps caught no males. Z7 - 14:OAc as a single component was signifi cantly more attractive than Z5 - 14:OAc alone. The combination of Z7 - 14:OAc and Z5 - 14:OAc showed an evidently synergistic effect and attracted much more males than the individual compounds in the field. Addition of Z7 - 14:OH to the blend of Z7 - 14:OAc and Z5 - 14:OAc enhanced slightly the trap catches. We conclude that the major components of the sex pheromone of H. arenicola are Z7 - 14:OAc and Z5 - 14:OAc. Currently, a triangle trap baited with the synthetic compounds Z7 - 14:OAc, Z5 - 14:OAc, and Z7 - 14:OH in a 1:0.4:0.25 ratio at 825 μg/trap dosage can be effectively used to monitor the H. arenicola population level and catch the males within the desert regions in China.

Onion thrips (Thrips tabaci) is the main insect pest of onion (Allium cepa), and feeding damage routinely causes serious yield losses. Lower amounts of epicuticular waxes on onion leaves have been associated with fewer onion thrips and less feeding damage, and research is needed to assess the relationships between amounts and composition of epicuticular waxes and feeding damage by onion thrips. This study used gas chromatography mass spectroscopy to determine amounts and types of epicuticular waxes on the foliage of onion accessions that had been field-selected for lower damage from onion thrips. Hentriacontanone-16 (H16), octaconasol-1, and triacontanol-1 were the most prevalent waxes on the foliage of these selections. Amounts of H16 were significantly lower on selections visually classified as having glossy or semiglossy foliage. Semiglossy selections were identified with similar amounts of total epicuticular wax as waxy phenotypes, due primarily to lower amounts of H16 and higher amounts of other waxes. These semiglossy selections suffered significantly less feeding damage from onion thrips in a field evaluation, supporting the identification of unique wax profiles toward the development of thrips-resistant onion.

The aims of our research include the phytochemical characterization of the essential oils extracted from different lavender species ( Lavandula vera L., L. intermedia L., L. pyrenaica DC., and L. stoechas subsp. stoechas) that are important from therapeutic and economic aspects, as well as the optimization of the tube dilution method for microbiological investigation of the effect of essential oils on the selected Pseudomonas strain. The chemical and percentage compositions of the essential oils were analysed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). The main component of the essential oils in the majority of lavender species was linalool, while L-fencone was identified in the largest amount in L. stoechas subsp. stoechas. Tube dilution is one of the most frequently used methods for microbiological investigation. Its advantages are that it is easy to perform and provides information about minimum bactericidal concentration (MBC). Because of the lipophilic character of essential oils, the tube dilution method should be optimized for the detection of antibacterial activity of these plant extracts, and, therefore, emulsions containing 0.2% polysorbate 80 were prepared from the samples. In each of the eight investigated essential oils an inhibitory effect was detected, and MBC values were in the range 12.5-50 μL/mL in seven cases.

Twenty-five compounds were identified from the essential oil of Globba pendula Roxb. by gas chromatography–mass spectrometry (GC-MS) with Selinene<d-> (22.09%) as the main component, followed by Ishwarane (6.52%). The essential oil was found to possess moderate NO inhibitory effect with IC50 of 41.68 ±4.51 µg/ml, significant cytotoxic activity against MCF7 and Hep3B cell lines with IC50 of 28.15±1.08 and 35.24±0.06 (µg/ml), respectively. This is the first report on volatile compounds and biological activities of essential oil of Globba pendula Roxb. collected at An Giang province, Vietnam.

The essential oil from the stem bark and leaves of Dennettia tripetala Bak. f. (Annonaceae) growing wild in Ondo State, Nigeria, has been characterized by combined gas chromatography (GC) and gas chromatography-mass spectrometry (GC- MS) analyses. Overall, thirty-six components have been fully identified, thirty-two in the stem-bark oil, and only seven in the leaf oil. In both oils, 2-phenylnitroethane was the main component, ranging between 70 – 76% of the total oils. The profile of the stem bark oil was characterized by a large number of sesquiterpenes, whereas among the few components in the leaf oil, linalool reaches over 17%. When both oils were assayed for antimicrobial activity, only Staphylococcus aureus was susceptible to the stem-bark oil which was more active than leaf oil. For protective effects against UV radiation–induced peroxidation in phosphatidylcholine (PC) liposomes, stem-bark oil also showed greater effectiveness. Activity of the leaf oil against Trichomonas gallinae, was also remarkable.

AbstractCallosobruchus maculatus (F.) (Coleoptera: Bruchidae) is an important stored grain pest of Lathyrus sativus L. (Leguminosae), commonly known as khesari, in India, Bangladesh and Ethiopia. Volatiles were collected from four varieties, i.e., Bio L 212 Ratan, Nirmal B-1, WBK-14-7 and WBK-13-1 of uninfested khesari seeds, and subsequently identified and quantified by gas chromatography mass spectrometry and gas chromatography flame ionization detector analyses, respectively. A total of 23 volatiles were identified in the four varieties of khesari seeds. In Bio L 212 Ratan and WBK-13-1 seeds, nonanal was the most abundant followed by farnesyl acetone; whereas farnesyl acetone was predominant followed by nonanal in Nirmal B-1 and WBK-14-7 khesari seeds. The olfactory responses of female C. maculatus toward volatile blends from four varieties of khesari seeds, and individual synthetic compounds and their combinations were examined through Y-shaped glass tube olfactometer bioassays. Callosobruchus maculatus showed significant preference for the whole volatile blends from Bio L 212 Ratan seeds compared to whole volatile blends from other three varieties. The insect exhibited attraction to five individual synthetic compounds, 3-octanone, 3-octanol, linalool oxide, 1-octanol and nonanal. A synthetic blend of 448, 390, 1182, 659 and 8114 ng/20 μl methylene chloride of 3-octanone, 3-octanol, linalool oxide, 1-octanol and nonanal, respectively, was most attractive to C. maculatus, and this combination might be used for insect pest management program such as baited traps.
