Characterization and detection of Passiflora mottle virus and other two potyviruses causing passionfruit woodiness disease in Vietnam

Passionfruit plantation in Vietnam increased to 10,000 ha in 2019. However, the outbreaks of passionfruit woodiness disease (PWD) have become a serious threat for the production. In this study, five virus isolates DN1, DN4, NA1, GL1 and GL2 were collected from different areas of Vietnam. Their causal roles for PWD were verified by back inoculation to passionfruit. Analyses of coat protein (CP) and genomic sequences revealed that GL1 isolate is closely related to East Asia Passiflora virus (EAPV) AO strain of Japan (polyprotein nt/aa identities of 98.1% / 98.2%), while GL2 isolate is related to Telosma mosaic virus (TelMV) isolate PasFru, China (polyprotein nt/aa identities of 87.1% / 90.9%). CP comparison, host range and cytological characterization indicated that DN1, DN4 and NA1 are potyviruses, but different from EAPV and TelMV. Phylogenic analyses of their CP and genome sequences indicated that these three isolates and passionfruit severe mottle-associated virus Fujian isolate of China belong to a distinct clade, which does not satisfy the threshold (76% nt identity of polyprotein) to be regarded as any of potyviral species. Thus, a new species name of “Passiflora mottle virus” has been proposed by ICTV. A rabbit antiserum was produced against the CP of DN1 and it can discriminate Passiflora mottle virus (PaMoV) from TelMV and EAPV in western blotting and ELISA without cross reactions. Field surveys of 240 samples by ELISA and RT-PCR disclosed that PWD in Vietnam is mainly caused by PaMoV; followed by EAPV, mixed-infection of PaMoV/EAPV, and rare cases of TelMV.

Hysterionica s.l. (including Neja) comprises ca. 15 species distributed mainly in the grasslands of southern Brazil, Uruguay, central and northern Argentina and Southern Paraguay. Based on field surveys, study of nomenclatural types and other specimens, and review of literature, a new species is proposed here, Hysterionica chamomilloides, endemic of mountainous regions of northern Uruguay, and southwestern Rio Grande do Sul state, Brazil. This work provides a detailed morphological description of the new species, information on its geographic distribution, habitat, phenology, conservation status, and discusses the main differences between H. chamomilloides and the morphologically most similar species. Illustrations, images and a key to the South American species of Hysterionica are also supplied.

Abstract Three Hungarian (No.2, 4 and 9), and a Moldavian (K) plum pox virus isolates were compared with a characterized Spanish isolate (5.15) by RT-PCR, ELISA, dot-blot and West­ern blot analysis. Monoclonal antibodies prepared against the external, intermediate and internal sequences of the coat protein of the Spanish isolate were able to differentiate the four isolates. Hungarian isolate No. 2 proved to be serologically identical to the Spanish isolate, while No. 4 showed appreciable differences and No. 9 could be recognized only by the monoclonal antibodies representing the intermedial and internal parts of the coat protein. K isolate showed a more distant relationship to other isolates. Our experiment provided the first demonstration of the presence of D type isolates in Hungary.

High-mountain regions are known to harbour considerable biodiversity, although it is not all well known. The terrestrial fauna of the world’s largest mountain range, the Himalayas, has been moderately well studied, but this is not the case with the limnic fauna, and especially molluscs. During intensive malacozoological field surveys conducted over the past 20 years, the bivalve family Sphaeriidae has been studied in Nepal along an elevational gradient from 100 to 4010 m above sea level (a.s.l.). Here we describe a new species of Sphaeriidae, Pisidium alexeii, sp. nov., based on comprehensive molecular phylogenetics, anatomy and shell morphology. The species can be clearly distinguished from all other sphaeriid species occurring in Nepal. A molecular phylogeny based on mitochondrial and nuclear data inferred the oriental biogeographical affinity of the new species. The species is ecologically restricted and only occurs at a few sites between 1010 and 1700 m a.s.l. A review and updated checklist of the sphaeriid fauna of Nepal is provided and biodiversity and biogeographical patterns are discussed.

During field surveys conducted in Albania in 2014, we encountered 90 species of butterflies. Anthocharis damone Boisduval, 1836 is a new species for the fauna of Albania while Apatura metis Freyer, 1829 is confirmed for the country. With these discoveries the number of butterfly species known to occur in Albania has risen to 200 and our surveys added many new sites for species considered rare and local in Albania. In addition, the life cycles of Lycaena ottomana (Lefèbvre, 1830) and Apatura metis from Albania are documented and discussed.

Cherry virus A (CVA), a member of the genus Capillovirus, has been reported in sweet cherry in Germany, Canada, and Great Britain. No data are available on the effects of CVA on fruit quality and yield of infected trees. Little cherry disease (LChD) occurs in most cherry growing areas of the world. Symptoms on sensitive cultivars include discolored fruit that remain small, pointed in shape, and tasteless. Three Closterovirus spp. associated with LChD have been described (Little cherry virus-1 [LChV-1], LChV-2, and LChV-3). Diseased local and commercial cultivars of sour cherry trees were found in a Prunus sp. germplasm collection and orchards in Poland during the 2003 growing season. The foliar symptoms included irregular, chlorotic mottling, distortion, and premature falling of leaves. Some of the diseased trees developed rosette as a result of decreased growth and shortened internodes. Severely infected branches exhibited dieback symptoms. Because the symptoms were suggestive of a possible virus infection, leaf samples were collected from 38 trees and assayed for Prune dwarf virus and Prunus necrotic ringspot virus using double antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA). RNA extracted from leaves was used in a reverse transcription-polymerase chain reaction (RT-PCR) with the One-Step RT-PCR with Platinum Taq (Invitrogen Life Technologies) and primer sets specific for CVA (1), LChV-1 (3), and LChV-2 (3). The RNA samples were also tested using RT-PCR for detection of Cherry mottle leaf virus (CMLV), Cherry necrotic rusty mottle virus (CNRMV), and Cherry green ring mottle virus (CGRMV) with specific primer sets (2). Amplification of a 397-bp coat protein gene product confirmed infection of 15 trees with CVA. A 419-bp fragment corresponding to the coat protein gene of LChV-1 was amplified from cv. Gisela rootstock and local cv. WVIII/1. To confirm RT-PCR results, CVA amplification products from local cv. WX/5 and LChV-1 from cvs. Gisela and WVIII/1 were cloned in bacterial vector pCR 2.1-TOPO and then sequenced. The sequences were analyzed with the Lasergene (DNASTAR, Madison, WI) computer program. The alignment indicated that the nucleotide sequence of cv. WX/5 was closely related to the published sequences of CVA (Genbank Accession No. NC_003689) and had an 89% homology to the corresponding region. The nucleotide sequence similarity between the 419-bp fragment obtained from cvs. Gisela and WVIII/1 was 87% and 91%, respectively, compared with the reference isolate of LChV-1 (Genbank Accession No. NC_001836). The sampled trees tested negative for LChV-2, CGRMV, CMLV, and CNRMV using RT-PCR. Some trees tested positive for PNRSV and PDV. The full-length genomes of two isolates of Rice yellow mottle virus from Ethiopia were sequenced. A comparison with 28 sequences from East Africa showed that they clustered within a new strain named S4et, related to the S4mg and S4ug strains found in the Lake Victoria Basin and Madagascar, respectively.

An integrative taxonomic analysis of newly discovered populations of Hemiphyllodactylus from Indochina recovered a new species from Peninsular Thailand and two others from Laos. The new Thai species, Hemiphyllodactylus pardalis sp. nov. described herein, is the sister species to all other species in the newly designated Indochina clade and has a unique spotted contrasting dorsal pattern not seen in any other species of Hemiphyllodactylus. This, along with its elongate and gracile body morphology, renders it remarkably convergent on the Philippine gecko Pseudogekko smaragdinus despite the fact it is deeply nested within Hemiphyllodactylus. The description of this new species underscores the depauperate nature of Hemiphyllodactylus diversity throughout Indochina in general and Thailand in particular compared to neighboring upland areas of Myanmar and Peninsular Malaysia. This especially underscores the need for continued herpetofaunal field surveys in montane areas of western Thailand.
