Effects of nutritional status and diet composition on fatty acid transporters expression in zebrafish (Danio rerio )

AbstractThe metabolic impact of circulating fatty acids (FAs) in patients requiring hemodialysis (HD) is unknown. We investigated the associations between plasma triglyceride (TG) FAs and markers of inflammation, insulin resistance, nutritional status and body composition. Plasma TG-FAs were measured using gas chromatography in 341 patients on HD (age = 55.2 ± 14.0 years and 54.3% males). Cross-sectional associations of TG-FAs with 13 markers were examined using multivariate linear regression adjusted for potential confounders. Higher levels of TG saturated fatty acids were associated with greater body mass index (BMI, r = 0.230), waist circumference (r = 0.203), triceps skinfold (r = 0.197), fat tissue index (r = 0.150), serum insulin (r = 0.280), and homeostatic model assessment of insulin resistance (r = 0.276), but lower malnutrition inflammation score (MIS, r =  − 0.160). Greater TG monounsaturated fatty acid levels were associated with lower lean tissue index (r =  − 0.197) and serum albumin (r =  − 0.188), but higher MIS (r = 0.176). Higher levels of TG n-3 polyunsaturated fatty acids (PUFAs) were associated with lower MIS (r =  − 0.168) and interleukin-6 concentrations (r =  − 0.115). Higher levels of TG n-6 PUFAs were associated with lower BMI (r =  − 0.149) but greater serum albumin (r = 0.112). In conclusion, TG monounsaturated fatty acids were associated with poor nutritional status, while TG n-3 PUFAs were associated with good nutritional status. On the other hand, TG saturated fatty acids and TG n-6 PUFAs had both favorable and unfavorable associations with nutritional parameters.

We examined the long-term effects of nutritional status and the acute effects of changes in exogenous carbohydrate- and lipid-substrate supply and utilization on fructose 2,6-bisphosphate (Fru-2,6-P2) concentrations in heart, gastrocnemius and soleus. Starvation decreased Fru-2,6-P2 concentrations in all three muscles. The acute administration of insulin and glucose increased skeletal-muscle Fru-2,6-P2 in the fed, but not in the starved, state, but cardiac Fru-2,6-P2 was unchanged. Cardiac and skeletal-muscle Fru-2,6-P2 concentrations were unaffected by acute increases in fatty acid supply produced by the administration of corn oil plus heparin, or by acute decreases in fatty acid supply produced by inhibition of lipolysis. Differences in cardiac and skeletal-muscle Fru-2,6-P2 concentrations observed in response to starvation were not reversed by administration of glucose or glucose plus insulin, or by inhibition of lipolysis, even though changes in citrate (heart), acylcarnitine (heart) and glycogen (skeletal muscle) were observed. Concentrations remained low for at least 8 h after chow re-feeding, but the fed value was restored by 24 h.

Rock lobster species are found worldwide and have a life history that includes development through a planktonic phyllosoma followed by a nektonic non-feeding puerulus that relies on stored energy during recruitment into near-shore habitats. Recruitment to adult populations of western rock lobster (Panulirus cygnus) is highly variable and is likely to be strongly influenced by shelf width and oceanic conditions affecting cross-shelf transport and nutrition. Since the nutritional status of newly settled pueruli will reflect the phyllosoma feeding environment and distance swum, we studied levels of lipid, fatty acid (FA) and protein of 422 pueruli and 79 first instar juveniles from four Western Australian locations. Lipid levels generally were inversely related to shelf width but were variable, suggesting pueruli may travel complex trajectories to coastal settlement. Lipid and FA composition of pueruli were consistent with spatial and seasonal variation in Leeuwin Current and coastal productivity regimes. Seasonal differences in FA composition occurred regardless of the year of settlement. Pueruli had lower lipid levels during ENSO years, when recruitment tends to be lower also. Measures of puerulus nutritional status appear to provide valuable insights into the processes underpinning recruitment in Panulirus cygnus and other commercially and ecologically important species.
