Holothuria scabra larvae on different seagrass substrates. By: Lisa Fajar Indriana, Yuli Afrianti, Sitti Hilyana, and Muhammad FirdausSandfish Holothuria scabra is marine commodities with a high economic value. Overfishing of natural stocks has compelled an interest to begin aquaculture practice. Settlement is a critical phase for the planktonic larvae as they will transform to benthic form in the presence of substrate. This study aims to evaluate the settlement preferences, growth, and survival rate of H. scabra larvae settled on different seagrass leaves. The research was conducted using the Completely randomized design with four different species of seagrass leaves and five replications. The treatments consist of Enhalus acoroides (L-1), Syringodium isoetifolium (L-2), Cymodocea serrulata (L-3), and Cymodocea rotundata (L-4). Initial number of larvae was 1,000 individuals and the substrate was set with same widthof 12 cm x 17 cm for each unit. Results of the experiment indicated that settlement preference and survival rate of H. scabra larvae was significantly affected by seagrass used as substrate while no significantly differences was observed for growth of larvae. E. acoroides showed the best result with 0.26 ind. cm-2 settelement preference and 10.66% survival rate, so that suitable to be used as settlement substrate in H. scabra hatchery.

Seagrasses habitats have both physical and ecological functions that support adjacent waters qualities and its dwelling organisms. There are varies of pressure on seagrass environmental, especially due to people activities that could decrease its function and habitat stabilities. The study aimed to determine about seagrass habitat stabilities from its species composition and/or densities. Line transect-plots and exploration methods were used in five locations of Manokwari coastal waters, that were, Andai, Rendani, Wosi, Briosi, and Tanjung Manggewa. There are five pioneer species (Cymodocea rotundata, Halodule pinifolia, H. uninervis, Syringodium isoetifolium, Halophila ovalis) and 3 climax species (Cymodocea serrulata, Enhalus acoroides, Thalassia hemprichii). The pioneer only found in Andai and Wosi, however both pioneer and climax encountered in three other locations. In Rendani and Tanjung Manggewa higher density of climax species (T. hemprichii) were 617.7 and 828.0 stands m-2, respectively, although in Briosi the higher pioneer (C. rotundata) of 570.7 stands m-2. These conditions showed that seagrass habitat in Rendani and Tanjung Manggewa are more stable compared to Briosi, as well as Andai and Wosi. It concluded that pioneer species found in newly formed habitat or disturbed, on the other hand, climax in more stable habitat.

AbstractMatrix metalloproteinases (MMPs) are pivotal for cancer cell migration and metastasis which are generally over-expressed in such cell types. Many drugs targeting MMPs do so by binding to the conserved catalytic domains and thus exhibit poor selectivity due to domain-similarities with other proteases. We report herein the binding of a novel compound [3-(E-3,4-dihydroxycinnamaoyloxyl)-2-hydroxypropyl 9Z, 12Z-octadeca-9, 12-dienoate; Mol. wt: 516.67 Da], (C1), isolated from a seagrass, Cymodocea serrulata to the unconserved hemopexin-like (PEX) domain of MMP2 (− 9.258 kcal/mol). MD simulations for 25 ns, suggest stable ligand-target binding. In addition, C1 killed an ovarian cancer cell line, PA1 at IC50: 5.8 μM (lesser than Doxorubicin: 8.6 µM) and formed micronuclei, apoptotic bodies and nucleoplasmic bridges whilst causing DNA laddering, S and G2/M phase dual arrests and MMP disturbance, suggesting intrinsic apoptosis. The molecule increased mRNA transcripts of BAX and BAD and down-regulated cell survival genes, Bcl-xL, Bcl-2, MMP2 and MMP9. The chemical and structural details of C1 were deduced through FT-IR, GC–MS, ESI–MS, 1H and 13C NMR [both 1D and 2D] spectra.

In India 14 seagrass species can be found with monospecific genera (Enhalus, ThalassiaandSyringodium),Cymodoceawith two species andHalophilaandHalodulerepresented by more than two taxonomically complex species. Considering this, the present study was made to understand the level and pattern of genetic variability among these species collected from Tamilnadu coast, India. Random amplified polymorphic DNA (RAPD) analysis was used to evaluate the level of polymorphism existing between the species. Out of the 12 primers tested, 10 primers amplified 415 DNA fragments with an average of 41.5 fragments per primer. Of the total 415 amplified fragments only 123 (29.7%) were monomorphic and the remaining 292 (70.3%) were polymorphic for Indian seagrass species. Among the 10 primers used four are identified as the key primers capable of distinguishing all the Indian seagrasses with a high degree of polymorphism and bringing representative polymorphic alleles in all the tested seagrasses. From the present investigation, this study shows that the RAPD marker technique can be used not only as a tool to analyse genetic diversity but also to resolve the taxonomic uncertainties existing in the Indian seagrasses. The efficiency of these primers in bringing out the genetic polymorphism or homogeneity among different populations of theHalophilaandHalodulecomplex still has to be tested before recommending these primers as an identification tool for Indian seagrasses.
