To investigate whether low-level lighting is necessary and which narrow-band light spectra are effective in seeds germination, seeds of 14 genotypes from begonia, echinacea, gerbera, petunia, and vinca were germinated under ultraviolet-B (UVB), blue (B), green (G), red (R), far-red (FR) light, or darkness. Light-emitting diode (LED) fixtures provided all spectrum treatments except for UVB, which was provided by a narrow-band fluorescent light. The photon flux density at seed level was ≈18 µmol m-2 s-1 for B, G, R, and FR, and 0.4 µmol m-2 s-1 for UVB. Based on daily germination observations, final germination percentage, germination onset time, germination time spread, and germination speed were compared among different spectrum treatments for each of the plant genotypes. There were no promotive effects on final germination percentage, germination onset time, and germination time spread under the narrow-band lights compared to darkness. For all plant genotypes, B had a similar effect as darkness on seed germination. FR inhibited seed germination relative to darkness by reducing final germination percentage by 31–88%, delaying germination onset time by 30–40%, and decreasing germination speed by 11–48% in some genotypes. Under R, G, and UVB compared to darkness, germination speed was promoted for begonia ‘Apricot Shade’, a light-requiring genotype, and inhibited for vinca ‘Burgundy’, a light-inhibited genotype. Therefore, lighting at low levels used in our study is unnecessary for seed germination of the tested species except light-requiring genotypes, where R, G, and UVB are the most promotive among the tested narrow-band lights.

Abstract. Ku-Or Y, Leksungnoen N, Onwinom D, Doomnil P. 2020. Germination and salinity tolerance of seeds of sixteen Fabaceae species in Thailand for reclamation of salt-affected lands. Biodiversitas 21: 2188-2200. Over the years, areas affected by salinity have increased dramatically in Thailand, resulting in an urgent need for reclamation of salt-affected areas using salinity tolerant plant species. In this context, seed germination is an important process in plant reproduction and dispersion. This research aimed to study the ability of 16 fabaceous species to germinate and tolerate salt concentrations of at 6 different levels (concentration of sodium chloride solution, i.e., 0, 8, 16, 24, 32, and 40 dS m-1). The germination test was conducted daily for 30 days, and parameters such as germination percentage, germination speed, and germination synchrony were calculated. The electrical conductivity (EC50) was used to compare the salt-tolerant ability among the 16 species. Our results showed that the germination percentage, germination speed, and germination synchrony of all species decreased with an increase in salinity concentration. The mean germination time increased from 2-4 days under 0 dS m-1 to 4-10 days under 16 dS m-1, indicating that the seeds germinated at a slower rate as the salinity levels increased. Seed germination percentage was more than 80% across all the species at salinity levels between 0 to 16 dS m-1. Sesbania grandiflora, Senna siamea, and Dalbergia cochinchinensis had a high EC50 value of 33.56, 32.93, and 30.83 dS m-1 respectively, suggesting that these species were the three most salt-tolerant species in this study. As such, the establishment of these species in salt-affected areas should be studied further in order to reclaim such affected lands.

Abstract: Chitosan is a biopolymer obtained from deacetylation of chitin; it has multiple applications in agriculture as an antifungal, soil conditioner, inducer of defense mechanisms, fruits postharvest coating, leaves and seeds, among others. The objective in this research was to evaluate the effect of chitosan coatings mixed with fungicide (dithiocarbamate) on the germination and germination speed of bean and maize seeds in storage and to determine the retention capacity of the fungicide in the coated seeds under different times of imbibition. Two coating treatments at concentrations of 0.1 and 0.5% chitosan in water, two coatings treatments at 0.1 and 0.5% chitosan supplemented with 0.5% fungicide and a coating without chitosan using only 0.5% fungicide in water were used in bean and maize seed; and as control seeds imbibed in distilled water were used; after treatments, germination percentage and germination speed were determined, also fungicide release were determined at 0, 1, 2 and 6 h of imbibition, and the effect of storage time on germination and germination speed was determined at 30, 60, 90, 120, 150 and 180 days of storage at 4 °C and 45% relative humidity. The fungicide release effect was determined by inhibiting Fusarium oxysporum conidia germination. There were no negative effects of coatings on seed germination after storage. The treatment that provided both greater retention of the fungicidal agent and released it gradually, was 0.5% chitosan mixed with fungicide concentration. Chitosan coating seeds mixed with fungicide do not cause negative changes in seed germination or germination rate.

The goal of this study was to evaluate concentrations of nanosized TiO2 at 0, 5, 20, 40, 60 and 80 mg L-1 with bulk TiO2 for possible stimulatory effects on wheatgrass seed germination and early growth stage. After 14 days of seed incubation, germination percentage improved by 9% following exposure to 5 ppm nanosized TiO2 treatment comparing to control. Similar positive effects occurred in terms of germination value and mean daily germination. Application of bulk TiO2 particles in 80 ppm concentration greatly decreased the majority of studied traits. Therefore phytotoxicity effect observed on wheatgrass seedling by application of bulk TiO2 particles in 80 ppm concentration. Exposure of wheatgrass seeds to 5 ppm nanosized TiO2 and bulk and nanosized TiO2 at 60 ppm obtained the lowest mean germination time but higher concentrations did not improve mean germination time. In general, there was a positive response by wheatgrass seed to some concentrations of nanosized TiO2. Usage of nanoparicles in order to improve germination and establishment of range plant in adverse environments similar to rangeland could be possible.

Contextualization: Lupinus bracteolaris is a heliophite species that vegetates low lands, open slopes and sandstone hills. Knowledge gap: Despite the high rusticity and potential for covering poor soils, there is still no information to assist in the analysis of its seeds. Purpose: To determine pre-germinative treatments, substrate, temperature, luminosity and water intake in the vigor of Lupinus bracteolaris seeds. Methodology: Fruits were collected from 20 matrix plants, then the seeds were processed and submitted to the following tests: pre-germinative methods (six treatments at 25°C); substrates and temperatures (sand and paper at 20, 25 and 30°C); photoblast test (white light, diffuse-green light and continuous dark under 20°C) and the imbibition curve (control and scarification between sandpaper under 20°C). Evaluations: percentage of germination and formed seedlings, germination speed index, average germination time and seedling formation, using a completely randomized design. Results and conclusions: According to the analyzed variables, the results showed superiority for the pre-germinative treatment between sandpapers for 20 seconds. In addition, the substrate paper germibox and the temperature of 20°C were the most suitable during seed germination. The seeds of Lupinus bracteolaris were classified as neutral-photoblastic and without dormancy.

Este trabalho objetivou avaliar a germinação e vigor de sementes de Plathymenia reticulata Benth. submetidas a diferentes tratamentos para superação de dormência. Os tratamentos testados foram: controle - sementes intactas (T0), escarificação mecânica com lixa d’água nº 60 (T1), desponte na região lateral do tegumento (T2), imersão em água fervente por 10 min (T3), em hipoclorito de sódio por 15 min (T4) e em solução com detergente por 15 min (T5). Foram determinados: peso de mil sementes, grau de umidade, porcentagem de germinação, índice de velocidade de germinação (IVG), tempo médio de germinação (TMG), curva de embebição, teste da condutividade elétrica e comprimento da radícula. A porcentagem de germinação das sementes submetidas ao T1 e T2 foi estatisticamente igual (respectivamente, 90 e 88%) e superiores aos outros tratamentos testados (12%). As sementes expostas aos T1 e T2 apesentaram porcentagem de absorção de água superiores e em menor tempo, assim como maiores IVG e menores TMG, comparadas aos demais tratamentos. O comprimento das radículas não variou entre os tratamentos testados. A condutividade elétrica da solução com as sementes do T0 foi superior aos outros tratamentos testados. Sementes de P. reticulata expostas ao T1 e T2 apresentam maior germinação e vigor.Palavras-chave: germinação, índice de velocidade de germinação, teste de condutividade elétrica, vigor, vinhático. DORMANCY BREAKING OF Plathymenia reticulata Benth. SEEDS ABSTRACT:This study aimed to evaluate the germination and vigor of Plathymenia reticulata Benth. seeds submitted to different dormancy breaking treatments. Testing treatments were: control - intact seeds (T0); mechanical scarification with sandpaper n. 60 (T1), coating cutting in the lateral side of tegument (T2), immersion in boiling water during 10 min (T3), in sodium hypochlorite solution during 15 min (T4), and in detergent solution during 15 min (T5). Moisture content, 1000-seed weight, germination percentage, germination speed index (GSI), mean germination time (MGT), imbibition curve, electrical conductivity test, and radicle length were determined. Germination percentage to T1 and T2 were statistically equal (90 and 88 %, respectively) and higher than others treatments tested (12%). Seeds exposed to T1 and T2 also presented higher and faster water absorption percentage, when compared to the others treatments, as well as higher GSI, and lower MGT. The rootlets length did not vary among treatments tested. Electrical conductivity of intact seeds was higher than others treatments. In this paper, we report the effects of heavy metals (HMs) (cadmium and mercury) on seed germination and seedling growth of Phragmites australis and Triarrhena sacchariflora, which are the two main typical emerging plants in Hongze Lake wetland. The results showed that there was a reduction in germination percentage, germination index and seedling length as HM concentration in the growing media increased for both treatments. The effect of HMs toxicity on seed germination and seedling growth of T. sacchariflora was more obvious than of P. australis. At the stage of seed germination, P. australis and T. sacchariflora were sensitive to Hg2 +  and Cd2 + , respectively, and Hg2 +  was more toxic than Cd2 +  at the stage of seedling growth. The effect of HMs toxicity is not invariable during plant growth. Compared to the stage of seedling growth, P. australis and T. sacchariflora are more susceptible to HMs at the stage of seed germination. In addition, we calculated the ecological thresholds of P. australis to Cd and Hg are 19.32 and 1.08 mg kg−1, and that of T. sacchariflora to Cd is 4.62 mg kg−1 based on the lab simulation. The results also indicated that the species of P. australis is more tolerant than T. sacchariflora to the HMs and is a better candidate for restoration in Hongze Lake wetland ecosystem.

Allelopathic effects of lantana (Lantana camaraL. ♯3LANCA) residues (root, shoot), foliar leachates, and the soil (where lantana was grown) on milkweedvine (Morrenia odorataLindl. ♯3MONOD) seed germination and growth over a 30-day period were examined. Foliar leachates or the soil collected from the field where lantana had been growing had no effect on the final germination percentage or the seedling growth of milkweedvine. Incorporation of dried lantana shoot or root material into soil had no effect on the final percentage germination but caused significant reductions in milkweedvine growth over a 30-day test period. Roots were more inhibitory than shoots. Fifty percent of milkweedvine seedlings died within 15 days after germination at 1% (w/w) dried lantana root incorporation into the soil, and higher concentrations increased seedling death. Lantana roots incorporated into the soil produced foliar symptoms such as wilting and desiccation, whereas lantana shoots incorporated into the soil produced yellowing of the foliage of milkweedvine. Allelopathic activity of lantana residues was still strong even after decomposition of lantana residues for 4 weeks prior to the planting of milkweedvine seeds.

This study was carried out to investigate the effects of light spectra, additional UV-A, and different growth regulators on the in vitro germination of Senecio cineraria DC. Seeds were surface-sterilized and inoculated in MS medium to evaluate the following light spectra: white, white plus UV-A, blue, green, red or darkness. The maximum germinability was obtained using MS0 medium under white light (30%) and MS + 0.3 mg L-1 GA3 in the absence of light (30.5%). S. cineraria seeds were indifferent to light. Blue and green lights inhibited germination. Different concentrations of gibberellic acid (GA3) (0.1; 0.4; 0.6; 0.8; 1.0 and 2.0 mg L-1) and indole-3-acetic acid IAA (0.1; 0.3 and 1.0 mg L-1) were evaluated under white light and darkness. No concentration of GA3 enhanced seed germination percentage under white light. However, when the seeds were maintained in darkness, GA3 improved germination responses in all tested concentrations, except at 1.0 mg L-1. Under white light, these concentrations also increased the germination time and reduced germination rate. Germination rate, under light or darkness, was lower using IAA compared with GA3.

We investigated the effects of gibberellic acid (GA3) and kinetin on seed germination of S. sesban L. and S. rostrata L. The matured seeds used for this study were obtained from International Institute for Tropical Agriculture (IITA) Ibadan, Nigeria. The seeds were harvested in 2014 and stored dry in a glass container and kept (15°C) in the refrigerator. The viability of the seeds were determined by floating the intact seeds in water and water uptake (imbibition) was carried out. Four replicates of 20 seeds per replicate were germinated and the seeds observed daily and final count was recorded after 14 days of incubation at 30°C. Intact seeds were soaked in gibberellic acid (GA3) and kinetin for 24h and germination percentage taken. The results from the water absorption demonstrated that the seeds of S. rostrata is more permeable compared to S. sesnban as indicated by higher water absorption of seeds of S. rostrata (70%) to S. sesban (25.4%) after 24hrs of incubation. The anatomy of the seed coats indicated the presence of water and gas impermeable tissues namely cuticle, macrosclereids, osteosclereids and disintegrated parenchyma layer. Generally, 0.1 mM kinetin and GA3 enhanced significant germinations compared to the control with 0% germination for the 14 days period of germination. The percentage germination of seeds of S. sesban and S. rotrata subjected to different treatments and germinated in 0.1 mM GA3 and water showed a progressive decrease in germination. From our study, 0.1 mM GA3 and Kinetin significantly enhanced seed germination of S. sesban and S. rostrata.

Abstract The greatest diversity of ferns occurs in the tropics, where approximately three in four of the species are found. In the Brazilian flora, the ferns have megadiversity, being an important plant group. The objective was to evaluate the spores germination of ferns Polypodium aureum in different types of culture medium. The sporangia were removed from fertile fronds and sterilized with sodium hypochlorite 0.57% during 30 minutes. After sterilization, the sporangia were seeded in culture media agar, MS, SH, White and B5. The experimental design was completely randomized being composed of 5 treatments (agar media, B5, SH, White and MS) and 10 replications. The ratings occurred every 3 days over a period of 45 days, counting the number of spores germinated. After this period, the germination percentage, germination time on medium, germination speed index and timing of germination were calculated. The germination percentage and the index of synchronization have not changed in function of different culture media. The medium White presented the highest average for the germination speed index whereas the agar media, MS, SH and B5 had the same germination speeds. The percentage of mortality was higher in MS medium, the White medium presented the lowest percentage of mortality, and the media composed by agar, SH and B5 showed intermediate values. The lowest average germination time occurred at the media SH and B5, since the environment composed by agar showed higher average germination time. The media MS and White showed similar results to the agar, SH and B5. Based on the obtained results, it can be concluded that the White medium is the most effective on the germination of spores of Polypodium aureum.
