Monosomic analysis of leaf peroxidase Px0 and Px9 loci in hexaploid oats

Inheritance of the peroxidase isoenzymes of the flag leaf blade was examined by isoelectrofocusing in the hexaploid oats Avena byzantina cv. Kanota, Avena fatua ssp. compacta, and Avena sativa cv. Cherokee. Two independent peroxidase loci (Px0 and Px9) were detected in the F2 from the 'Kanota' × compacta cross. The Px0a derived from compacta expressed the highest peroxidase activity and was accompanied by a post-transcriptionally modified form or mozyme. A monosomic analysis of the Px0 and Px9 loci revealed that they were located on chromosomes 18 and 6, respectively. Phenotypic expression of the peroxidases varied in each genotye at the Px0 and Px9 loci. Phenotypes of the homozygote (Px0aPx0a) and the hemizygote (Px0a—) were similar to each other. The heterozygote (Px0aPx0b) had half the enzymatic activity of the others. Px9b of compacta was functional as a suppressor but that of 'Cherokee' was nonfunctional.Key words: monosomic analysis, peroxidase loci, isoenzyme, hexaploid oats.

The response of wheat (Triticum aestivum L.) and wild oat (Avena fatua L.) to barban (4-chloro-2-butynyl-m-chlorocarbanilate) was studied as influenced by plant morphology and air temperature after application. Growth of wheat and wild oat seedlings was reduced by barban at 0.3 μg and 0.6 μg applied to the first node, respectively. Barban application to the base and midpoint of the first leaf blade required a lower dose to reduce wild oat growth than wheat growth. Increased tillering occurred from barban injury to the main culm in wheat. Wheat and wild oat susceptibility to barban increased as the post-treatment temperature decreased from 32 to 10 C. Barban selectivity for wild oats in wheat was greater at 27 and 21 C than at 16 and 10 C.

A total of 110 rice varieties of which 108 local (From Bangladesh Rice Research Institute) and two hybrid varieties (From ACI Ltd. Bangladesh) were used to identify the morphological traits during July, 2008 to June, 2009. These varieties represented four types viz. T. Aman (n=92), B. Aman (n=15), Boro (n=2) and Jhum accession (n=1) as described by Bangladesh Rice Research Institute’s literature. Though all these materials were varied ecotypically but grown in one (T. Aman) season. Irrespective of groups of all, 90 varieties were appeared distinctly morphologically from others due to seed colour; stigma colour; anthocyanin colouration of leaf sheath, nodes and lemma:palea, presence of awn and nodal root. Among 90 distinct varieties 19 were distinctly different from others by four traits, nine by three traits, 24 by two traits and 38 varieties by only single trait. The rest 20 varieties were distinguished flag leaf: attitude of the leaf blade. This was done to assess the quality traits of distinctness as major concern and was noninfluenced by the environment. Soluble peroxidase activity from pedicels of seedless table grape cv. Sultana was highly stimulated by post-bloom applications of gibberellic acid (GA3) to vines. The increase in peroxidase activity was mainly due to the induction of a basic peroxidase isoenzyme (pI > 9; BPrx-HpI). The activity of two other peroxidase isoenzymes of pI 6.5 and 3.2 was not altered by the hormone treatment. BPrx-HpI was induced by GA3 in pedicels and rachis but not in berries, although in berries peroxidase activity was also stimulated by post-bloom GA3 applications. BPrx-HpI oxidised guaiacol and ortho-phenylenediamine (o-PDA), while the others peroxidases found in the pedicel and in the berry oxidised only o-PDA. Hence, BPrx-HpI was characterised as a guaiacol-peroxidase showing no activity towards ascorbic acid (ASC). The possible role of BPrx-HpI in pedicel lignification and berry-drop caused by GA3 applications to cv. Sultana vines is discussed.

The objective of this study was to investigate relationships between leaf area index, leaf chlorophyll concentration, yield components and grain yield in oat (Avena sativa L.). Ten oat varieties were analyzed in field conditions regarding those traits. Flag leaf chlorophyll concentration range between 451.51 and 747.79 units of μmol of chlorophyll per m2. Also, leaf area index range between 13.68 to 32.84 cm2. Significant correlation indices were highlighted between yield components and leaf area index, yield/yield components and chlorophyll concentration of flag leaf.

Experiments were conducted in controlled environmental chambers to determine the influence of temperature on the phytotoxicity of seven soil-applied herbicides. Diclofop {2-[4-(2,4-dichlorophenoxy)phenoxy] propanoic acid} soil incorporated or surface applied, was more toxic to wild oat(Avena fatuaL.) shoots at 10 and 17 C than at 24 C. Efficacy of diclofop was enhanced with soil incorporation. Diclofop toxicity to wild oat roots was not influenced by a change in temperature. EPTC(S-ethyl dipropylthiocarbamate) stimulated sugarbeet(Beta vulgarisL. ‘American Crystal Hybrid B’) shoot dry weight production at 10 C and caused dry weight reduction at 24 C. Atrazine [2-chloro-4-(ethylamino)-6-(isopropylamino)-s-triazine] toxicity to barley(Hordeum vulgareL. ‘Larker’) and alachlor [2-chloro-2′,6′-diethyl-N-(methoxymethyl)acetanilide] toxicity to oats(Avena sativaL. ‘Chief’) increased with increased temperature from 10 to 17 C. Temperatures within the range of 10 to 24 C did not affect trifluralin (α,α,α-trifluoro-2,6-dinitro-N,N-dipropyl-p-toluidine) toxicity to barley or BAY-5653 [N-(2-benzothiazolyl)-N-methylurea] or chloamben (3-amino-2,5-dichlorobenzoic acid) toxicity to oats.

ABSTRACT Eighteen nodules from thirteen patients with non-toxic nodular goitre were studied. The nodules were classified, according to the 131I scintiscanner, into »cold« and »warm« types. The histological diagnosis of all the glands was multinodular colloid goitre. An enzymatic system with iodide-peroxidase activity was prepared from nodular tissue obtained by surgical thyroidectomy. The enzymatic activity was determined by spectrophotometry at 287.5 nm by measuring the formation of triiodide ion. The group of »cold« nodules showed an average of 333 units of enzymatic activity and the »warm« nodules 940 units. The significance of the correlation between the capacity for iodide-uptake, the iodide-peroxidase activity and the involution of the metabolic steps in the nodular goitre is discussed.

An experiment was conducted to study the heterosis of 27 F1 hybrids produced from nine CMS lines and three restorer lines of rice with their parental lines and BRRI Dhan29 as standard check for 17 yield and its contributing traits. Mid parent, better parent and standard heterosis of most of the crosses were significant for most of the characters studied either in positive or in negative direction. Desirable and significant mid parent and better parent heterosis was observed in 13 and seven cross combinations, respectively for grain yield and most of its related traits. Considering more than 20% mid parent and better parent heterosis for grain yield along with most of its related traits, nine and five cross combinations, respectively were identified as good heterotic combinations over mid parental and better parental value. On the basis of individual trait significant and desirable mid parent heterosis was observed in eight cross combinations for plant height, 12 for leaf blade length, nine for flag leaf blade length, 13 for flag leaf sheath length, eight for tillers per hill, eight for panicles per hill, 14 for days to 50% flowering, four for days to maturity, four for panicle length, 14 for panicle weight, nine for primary branches per panicle, 16 for secondary branches per panicle, six for filled grains per panicle, 15 for 1000 seed weight, 13 for grain yield per hill and 15 for harvest index. Significant and desirable better parent heterosis was observed in 12 cross combinations for plant height, 11 for leaf blade length, 5 for flag leaf blade length, nine for flag leaf sheath length, seven for tillers per hill, seven for panicles per hill, 25 for days to 50% flowering, one for days to 100% flowering, seven for days to maturity, three for panicle length, nine for panicle weight, five for primary branches per panicle, 12 for secondary branches per panicle, four for filled grains per panicle, nine for 1000 seed weight, seven for grain yield per hill and nine for harvest index. None of the crosses were identified as good heterotic over standard check for grain yield per hill but many good and desirable heterotic crosses were identified over standard check for most of the yield related characters. This research was carried out in experimental field of Field Crops Department of Agricultural Faculty of Namik Kemal University in randomized split block design with three replications per treatment during 2004-2005 and 2005-2006. The objective of this study was to find out the contribution rates of awn, flag leaf, 1st upper leaf blade, 2nd upper leaf blade and other leaf blades to main yield components in three durum wheat cultivars (cv. Kiziltan-91, Kunduru-1149, and Yelken-2000). The results of this experiment showed that removal of awn, flag leaf, 1st upper leaf blade, 2nd upper leaf blade, and other leaf blades reduced significantly spike weight, number of grains per spike, grain weight per spike, and 1000-grain weight except the number of spikelets per spike. The translocation of labelled assimilates from the flag leaf blade to the ear and other parts of the plant was followed in wheat, 15-20 days after anthesis. The velocity of assimilates moving through the top internode varied from 80 to 100 cm/hr and the velocity of movement down the leaf sheath was about half this value. Removing grains from the head reduced the velocity of the assimilates moving up the stem from the node and increased their downward velocity in the stem. How-ever, the rate of movement out of the leaf was unaffected by the removal of grains. With full grain development assimilates from the flag leaf moved directly from the node of insertion of the leaf up the stem to the ear. The results are discussed in relation to the vascular anatomy of wheat and the mechanism and factors controlling translocation.
