A novel phylogenetic group withinThozetella(Chaetosphaeriaceae): a new taxon based on morphology and DNA sequence analyses

A new species, Thozetella pinicola , was isolated from leaf litter of Pinus elliottii Engelm. in Hong Kong. This taxon is described and compared with existing species in the genus. It occurs on the substrate as creamy white sporodochia and has short black conidiophores. Morphological characters are typical of Thozetella and it most closely resembles Thozetella falcata , Thozetella gigantea and Thozetella nivea , but may be distinguished by its distinct microawns and different conidial size. To gain further taxonomic insight into the phylogenetic relationships of our new taxon and its allies, we sequenced and analysed 6 different regions of 3 genes (ribosomal DNA and protein coding genes: RNA polymerase II largest subunit (RBP2) and β-tubulin). Resulting phylogenies are compared with existing morphological information. Molecular data support the relationship between Thozetella species and the Chaetosphaeriaceae (Chaetosphaeriales, Sordariomycetes). In addition, we recovered a new phylogenetic lineage (or group) within the existing phylogenetic framework of Thozetella as previously proposed. In particular, there is a close association between T. pinicola and T. nivea, which is strongly supported. The affinities of these 2 newly sequenced taxa are discussed in light of morphological and molecular characters.

Molecular data from the 28S D2 ribosomal nuclear gene fragment were utilised to construct a phylogeny for the Australian Ennominae. Sequences were obtained from 68 geometrid and 5 outgroup species. Sequences from a smaller subset of 17 species also were analysed using the nuclear protein-coding gene EF-1a. Species were sampled from all major subfamilies of the Geometridae as no a priori assumptions could be made confidently about possible sister groups to the Australian Ennominae.The major findings from these analyses were as follows:(a) Drepanidae are a sister group to Geometridae;(b) Larentiinae are derived basally within the Geometridae; the Sterrhinae are the penultimate basally derived group;(c) Oenochrominae s. str. are closely related to the Geometrinae.(d) Ennominae are not monophyletic;(e) Tasmanian Archiearinae are misplaced in the Archiearinae and have close affinities to Australian Nacophorini (Ennominae);(f) Australian Nacophorini are not monophyletic.These results are at odds with traditionally held beliefs on the origins of Geometridae but are in broad agreement with and elaborate on the findings of Abraham et al. (2001). The implications of these findings in relation to key morphological characters are discussed using the proposed phylogenetic framework.

Several soil samples from different habitats in Badakhshan province of Afghanistan were collected to isolate and characterize bacteria feeding nematodes. The Galleria mellonella-baiting method was used for the isolation of the Afghan insect-associated nematodes. The nematodes were studied using morphological and morphometric data. The Oscheius specimen was characterized by a longer body (630–820 µm) and shorter pharynx (125–145 µm), whereas other morphological characters were not unusual. The Diploscapter specimen had an annulated cuticle, with lip region width 1.5 times shorter than the stoma, and had separated pharyngeal corpus from the isthmus and vulva located in the middle of the body. The molecular data were derived using three loci; 18S, 28S (D2/D3 segment), and ITS rRNA region, which were utilized to measure the genetic distance. The phylogenetic analysis was conducted to reconstruct the relationship tree. Both morphological and molecular approaches confirmed the identity of nematode isolates as Oscheius tipulae and Diploscapter coronatus. This is the first report of insect-associated nematodes from the soil of Afghanistan. Both species were capable of infecting and killing G. mellonella larvae in less than 96 h.

In this article, we present the nearly complete mitochondrial genome of the Subalpine Kashmir vole Hyperacrius fertilis (Arvicolinae, Cricetidae, Rodentia), assembled using data from Illumina next-generation sequencing (NGS) of the DNA from a century-old museum specimen. De novo assembly consisted of 16,341 bp and included all mitogenome protein-coding genes as well as 12S and 16S RNAs, tRNAs and D-loop. Using the alignment of protein-coding genes of 14 previously published Arvicolini tribe mitogenomes, seven Clethrionomyini mitogenomes, and also Ondatra and Dicrostonyx outgroups, we conducted phylogenetic reconstructions based on a dataset of 13 protein-coding genes (PCGs) under maximum likelihood and Bayesian inference. Phylogenetic analyses robustly supported the phylogenetic position of this species within the tribe Arvicolini. Among the Arvicolini, Hyperacrius represents one of the early-diverged lineages. This result of phylogenetic analysis altered the conventional view on phylogenetic relatedness between Hyperacrius and Alticola and prompted the revision of morphological characters underlying the former assumption. Morphological analysis performed here confirmed molecular data and provided additional evidence for taxonomic replacement of the genus Hyperacrius from the tribe Clethrionomyini to the tribe Arvicolini.

Rotylenchus is a widely-distributed economically important plant-parasitic nematode group whose species-level identification relies largely on limited morphological characters including character-based tabular keys and molecular data of ribosomal and mitochondrial genes. In this study, a combined morphological and molecular analysis of three populations of R. goodeyi from Belgium, Poland and the Netherlands revealed important character variations of this species leading to synonymisation of R. rhomboides with R. goodeyi, and a high nucleotide variation within cox1 gene sequences in these populations. Additional Illumina sequencing of DNA from individuals of the Dutch population revealed two variants of mitogenomes each of approximately 23 Kb in size, differing by about 9% and containing eleven protein coding genes, two ribosomal RNA genes and up to 29 transfer RNA genes. In addition to the first representative whole genome shotgun sequence datasets of the genus Rotylenchus, this study also provides the full length mitogenome and the ribosomal DNA sequences of R. goodeyi.

Crocus musagecitii is described as a new species. Diagnostic morphological characters, a full description and detailed illustrations are provided on the basis of the type specimen and wild specimens. Morphologically, C. musagecitii is close to Crocus biflorus subsp. pseudonubigena. Crocus musagecitii differs from C. biflorus subsp. pseudonubigena by the lack of stripes or narrow purplish tongue on outside of outer tepals, wider tepals, and homogenously yellow anthers. In order to clarify the phylogenetic position of this species within the Crocus adamii species complex, we sequenced the internal transcribed spacer region (ITS: ITS1 + 5.8SrDNA + ITS2) and 5’ external transcribed spacer (ETS) of the nuclear ribosomal DNA (rDNA). A phylogenetic tree obtained by Bayesian phylogenetic inference is given. Phylogenetic analyses revealed that the new taxon is close to C. munzurensis. Crocus musagecitii differs from its phylogenetically closest relative C. munzurensis by the corm tunics (C. musagecitii: coriaceus; C. munzurensis: membranous), the number of leaves (C. musagecitii: up to 8; C. munzurensis: up to 4) and non-hairy leaf margins.

Phellinus vietnamensis sp. nov. is described from Vietnam based on morphological characters and molecular data. Morphologically, it is characterized by perennial, pileate basidiomata, a dimitic hyphal system, hooked hymenial setae, and colorless, broadly subglobose to ovoid, thick-walled basidiospores 5.5–6 × 4.8–5.2 μm. Phylogenetically, the status of Phellinus vietnamensis is strongly supported based on sequences of the nuclear internal transcribed spacer (ITS) regions, the translation elongation factor 1-α gene (EF1-α) nuclear large subunit rDNA (nrLSU) and the second largest subunits of RNA polymerase II (RPB2).

Dasyphyllum Kunth is the most diverse genus of the South American subfamily Barnadesioideae (Asteraceae), comprising 33 species that occur in tropical Andes, Atlantic Forest, Caatinga, Cerrado, and Chaco. Based on distribution, variation in anther apical appendages, and leaf venation pattern, it has traditionally been divided into two subgenera, namely, Archidasyphyllum and Dasyphyllum. Further, based on involucre size and capitula arrangement, two sections have been recognized within subgenus Dasyphyllum: Macrocephala and Microcephala (=Dasyphyllum). Here, we report a phylogenetic analysis performed to test the monophyly of Dasyphyllum and its infrageneric classification based on molecular data from three non-coding regions (trnL-trnF, psbA-trnH, and ITS), using a broad taxonomic sampling of Dasyphyllum and representatives of all nine genera of Barnadesioideae. Moreover, we used a phylogenetic framework to investigate the evolution of the morphological characters traditionally used to recognize its infrageneric groups. Our results show that neither Dasyphyllum nor its infrageneric classification are currently monophyletic. Based on phylogenetic, morphological, and biogeographical evidence, we propose a new circumscription for Dasyphyllum, elevating subgenus Archidasyphyllum to generic rank and doing away with the infrageneric classification. Ancestral states reconstruction shows that the ancestor of Dasyphyllum probably had acrodromous leaf venation, bifid anther apical appendages, involucres up to 18 mm in length, and capitula arranged in synflorescence.

A new polypore, Rhodonia tianshanensis, collected from West Tianshan Nature Reserve in Xinjiang Autonomous Region (northwest China), is described and illustrated based on morphological characteristics and molecular evidence. It is characterized by having resupinate basidiomata with an oblique tube layer, fusoid cystidioles in the hymenium, and cylindrical basidiospores. Based on multiple loci DNA sequences including the internal transcribed spacer (ITS), the large subunit (nLSU), and the second subunit of RNA polymerase II (RPB2) regions, our phylogeny strongly supported R. tianshanensis as a new species belonging to the genus Rhodonia.

Microweiseinae is a quite recently established subfamily within ladybird beetles (Coccinellidae). According to recent analyses of morphological and molecular data, it has been divided into three tribes. Members of the subfamily are distributed mostly in tropical and subtropical regions of the world. Despite several recent taxonomical studies of this group, its diversity and distribution is still not fully understood. Recent field collecting on Madeira Island resulted in the discovery of interesting specimens belonging to a yet unknown taxon, described here as Madeirodula atlantica gen. et sp. nov. Phylogenetic analysis of morphological characters indicate that the new taxon form a distinct branch within the subfamily Microweiseinae, for which we propose a new tribe Madeirodulini trib. nov. Evolutionary trends within the subfamily are discussed, and an updated key to the tribes of Microweiseinae is provided.

The Balkan Peninsula and the amphi-Adriatic region are biodiversity centres with a high number of Campanula species, some of them with restricted and isolated distributions. Among those is the morphologically, karyologically and genetically well-defined linage of isophyllous Campanula species of the garganica clade, taxonomically treated as Campanula ser. Garganicae. In the central Adriatic, on the Island of Vis (Croatia), an isophyllous Campanula was found growing in rocky crevices of calcareous sea cliffs among chasmophytic vegetation with extremely challenging ecological conditions. Based on morphological characters (monopodial growth form, cordate to ovate basal leaves, campanulate corolla, presence of obtuse hairs at the base of filaments, capsule opening by basal pores and brown shiny seeds) the unidentified plant from the Island of Vis falls into ser. Garganicae. To establish the phylogenetic position and relationships of the unknown Campanula among other species of the garganica clade, nuclear ITS and chloroplast trnL-F sequences as well as comparative morphology, karyology and scanning electron microscopy of seeds were used. Due to its campanulate corolla the plants from Vis are morphologically close to C. portenschlagiana from the eastern Adriatic, from which they differ in several quantitative floral characters. The karyological analysis revealed a diploid chromosome number (2n = 34), while SEM analysis showed that seed coat of new taxon has weakly striate testa, thin wavy striped radial walls and slightly marked lumen. Additionally, molecular data show that the Vis taxon is clearly separated as a distinct species from other species of ser. Garganicae. Therefore, it is described and illustrated as a new species, Campanula teutana Bogdanović & Brullo.  Because of the small number of individuals within the single population, C. teutana should be included on the Red List of the vascular flora of Croatia as an endangered species (EN).
