The control of adventitious root production in tissue-cultured Douglas-fir

Rooting percentage and root number in tissue-cultured Douglas-fir (Pseudotsugamenziesii (Mirb.) Franco) were influenced by the rooting substrate, the concentration of sucrose and boron in the rooting medium, shoot height, and shoot generation. Peat–perlite was a better substrate than agar, producing 70% compared with 0% rooted shoots, respectively. On peat–perlite, cell divisions were organized and were associated with tracheid nests, whereas on agar proliferation was unorganized and not restricted to the nests. A sucrose concentration of 4% was optimal for the production of nodular or rooted shoots. At 4% sucrose and 3 mg •L−1 boric acid, 100% of the shoots rooted, and the mean root number was 11. Rooting percentage and root number were significantly higher with shoots that were 3 cm tall than 2 or 1 cm tall shoots. Finally, it was found that shoot responses were more rapid in third and fourth generation shoots than in second generation shoots. At least 80% were rooted or nodular after 4 weeks, compared with only 36% from the second generation.

Anaplastic lymphoma kinase-rearranged non-small-cell lung cancer (ALK+ NSCLC) is a model disease for the use of targeted pharmaceuticals in thoracic oncology. Due to higher systemic and intracranial efficacy, the second-generation ALK tyrosine kinase inhibitors (TKI) alectinib and brigatinib have irrevocably displaced crizotinib as standard first-line treatment, based on the results of the ALEX and ALTA-1L trials. Besides, lorlatinib and brigatinib are the preferred second-line therapies for progression under second-generation TKI and crizotinib, respectively, based on the results of several phase II studies. Tissue or liquid rebiopsies at the time of disease progression, even though not mandated by the approval status of any ALK inhibitor, are gaining importance for individualization and optimization of patient management. Of particular interest are cases with off-target resistance, for example MET, HER2 or KRAS alterations, which require special therapeutic maneuvers, e.g., inclusion in early clinical trials or off-label administration of respectively targeted drugs. On the other hand, up to approximately half of the patients failing TKI, develop anatomically restricted progression, which can be initially tackled with local ablative measures without switch of systemic therapy. Among the overall biologically favorable ALK+ tumors, with a mean tumor mutational burden uniquely below 3 mutations per Mb and the longest survival among NSCLC currently, presence of the EML4-ALK fusion variant 3 and/or TP53 mutations identify high-risk cases with earlier treatment failure and a need for more aggressive surveillance and treatment strategies. The potential clinical utility of longitudinal ctDNA assays for earlier detection of disease progression and improved guidance of therapy in these patients is a currently a matter of intense investigation. Major pharmaceutical challenges for the field are the development of more potent, fourth-generation TKI and effective immuno-oncological interventions, especially ALK-directed cell therapies, which will be essential for further improving survival and achieving cure of ALK+ tumors.

Responses to changes in intravascular pressure of isolated rat mesenteric arterioles were investigated under no-flow conditions. First-, second-, third-, and fourth-generation arterioles were isolated and cannulated. Vascular diameters were measured with an image-shearing device and recorded. The arterioles (except for the first-generation vessels) developed spontaneous tone, corresponding to the step increases in intravascular pressure (from 20 to 160 mmHg, by 20-mmHg steps). For example, at 80 mmHg pressure the mean diameters of first-, second-, third-, and fourth-generation vessels were 286.9 +/- 5.0, 203.4 +/- 8.2, 92.5 +/- 4.6, and 35.6 +/- 4.8 microns, respectively; by use of a Ca(2+)-free solution containing ethylene glycol-bis(beta-aminoethyl ether)-N,N,N',N'-tetraacetic acid (1 mM) and sodium nitroprusside (SNP; 10(-4) M) the passive diameters of these vessels were 295.6 +/- 6.3, 238.4 +/- 11.7, 120.3 +/- 3.7, and 59.4 +/- 3.1 microns, respectively, demonstrating that the degree of pressure-induced constriction increased with the increasing order of generations (3, 14, 24, and 43%, respectively). The vasoactive function of endothelium and vascular smooth muscle was assessed by the responses of arterioles to acetylcholine (ACh; 10(-6) M) and SNP (10(-7) M) before and after removal of the endothelium with air. After removal of the endothelium, dilation to ACh was abolished while dilation to SNP was retained. Removal of the endothelium did not significantly alter the changes in the diameter of arterioles in response to step increases in intravascular pressure.(ABSTRACT TRUNCATED AT 250 WORDS)

: The study evaluates production parameters (height, diameter at breast height, volume) of Douglas fir (<I>Pseudotsuga menziesii</I> [Mirb.] Franco) at mesotrophic sites of the Křtiny Training Forest Enterprise in mature stands. In total, 29 mixed stands were assessed with the registered proportion of Douglas fir at an age of 85 to 136 years. Comparing the 10 largest Douglas firs with the 10 largest spruces or larches higher, and as a rule markedly higher, production potential of introduced Douglas fir was found in all assessed stands. There were also groups of trees where the volume of Douglas fir was twice to 3 times higher than the volume of spruce or larch (see Tabs. 5 to 10). For example, in stand 177B11, the mean volume of 9.12 m<sup>3</sup> was recorded in the 10 largest Douglas fir trees but the volume of spruce reached only 3.17 m<sup>3</sup> and the volume of larch was 3.70 m<sup>3</sup>. Differences in mensurational parameters of Douglas fir found on the one hand and of Norway spruce (<I>Picea abies</I> [L.] Karst.) or European larch (<I>Larix decidua</I> Mill.) on the other hand compared by ANOVA tests were statistically highly significant. Annual ring analyses have shown that at present the volume increment of particular Douglas fir trees ranges from 0.12 to 0.16 m<sup>3</sup> per year in mature stands (i.e. about 1.5 m<sup>3</sup> every 10 years).

Abstract Background Sepsis frequently leads to acute kidney injury. In severe cases, patients may require continuous renal replacement therapy (CRRT) which involves placement of a dialysis catheter and an extracorporeal blood filtration circuit. CRRT is commonly considered to “mask” fever, though this phenomenon has not been investigated. Methods We queried an institutional database of all patients on CRRT from 2007 to 2015 for inpatient temperature data and antibiotic administration records. Receipts of piperacillin–tazobactam, a carbapenem, or a third or fourth-generation cephalosporin, indicating a serious infection, were considered intervention arm. We analyzed temperatures recorded in the intensive care unit before, during, and after CRRT. Patients were divided into groups that did not receive antibiotics as well as those who did. Temperature data were Winsorized to correct for outliers. We also performed descriptive statistics for each group. Results There were 237,988 temperature readings for 1,568 ICU patients on CRRT. 1,153 patients received broad-spectrum antibiotics in ICU. In patients who received antibiotics in ICU and were presumed to have an infection, the mean temperature was 37.2°C prior to initiation of CRRT, 36.8°C while on CRRT, and 37.2°C following discontinuation of CRRT. In the 415 patients who did not receive IV antibiotics, the mean temperature was 36.9°C prior to initiation of CRRT, 36.6°C while on CRRT, and 37.0°C following discontinuation of CRRT. During each of the periods before, during, and after CRRT, patients who received antibiotics had significantly higher temperatures than those who did not (P < 0.001). Patients receiving antibiotics were generally younger (mean 60 years vs. 64 years, P < 0.001), had longer ICU stays (mean 29 days vs. 12 days, P < 0.001) and spent more time being ventilated (mean 23 days vs. 7 days, P < 0.001). The mean SOFA score on day one was similar (mean 11.1 in the antibiotic group and 10.5 in the other group). Conclusion This investigation suggests that patients have slightly lower temperatures while on CRRT, by on average less than half a degree. A similar effect is seen in both patients with infections as well as those without. Further work will be needed to determine what constitutes a true febrile response in this population. Abstract Background: A new commercially available (so-called second-generation) IRMA for parathyroid hormone (PTH) separately detects intact PTH and its N-truncated fragments; however, no studies have compared the first- and second-generation IRMAs for PTH in patients with primary hyperparathyroidism (PHPT) to assess their respective diagnostic accuracies. Methods: We concomitantly investigated 39 postmenopausal patients with PHPT and a control group of 70 healthy postmenopausal women matched for age, renal function, and vitamin D status. In all individuals, PTH was measured with a classic IRMA (PTH-S; DiaSorin Inc.), which uses antibodies directed against epitopes 1–34 and 39–84, and a new method (Scantibodies Laboratory. Inc.), which uses antibodies against epitopes 1–4 and 39–84 (PTH-W) and epitopes 7–34 and 39–84 (PTH-T). We also assayed serum PTH in 10 PHPT patients every 24 h for 5 days after successful surgery. Results: The different assays gave serum PTH values that were &gt;2 SD higher than values for the control population in 59% (PTH-S), 77% (PTH-W), and 82% (PTH-T) of patients with PHPT. However, ROC curve analysis showed no significant differences among the three PTH assays, demonstrating overlapping diagnostic sensitivities. In PHPT patients, the correlation among the assays was highly significant (r = 0.91–0.92; P &lt;0.001). The ratio PTH-W:PTH-T × 100 showed a gaussian distribution in both PHPT patients and controls, whose mean (SD) values [63.4 (13.3)% vs 64.5 (9.5)%, respectively] did not differ significantly. After parathyroidectomy, the mean percentages of variation in PTH detected with all of the assays were quite similar. Conclusions: The distribution of the PTH-W:PTH-T ratio in patients and controls suggests that PHPT does not markedly influence the rate at which biologically inactive fragments are generated by central or peripheral cleavage of PTH. The similar postoperative curves seem to contradict the hypothesized effect of acute hypocalcemia in modulating the central secretion of hormonal fragments. Our results indicate that the three investigated assays have similar diagnostic sensitivities in PHPT.

Logs of Pseudotsugamenziesii (Mirb.) Franco that had been on the ground for up to 313 years were grouped into five decay classes that ranged from 1, essentially undecayed, to 5, soft and incorporated into the forest floor but still identifiable. The mean residence times on the forest floor were 7, 17, 33, 82, and 219 years for decay classes 1 through 5, respectively. The single-exponential model of litter decomposition was fitted to the density of these logs. The summation-exponential model was constructed by summing single-exponential models fitted to lignin, cellulose, and the acid detergent soluble fraction. Both models gave virtually identical, statistically significant fits to the data. Wood density of these Douglas-fir logs decreased more slowly than that of most species other researchers have studied. The single-exponential model gave mineralization rates (k) of 0.0063 and 0.0070 year−1 when residence time and decay class age (mean residence time of the decay class), respectively, were used as the independent variable. Lignin decayed more slowly than cellulose or the fraction soluble in hot acid detergent, both of which decayed at rates that were not significantly different; thus, the summation-exponential model is recommended when these constituents are of interest.

ABSTRACTMean annual quantities of solar radiation absorbed during various stages of regeneration of a tropical rain forest in the upper Rio Negro valley of Colombia and Venezuela were estimated for the consecutive intervals between clear-cut and 1,3, 10, 20, 35, 60, 80 and 200 years of growth. Forest phytomass and litter fall data from each of these stages were used to calculate the mean annual net dry matter production per unit of absorbed photosynthelically active radiation (PAR), the PAR conversion efficiency. The quantities of PAR absorbed by the forest stands were calculated from the leaf area index values with an extinction coefficient for PAR of 0.74, a PAR albedo of 0.04, and an annual mean incoming PAR of 2.86GJ m-2y-1. Efficiency decreased with increase in successional stage. During the first 10 years of regrowth, the efficiency of conversion of PAR into above-ground phylomass averaged 0.23 g MJ-1, decreasing to 0.07 g MJ-1 over the following 50 years. Inclusion of annual root production in the calculations resulted in a small increase in PAR conversion efficiency; however, efficiency was more than doubled for some periods when the annual leaf and twig lillerfall were included. Efficiency values for above-ground production were much lower than PAR conversion efficiency values estimated for above-ground production of temperate forests.

Saudi women have recently started using oral contraceptives (OCs), which has led to risk of venous thromboembolism (VTE). The risk varies with the type of OC generations used, and with OC use the risk for VTE increases by 2- to 6-fold. This study evaluated the effect of OC types in relation to ABO blood group on the risk of VTE among Saudi women. Thrombin generation (TG) was measured in the plasma of the women in the presence and absence of platelet rich plasma, platelet poor plasma and thrombomodulin or activated protein C. OC usage increased TG parameters ETP and Peak height by 9.81% and 16.04%, respectively. An increased risk of VTE was seen among women on third generation OCs as compared to those on second generation products. Within OC generations, we found that for women using fourth generation OCs, their ETP increased by 36.18% as compared to those using second generation and by 6.07% in those using third generation compared to those using second generation. There was significant difference with respect to ABO blood groups and OC generation types, but larger sample size is required. Women who are 40 years and older and using third generation OC had a higher risk of having thrombosis (11.84%), as compared to those using second generation OC (8.79%) and to those using fourth generation OC (5.03%). An association between different OC groups and non-O blood group in thrombosis generation was noted. TG parameters were significantly increased in relation to BMI when comparing to OC users versus non-users. In addition, inhibition of TG parameters in the presence of recombinant human thrombomodulin (TM) and activated protein C (APC) were significantly increased.

The mean number of pollen grains produced per pollen cone (P) ranged from 37 310 to 62 960 ([Formula: see text], SE = 1584) among eight seed-orchard clones of Douglas-fir (Pseudotsugamenziesii (Mirb.) Franco) and correlated significantly (r = 0.87, p < 0.05) with the mean number of microsporophylls per cone but not significantly with pollen cone length. The number of pollen grains per milligram (G) also differed among clones (range 2323–3112; [Formula: see text], SE = 32) and correlated significantly and negatively (r = −0.86) with grain diameter. Although this study provides evidence for genetic variation in both P and G, the differences were relatively small. Adjusting pollen-cone counts for P would add little, if any, precision to estimates of pollen production in seed-orchard clones. If balanced clonal representation in pollen mixes is desired, equal weights of pollen would probably suffice for most breeding applications.

Background. Aim of this prospective study was to evaluate safety and efficacy of second generation Wallis system for degenerative lumbar disc disease. Material and methods. 25 patients underwent discectomy and insertion of Wallis system. Outcome assessment was done using Modified Oswestry disability index (ODI) and visual analogue scale (VAS) for backache and leg pain. Results. The mean follow-up was 20.5 months. The mean VAS for backache and leg pain showed significant improvement from 7.2 to 3.0 (p<0.001) and mean modified Oswestry disability index showed significant improvement from 59.1 to 24.7 (p<0.001). Only one patient (4%) underwent revision surgery for persistent pain due to granulation tissue deep to the implant. There were no other complications. Conclusion. The Wallis spine system is a safe and effective implant for treating degenerative lumbar disc disease.
